1.Simvastatin inhibits expression of pluripotent markers Oct3/4, Nanog, and Sox-2 in human breast cancer MCF-7 Cells

Objective:To investigate the effect of simvastatin on expression of pluripotent markers Oct3/4, Nanog, and Sox-2 in human breast cancer MCF-7 cells. Methods:Quantitative reverse transcription-polymerase chain reaction (qRT-PCR), immunofluo-rescent staining, flow cytometry, and Western blot were used to detect the expression of pluripotency markers Oct3/4, Nanog, and Sox-2 in human breast cancer MCF-7 cells treated with different doses of simvastatin. Results:qRT-PCR revealed the more signifi-cant inhibition of gene expressions of Oct3/4, Nanog, and Sox-2 in human breast cancer MCF-7 cells when subjected to high doses of simvastatin (10, 50, and 100 μmol/L) compared with the control group (P<0.05). By contrast, no significant difference was observed be-tween the expressions of low-dose simvastatin treatment (1 μmol/L) and control (P>0.05). The inhibitory effect of simvastatin on the gene expressions of Oct3/4 and Nanog was more significantly apparent at 50 and 100 μmol/L dosages than at 10 μmol/L (P<0.05). By contrast, no significant difference in the inhibitory expression of Sox-2 was observed among the three high-dose treatments (P>0.05). Between the two higher-dose treatments (50 and 100 μmol/L), no significant difference in the inhibitory expressions of Oct3/4, Nanog, and Sox-2 in MCF-7 cells was found. Meanwhile, in the 10 μmol/L simvastatin treatment, immunoflurescent staining showed a marked reduction in the protein expression of all three pluripotent markers in MCF-7 cells, and flow cytometry demonstrated a decrease of Oct3/4-, Nanog-, and Sox-2-positive cells (P<0.05). Western blot further revealed that the protein expression of Oct3/4, Nanog, and Sox-2 in MCF-7 cells was significantly declined by the same simvastatin dose (P<0.05). Conclusion: Simvastatin can inhibit the expression of pluripotent markers Oct3/4, Nanog, and Sox-2 in human breast cancer MCF-7 cells, proving the anti-cancer properties of simvastatin.
2.Supercapsular percutaneously-assisted total hip approach for the elderly with femoral neck fractures:study protocol for a prospective, open-label, randomized, controlled clinical trial

BACKGROUND:Artificial hip replacement by the posterolateral approach is generally introduced as a treatment for femoral neck fracture in the elderly, but it is limited by the need for a large incision, cutting off the extortor, and postoperative hip dislocation. The supercapsular percutaneously-assisted total hip (SuperPATH) approach was developed based on the original lateral approach. The SuperPATH approach for hip replacement is characterized by the reduced chance of damaging blood vessels and nerves, reducing intraoperative blood loss, retaining all the joint capsules and supinator, and significantly reducing the incidence of postoperative hip dislocation. OBJECTIVE:We hypothesized that the SuperPATH approach for hip replacement can achieve better efficacy in the elderly with femoral neck fractures and promote the recovery of postoperative hip function compared with the posterolateral approach. METHODS/DESIGN:This is a prospective, single-center, open-label, randomized control ed clinical trial that will be completed at the Pudong Hospital of Fudan University, Shanghai, China. Forty patients with femoral neck fractures were randomly divided into two groups to undergo femoral head replacement by the SuperPATH approach in the experimental group, and by the conventional posterior approach in the control group. The follow-up period is 1 week and 6 months. The main outcome measures are Harris hip scores at baseline, and at 1 week and 6 months postoperatively to assess functional recovery of the hip. Secondary outcome measures to evaluate the advantages of the SuperPATH approach include the length of surgical incision, intraoperative blood loss, operative time, 24-hour postoperative drainage, and hospital stay. Other outcome measures include the incidence of adverse reactions at 1 week and 6 months after surgery. The study protocol was approved by the Ethics Committee of the Pudong Hospital of Fudan University, China, and performed in accordance with the guidelines of the Declaration of Helsinki, formulated by the World Medical Association. Written informed consent was obtained from all participants. DISCUSSION:The aim of this trial is to test our hypothesis that the SuperPATH approach is safer and more reliable for hip functional recovery compared with the posterolateral approach for the artificial hip replacement in femoral neck fractures of the elderly.
3.Dose selection and effect of Oolong tea on the expression of β3-adrenergic receptor gene for weight reducing

BACKGROUND:Obesity is an important problem concerned domestically and internationally. How to control the body mass and to reduce the weight without any effect on normal food intake is the focus for study.OBJECTIVE:To probe into the weight-reducing effect of Oolong tea and its effect on the mRNA level of β3-adrenergic receptor(β3-AR).DESIGN: Completely randomized grouping, controlled trial SETTING:Department of Nutrition and Food Hygiene, College of Public Health, Nanjing Medical University; Institute of Pediatric Medicine, Nanjing Medical University.MATERIALS: This experiment was conducted in the laboratory of Nanjing Medical University from September 2003 to February 2004. The obese rat models were made with the diet of high energy and high fat in male rats weighing about 80 g. Thirty-two male obese rats were selected.And Oolong tea extract was prepared, whose concentration was equivalent to 0.24 g of tea.INTERVENTIONS:Thirty-two male obese rats were divided randomly in-to 4 groups: obese control group, low, middle and high dose of oolong tea groups. There were 8 rats in each group. The rats in the control group were fed by gavage with distilled water every day. The other rats in low, middle or high dose of Oolong tea groups were fed by gavage with 0.4 g/kg, 1.2 g/kg, and 2.4 g/kg of Oolong tea respectively. They were all fed with diet of high energy and high fat. Each rat was raised in separate cage. The room temperature for the rats remained about 22 ℃ with the humidity of 55%. The rats were free access to water, but the diet was fed twice a day at a fixed amount. If it was finished, no more diet would be added. Thirty days later, body mass, maximal diameter of adipocytes were measured, and β3-ARmRNA levels in adipose tissues were measured.MAIN OUTCOME MEASURES: Body mass, increased body mass,the weight of adipose tissues in retroperitoneal, peri-epididymal and interscapular regions and the maximal diameter of adipocytes were measured,β3- AR mRNA levels in the adipose tissues above were measured with the method of RT-PCR.RESULTS:According to intention-to-treat analysis, thirty-two male rats entered result analysis. [1]Body mass: Increased weight of rats in 1.2 g/kg and 2.4 g/kg Oolong tea groups were significantly lower than that in the rats of control group and rats in 0.4 g/kg Oolong tea group (58.4±46.7,68.1±30.4,125.7±34.4,96.3±26.2,P < 0.01), but the amount of total diet consumption was similar in each group (P > 0.05). [2]Lipid coefficient in retroperitoneal and peri-epididymal regions of rats in 1.2 g/kg and 2.4 g/kg Oolong tea groups was lower than that in the rats of control group and rats in 0.4 g/kg Oolong tea group,(1.57±0.53,2.14±0.90 to 2.71±0.49,2.50±0.53, 1.14±0.38,1.43±0.53 to2.00±0.00,1.88±0.35), but there was no significant difference among groups of the ratio in inter-scapular regions (P >0.05). [3]The maximal diameter of adipocytes: The maximal diameter in retroperitoneal, periepididymal and inter-scapular regions of the rats in 0.4 g/kg, 1.2 g/kg and 2.4 g/kg Oolong tea groups was significantly lower than that in the rats of control group[(113±24), (86±29), (90±23), (120±30)μm;(94±20), (80±18), (64±17), (111±21)μm; (24±11), (21 ±11), (22±10),(27±11)μm,P < 0.05]. [4]β3-AR mRNA levels in adipose tissues:The β3-AR mRNA levels in retroperitoneal, peri-epididymal and interscapular regions of the rats in 1.2 g/kg and 2.4 g/kg Oolong tea groups were significantly higher than those in rats of control group and rats with0.4 g/kg of Oolong tea (0.72±0.11,0.64±0.112,0.40±0.08,0.34±0.10 for retroperitoneal region, 1.06±0.21,1.02±0.24,0.42±0.15,0.43±0.11 for epididymal region, 1.01±0.42,0.70±0.17,0.42±0.10,0.49±0.16 for interscapular region, P < 0.01).CONCLUSION:Oolong tea was of weight~reducing effect,which may be related to its effect to increase β3-AR mRNA level The middle dose (1.2 g/kg) may be optimal.These results may be helpful to the theory of weight reducing with tea and the selection ofoptimal dose.
4.Clinical research progress on the efficacy and safety of bevacizumab in treating ad-vanced non-small cell lung cancer

Antiangiogenesis therapy is one of the most common anticancer therapies. Bevacizumab is a monoclonal antibody that blocks the binding of the vascular endothelial growth factor to its high-affinity receptors. It is the only antiangiogenic agent approved for the first-line treatment of advanced non-small cell lung cancer (NSCLC). Many recent studies have attempted to determine the suit-able partners of bevacizumab in first-line treatment of NSCLC and evaluate its efficacy and safety as a second-line or beyond and con-tinuous treatment of beyond disease progression in patients with advanced NSCLC. This review summarizes current clinical research about the efficacy and safety of bevacizumab in the treatment of advanced NSCLC.
5.A preliminary study on visual cortex and optic radiation with diabetic retinopathy by 1H-MR spectroscopy

Objective To study the metabolic change of proton magnetic resonance spectroscopy (1 H-MRS)in the visual cortex and optic radiation region of patients with diabetic retinopathy (DR).Methods 1H-MRS was performed in 20 patients with DR and 20 healthy volunteers on GE 1.5 T MR system respectively.Metabolic peaks of N-acetylasparte(NAA),creatine(Cr,in 3.02 and 3.94 ppm),choline-containing compounds(Cho)and myo-inositol(mI)were observed,and the ratios were analyzed by each other.Independent-samples t test was performed between two sets of data.Results In both visual cortex and optic radiation,the ratios of ml/Cr and mI/CrSee in DR group(0.664 ± 0.052 and 1.453±0.068 in visual cortex,0.717±0.074 and 1.484±0.114 in optic radiation)were significant higher than those in normal group(0.602±0.047 and 1.249±0.044 in visual cortex,0.679 ± 0.075 and 1.334±0.089 in optic radiation,P<0.05).In DR group,the ratios of CrSec/Cr,Cho/Cr and NAA/Cr in visual cortex and optic radiation were 0.458±0.043 and 0.488±0.052,0.481±0.057 and 0.807±0.110,1.633±0.105 and 1.709±0.140 respectively.In control group.the ratios of those were 0.484±0.041 and0.502±0.056,0.471±0.065 and 0.786±0.109,1.625±0.098 and 1.716±0.135 respectively.The ratios of CrSec/Cr,Cho/Cr and NAA/Cr had no statistic difference between two groups(P<0.05).Conclusion mI/CrSec is a typical change in the visual cortex and optic radiation region,1H-MRS as a noninvasive examination could provide biochemical and metabolic informations for diabetic patients.
6.Relative study of chemosensitivities of peripheral blood lymphocytes and cancer cells of lung cancer in vitro

Objective To evaluate the correlation of chemosensitivity of cancer cells and peripheral blood lymphocytes to 8 chemotherapy agents in vitro,which was helpful to guide clinical chemotherapy for patients with lung cancer. Methods The chemosensitivity of the peripheral blood lymphocytes and tumor cells to 8 clinical routine che-motherapeutic agents was tested in 40 cases of lung cancer by using MTT method. SPSS 10. 0 was used to analyze the above indicators. Results There was no significant difference in the sensitivity of 8 clinical routine chemotherapeutic agents between peripheral blood lymphocytes and tumor cells of patients with lung cancer(P > 0. 05) ,which was posi-tive correlated to chemotherapeutic drugs between peripheral blood lymphocytes and tumor cells. Conclusion The chemosensitive test of the peripheral blood lymphocytes with MTT method in vitro was valuable for reference of selec-ting anticancer drugs in clinic for patients with lung cancer.
7.Analysis of food allergen spectrums in food anaphylaxis in Guangzhou

Objective To analyse distribution of age and gender characteristics of specific IgG(sIgG)antibodies and specific IgE (sIgE)antibodies of 13 types of food allergens in patients with food anaphylaxis,and to explore the relationship between sIgG and sIgE in food anaphylaxis.Methods 314 cases of patients from 2009 to 2012 were selected as subjects,and divided into underage group(1 63 cases)and adult group(1 5 1 cases).Serum sIgG of 13 types of food allergens were detected by using enzyme linked immu-nosorbent assay,serum sIgE of these food allergens were detected by using immune capture.Results 80.25% of the patients were sIgG-positive,and no obvious gender differences were found;while the positive rates of sIgG in the underage group(94.48%)were higher than that in the adult group(64.90%),there were statistically significant differences(P < 0.05 ).34.39% of the patients were sIgE-positive.The positive rates of sIgE in male patients(40.68%)were higher than that in female patients(26.28%),and that in the underage group(55.21%)were also higher than that in the adults group(1 1.92%),there were statistically significant differences(P <0.05).Conclusion The total positive rates and its distribution characteristics of sIgG and sIgE of same food aller-gens were obviously different.Food anaphylaxis might be associated with age,gender,food types and individual diversity.
8.Clinical effects of intense pulsed light on superficial strawberry hemangioma in proliferative phase

Objective To evaluate the clinical effect of intense pulsed light(IPL) on infantile superficial strawberry hemangioma in proliferative phase.Methods 68 patients,with age range from 1 day to 24 months,received IPL therapy with a proper energy and intervals.The therapeutic effects of the patients were evaluated through comparison of the photographs before and after treatment.Results AII patients with IPL therapy had a general effective rate of 76.47%.Few patients had adverse effects such as burned vesicle, hyperpigmentation,etc.Conclusion The intense pulsed light can be used as an earlier therapeutic scheme for infantile superficial strawberry hemangioma in proliferative phase.
9.Fingerprint analysis of Flos Genkwa by HPLC

Objective To establish the chromatographic fingerprint analysis for the quality control of Flos Genkwa.Methods RP-HPLC Method was applied to establish the chromatographic fingerprint.The separation was performed on a Kromasil C18 column(250 mm?4.6 mm,5 ?m)with a gradient elution composed of methanol and 0.05% phosphate acid.The column temperature was set at 35 ℃ and the flow rate was 1.0 mL/min.The detective wavelength was at 238 nm.Results Twenty-one characteristic peaks were established in the fingerprint.The mutual model of Flos Genkwa was established and the similarities were calculated.The similarity of 17 samples in all the 19 samples was more than 0.90.Conclusion The method is simple and accurate with good reproducibility.It can be used for the quality control of Flos Genkwa.
10.Study on chemosensitivity test in vitro between peripheral blood lymphocytes and tumor cells in elderly lung cancer

Objective To explore the correlation of chemosensitivity in vitro of anti-cancer drugs between peripheral blood lymphocytes and tumor cells in non-small cell lung cancer of elderly patients.Methods The MTT method was used to test the sensitivity of the peripheral blood lymphocyte and the tumor cells of 52 patients with nonsmall cell lung cancer to 13 kinds of anti-cancer drugs.Results The sensitivity test in eleven drugs include CDDP,CBP,LOHP,PTX,CTX,ICTX,THP,VP-16,GEM,VCR and NVB in the peripheral blood lymphocyte were associated with that in the tumor cells.But no dependablity in two drugs,ADM and HCPT.Conclusion The peripheral blood lymphocytes could be replace tumor cells to test the chemosensitivity of anti-cancer drugs in patients with non-small cell lung cancer.
