ObjectiveTo investigate the genomic and non-genomic effects of 17β-estradiol on gallbladder smooth muscle strips of guinea pig and their possible mechanism. MethodsAfter ovariectomized operation (OVX) and subcutaneous injection of 17β-estradiol, the contents of serum estradiol and cholecystokinin-octopeptide (CCK-8) of the sham operation group (Sham) guinea pigs, the OVX group, the OVX and subcutaneous injection 17β-estradiol for 1 day (OVX+E2, 1 d), 3 days (OVX+E2, 3 d) and 7 days (OVX+E2, 7 d) were detected by EILSA respectively. The effects of CCK-8/Ach on constriction of gallbladder muscle strip were observed among various groups by using tension transducer, and the acute effects of 17β-estradiol on guinea pig gallbladder smooth muscle strips were observed to probe its possible mechanism. ResultsCompared with the Sham group, the serum contents of estradiol and CCK-8 decreased in OVX group (P< 0. 05) whereas the sensitivity of OVX guinea pigs gallbladder muscle strips to CCK-8/Ach increased (P<0.05). With the extension of the subcutaneous injection 17β-estradiol time (for 1, 3, 7 days), both the serum estradiol and CCK level increased (P< 0.05) while the guinea pig gallbladder strips sensitivity to CCK-8/Ach decreased (P<0.05).17β-estradiol at concentration ranging from 10 9 to 10-7 mol/L have no effect on the guinea pig gallbladder strips contraction (P>0.05), but at concentration of 10-6 and 10 5 mol/L, it can inhibit the gallbladder contraction (P<0.05). The blocking agents, such as nimodipine, atropine, devazepide, ICI 182,780 and Y-27632, can block the inhibited effects of 17β-estradiol. ConclusionThe 17β-estradiol can affect the gallbladder motility, both by genomic and non-genomic pathway.

Objective To study the effects of butylphthalide (NBP) on memory and apoptosis related protein as well as neuronal pathology in hippocampus of vascular dementia (VD) rats. Methods VD model was generated by the permanent occlusion of bilateral common carotid arteries in SD rats to produce the forebran ischemia. Male SD rats were randomly allocated into sham-operation group, VD model group, NBP treatment group and nimodipine treatment group. The function of memory was tested by the Morris water maze. The neuronal pathological changes and the expression of Bcl-2 and Bax proteins in the hippocampus were observed with hematoxylin-eosin (HE) staining and immunohistochemical staining, respectively. Results The impaired memory of VD rats was proved by the lengthened mean escape latency [(78.79±21.93)vs.(16.96±7.44),P<0.05] and the neuron in hippocampus was severely damaged. The decveased ratio of Bcl-2/Bax resulted from the overexpression of Bax proteins in VD model group versus the sham-operation group [(43.00±6.72)vs.(6.00±1.29),P<0.05]. The treatment of NBP notably improved the memory function of VD rats and reduced the hippocampus pathological injury (P<0.05). The expression of Bcl-2 protein raised [(33.14±8.05)vs.(21.81±4.97),P<0.05] along with reduced expression of Bax protein [(32.93±4.99)vs.(43.00±6.72),P<0.05] after NBP treatment. However, there was no significant difference in the treatment effects between nimodipine and NBP group (P>0.05). Conclusions NBP treatment could improve memory of VD rats and reduce the hippocampus pathological lesion by inhibiting the apoptosis related protein.

Objective To investigate the correlation between invasion ability and cytoskeleton remodeling of hepatocarcinoma cell by atomic force microscopy (AFM) and to explore mechanical properties during genesis, development and invasion of hepatocellular carcinoma (HCC).Methods Four HCC cell lines (MHCC-97H,MHCC-97L,SMMC-7721 ,Huh-7 )with different invasive ability were studied.Mechanical parameter (Young′s modulus)was measured by AFM.The pattern of cytoskeleton remodeling of HCC cell lines with different invasive ability was detected by immunofluorescent staining. The difference of cell invasive ability was tested by cell scratch experiment in other to verify mechanical data.Kruskal-Wallis H test was used to compare the differences between groups. Results The results of AFM indicated that Young′s modulus of cytoplasma area and nucleus area decreased gradually as cell invasion ability increased (χ2 =472.78,622.43,both P <0.01).According to invasive ability from low to high,Young′s modulus of cell cytoplasm area of Huh-7,SMMC-7721 ,MHCC-97L and MHCC-97H were 1 602.43 (845 .48,3 317.25)Pa,1 055 .28 (367.48,2 280.77)Pa,1 026.78 (369.20,2 019.96)Pa and 503.12 (366.11 ,700.31)Pa,respectively.Young′s modulus of cell nucleus area of Huh-7,SMMC-7721 ,MHCC-97L and MHCC-97H were 2 823.98 (1 262.78,4 440.07 )Pa,1 313.43 (590.71 , 2 678.62)Pa,1 285 .17 (583.29,1 961 .19)Pa and 655 .57 (441 .29,943.39)Pa,respectively.The results of immunofluorescent staining showed that the stronger the cell invasive ability, the worse cytoskeletal integrity and more irregular cell microfilament distribution. In cell scratch assay, the migration rate of MHCC-97H was 46.67% in 24 h and 86.47% in 48 h,that of MHCC-97L was 45 .70%in 24 h and 82.86% in 48 h,that of SMMC-7721 was 39.41 % in 24 h and 79.85 % in 48 h and that of Huh-7 was 34.60% in 24 h and 72.09% in 48 h,which showed that the cell migration orderly in creased as the cell invasion ability increased. Conclusions It seemed that HCC with higher invasive ability had lower Young′s modulus, softer cell, stronger deformability, worse cytoskeleton integrity and more irregular cell structure, and vice versa.

AIM: To explore the effects of sodium ferulate (SF) on function of macrophages in colonic tissue of the colitis rats in vivo. METHODS: The immunological colitis model of rats was produced. SF was used intracolonically for 21 days. The contents of malondialdehyde (MDA), nitric oxide (NO), prostaglandin E_2 (PGE_2) and the activity of superoxide dismutase (SOD), interleukin-1 (IL-1), TNF-?, myelopexoxidase (MPO), and the expression level of NF-?B p65 in colonic tissue of the rats were detected. RESULTS: SF (200,400,800 mg/kg) decreased the elevated contents of MDA, NO, PGE_2, the activity of IL-1, TNF-?, MPO, and the expression level of NF-?B p65, while increased the reduced activity of SOD in colonic tissue of the colitis rats in a dose-depended manner. CONCLUSION: SF restrained the activity of activated colonic macrophages and relieved the colonic inflammation reaction in vivo in colitis rats, which may be related to the suppression of NF-?B activation.
