1.Relationship between DNA damage repair and radioresistance in nasopharyngeal carcinoma CNE-2R cells

ABSTRACT:Objective To investigate the differential gene expression of nasopharyngeal carcinoma cell line CNE-2 and CNE-2R exposed to X-rays.Methods We used neutral comet assay to detect the intrinsic radiosensitiv-ity of tumor cells,immunofluorescence to detect the phosphorylated histone γH2AX and Rad5 1 foci formation and to assay the time-dose effects on cell radiation injury and radiation radiosensitivity change after irradiation.The dif-ferential gene expression was screened with cDNA microarray by DNA damage repair related gene chip.Differenti-ally expressed proteins screened were confirmed by Western blot.Results After 2 h of 4 Gy 9MeV-β-ray irradia-tion,CNE-2R cells had aggravated DNA damage compared to CNE-2 cells and the damage was more obvious with the prolongation of time.It suggested that cell DNA damage severity was more obvious.The immunofluorescence showed the positive rates ofγH2AX cells in CNE-2 cell line were higher than those in CNE-2R cell line.There were 37 different genes related to DNA damage repair in CNE-2 cell line compared with CNE-2R cell line,while 24 up-regulated and 13 down-regulated genes were found in SiHaR cells.In them,there were 6 genes whose ratios were 6 times higher and 3 genes whose ratios were lower than 0.1.Western blot verified the results of four differentially expressed proteins compared with CNE-2 cells,in which GADD45a and RRAD1 expression levels were significantly lowered in CNE-2R while the RAD9a and RCF2 protein significantly increased.Conclusion The radioresistance of CNE-2R cells is significantly related to DNA damage repair capacity in certain mutations at the gene level.It is possible to regulate cell radiosensitivity by regulating the related gene.
2. Active circulation vs single perfusion and single infusion for hyperthermic intraperitoneal chemotherapy in patients with peritoneal effusion of ovarian cancer

Objective: To compare the efficacy between active circulation mode and single-irrigation and single-pumped mode for hyperthermic intraperitoneal perfusion in ovarian cancer patients with intraperitoneal effusion. Methods: Fifty-three ovarian cancer patients with intraperitoneal effusion treated with active circulation mode of hyperthermic intraperitoneal perfusion between March 2016 and May 2017 were taken as study group, and another 51 ovarian cancer patients with intraperitoneal effusion treated with single-irrigation and single-pumped mode of hyperthermic intraperitoneal perfusion between July 2015 and February 2016 were taken as control group. All of the patients received hyperthermic intraperitoneal chemotherapy with paclitaxel and cisplatin. Each treatment was administered 3 times and treated for 2 cycles. The temperature duration time of perfusion fluid into and out of the abdominal cavity and the response rate of two groups were compared. Results: The temperature threshold of perfusate into the abdominal cavity of two groups can be controlled between 43.0-43.5 °. The abdominal temperature of study group was 41.0-41.5 ° and remained constant. The response rates of peritoneal effusion in study group and the control group were 86.8% (46/53) and 64.7% (33/51), respectively; there was a significant difference between the two groups (P < 0.05). Except for abdominal pain, the incidence rates of adverse reactions had no statistical differences (all P > 0.05). Conclusion: Active circulation mode of hyperthermic intraperitoneal perfusion has the advantages of constant temperature, short treatment time, high safety and remarkable curative effect in the treatment of peritoneal effusion of ovarian cancer, and is worthy of promotion in clinical practice.
3. Application of triple exits linker for temperature regulation of ovarian cancer patients with ascites during hyperthermic peritoneal perfusion

Objective: To investigate the efficacy of triple exits linker for temperature regulation of ovarian cancer patients with ascites in hyperthermic intraperitoneal perfusion using HGGZ-102 type body cavity heat perfusion machine. Methods: From April 2013 to September 2014, 39 ovarian cancer patients with ascites did not use triple exits linker (as the control group). From October 2014 to May 2015, 45 ovarian cancer patients with ascites used triple exits linker during hyperthermic intraperitoneal perfusion (as the study group). The patients in two groups were treated with TP regimen (paclitaxel on d 1, and hyperthermic peritoneal perfusion with cisplatin on d 1 and d 8; repeated every 3 weeks with a total of six cycles). The temperature of the drugs inflowing into abdominal cavity and outflowing from abdominal cavity in two groups was examined. Results: In the study group, the threshold temperatures of the drugs inflowing into abdominal cavity for three cycles were precisely controlled at 43.5-44.5 , and the maximal temperature when drugs outflowing from abdominal cavity reached target temperature (39.5-40.5 ). The effective rates of ascite subsided were 71.1% and 46.2% in the study group and the control group, respectively. There was significant difference between the two groups (P < 0.05). The incidence rate of adverse reaction between the two groups had no statistically significant difference (P > 0.05). Conclusion: Application of triple exits linker for temperature regulation of ovarian cancer patients with ascites in hyperthermic intraperitoneal perfusion can control the temperature accurately and improve the therapeutic effect.
4.Preliminary study of superb microvascular imaging monitoring angiogenesis in tibial transverse transport

Objective:To investigate the application value of superb microvascular imagingin monitoring angiogenesis in the treatment of chronic ischemic diseases of lower limbs by tibial transverse transport.Methods:From May 2019 to December 2020, 12 patients with ischemic diseases of lower limbs who received tibial transverse transport therapy were retrospectively analyzed, including 7 male patients and 5 female patients, aged 30-86 years, with an average age of 64.0±16.6 years. Among them, there were 10 patients with diabetic foot, with a disease course of 5-30 years (mean 14.4±8.3 years), and 2 patients with arteriosclerosis obliterans. The results of lower extremity superb microvascular imaging before and after treatment were recorded and compared.Results:After treatment, 12 patients were found to have obvious new vessels around the artery of lower extremity, the number of vessels 1-8, the average number of 3.25±2.73. We found that a total of 12 patients 20 inherent lower limb artery collateral established, collateral circulation was found around 12 dorsal metatarsal artery (DMA) in 11 patients, around 3 dorsal pedis artery (DPA) in 2 patients, around 3 anterior tibial artery (ATA) in 2 patients, and around 1 posterior tibial artery (PTA) in 2 patients. The lateral branches were established in ipsilateral lower limbs in 6 patients, contralateral lower limbs in 2 patients, and bilateral lower limbs in 4 patients.Conclusion:The phenomenon of angiogenesis is obvious in the clinical application of tibial transverse transport. Superb microvascular imaging can effectively reflect the degree of promoting the establishment of collateral circulation by tibial transverse transport.
5.Effects of different ventilation modes on respiratory mechanics, hemodynamics and biochemical metabolism in patients undergoing laparoscopic colorectal cancer surgery

Objective:To compare the effects of pressure controlled ventilation (PCV) and volume controlled ventilation (VCV) on respiratory mechanics, hemodynamics and biochemical metabolism in patients undergoing laparoscopic colorectal cancer surgery.Methods:The clinical data of 78 patients underwent laparoscopic colorectal cancer surgery from August 2019 to June 2020 in Inner Mongolia People′s Hospital were retrospectively analyzed. Among them, 39 patients were treated with PCV (PCV group), and 39 were treated with VCV (VCV group). The respiratory mechanics, hemodynamics and biochemical metabolism indexes 10 min after anesthesia induction (T 1), 10 min after pneumoperitoneum + low head and foot height (T 2), 60 min after pneumoperitoneum + low head and foot height (T 3) and 120 min after pneumoperitoneum + low head and foot height (T 4) were recorded. The respiratory mechanical indexes included mean airway pressure (P mean), airway peak pressure (P peak), pressure of end tidal carbon dioxide (P ETCO 2) and dynamic lung compliance (C Ldyn); hemodynamic indexes included mean arterial pressure (MAP) and heart rate; and biochemical metabolic indexes included base excess, serum natrium, serum potassium, negative logarithm of the hydrogen ion concentration (pH) and blood glucose. The MAP and heart rate T 1 to T 4 in PCV group were significantly lower than those in VCV group, and there was statistical difference ( P<0.01 or <0.05). The base excess, serum natrium, serum potassium and pH T 1 to T 4 in PCV group were significantly higher than those in VCV group, while the blood glucose was significantly lower than that in VCV group, and there was statistical difference ( P<0.01 or <0.05). Conclusions:Compared with VCV, PCV can promote the recovery of respiratory mechanics index, stabilize hemodynamics and improve biochemical metabolism index in patients undergoing laparoscopic colorectal cancer surgery.
6.Expression of Ki67 and clinicopathological features in breast cancer.

OBJECTIVETo analyze the relationship between the expression level of Ki67 and clinicopathological features in breast cancer.
METHODSData of 918 female patients with invasive ductal breast carcinoma treated in the Cancer Hospital, Chinese Academy of Medical Sciences from Jan. to Dec. 2010 were analyzed retrospectively. The correlation of Ki67 expression and other clinicopathological features in the breast cancer was analyzed.
RESULTSAmong the 918 cases, the Ki67 index was 0.9% to 95% (mean value 27.8%). Taking the Ki67 index 14% as the boundary to divide the patients into two subgroups, 263 cases (28.6%) were ≤ 14%, and 655 cases (71.4%) were >14%. There were significant differences between the Ki67 expression and age, tumor size, axillary lymph nodes status, histological grade and the expressions of C-erbB-2, estrogen receptor (ER) and progesterone receptor (PR) (P < 0.05 for all). All the Ki67 indexes of Ki67 expression in luminal B (30.44%), HER-2 overexpression (36.77%) and triple negative (47.40%) subtypes were significantly higher than that in the luminal A subtype (21.36%)(P < 0.01). The expression level of Ki67 in triple-negative subtype (47.40%) was significantly higher than that in the non-triple-negative subtype (24.79%)(P < 0.001).
CONCLUSIONSKi67 index is significantly correlated with the age, tumor TNM stage, axillary lymph node status, histological grading, ER status, PR status and HER-2 status. A high expression level of Ki67 is a poor prognostic factor for breast cancer. The expression level of Ki67 should be detected routinely and it may become a useful prognostic marker in the treatment of breast cancer.

7. Clinical application of the thumb-tack needle for subcutaneous embedding therapy on treating insomnia with insufficiency of heart and spleen among breast cancer patients

Objective:
To explore the clinical therapeutic method and effect of the thumb-tack needle for subcutaneous embedding therapy on treating insomnia with insufficiency of heart and spleen among breast cancer patients.
Methods:
Totally 80 breast cancer patients with insomnia with insufficiency of heart and spleen treated in department of oncology, the Second Affiliated Hospital of Xi′an Jiao Tong University from March 2018 till November 2018 were selected and assigned into an experiment group and a control group by random number table, 40 cases in each one. The experiment group applied thumb-tack needle for subcutaneous embedding therapy, selecting Shenmen, Sanyinjiao, Zusanli, Anmian as major acupoint and Taibai as assistant acupoint. The control group used fake needles at the same places. After four-week treatment, we evaluated the therapeutic effect using the Pittsburgh Sleep Quality Index (PSQI) and observed side effects.
Results:
Before treatment, no significantly statistical differences were observed between the two groups on global and each dimension′s PSQI scores (
