1.Experimental research on Chinese herbal prescriptions to treat rheumatoid arthritis and their anti-inflammatory mechanism.

Objective:To investigate the effects of Chinese medicine prescriptions Gancaofuzi-tang and Tongbi-fang on adjusting the dynamic expression of related cytokines and further ameliorating the histopathology of synovitis in rats with Ⅱ collagen induced arthritis(CIA).Methods:The experimental Wistar rat model of CIA was set up successfully.The rats divided into five groups in random: normal(N),Gancaofuzi-tang treatment(GF),Tongbi-fang treatment(TB),Puwei western medicine treatment(PW) and model control(C).The different medicine was affused into stomach 1.5 ml/rat,once daily.The rats were sacrificed after 6 weeks of medicine administration.Foot swelling degrees of the rats were measured every other day,and then calculate swelling rates.Getting blood after 3 weeks of treatment from rat's innercanthus to detect the level of serum Sialic Acid(SA) and the level of serum IL-2 and TNF-? by ELISA.The synovial histopathology was observed with HE staining under optical microscope.Results:Gancaofuzi-tang and Tongbi-fang significantly decreased swelling rate of joints in CIA rats.The pathology of synovisl tissues was markedly ameliorated.After administration for 3 weeks,the levels of serum SA,IL-2 and TNF-? of the rats in GF and TB groups were significantly decreased compared with the control.Continuous administration for 6 weeks,the levels of the targets in GF and TB rats reached to near the normal level.Conclusion:All the results demonstrate that Gancaofuzi-tang and Tongbi-fang play anti-inflammatory and immunoregulatory roles in CIA rats through inhibiting activation of some inflammatory mediators,adjusting related cytokines expression,then eliminating the joint swelling and improving synovitis pathology,which might be looked as immune suppression effects similar to NSAID medicine.

3.Sedative and hypnotic effects of oil of spine date seeds from different extraction technologies in mice

Objective To study the sedative and hypnotic effects of oil of spine date seeds by different extraction technologies(compression and supercritical CO2 extraction,OSDSE and OSDSS) in mice.Equivalence test was used to test the effects.Methods Spontaneous activity and tolerance were observed by photoelectric counting method after long-term intragastric administration of OSDSE and OSDSS in mice.The sleeping time and delitescence induced by above threshold dose of pentobarbital sodium and sleeping number induced by below threshold dose of pentobarbital sodium were utilized to observe the hypnotic effect of OSDSE and OSDSS in mice.Diazepam was applied as a positive control drug.Results OSDSE and OSDSS could obviously reduce spontaneous activity,increase sleeping time and sleeping number in mice,while the sleeping delitescence was shortened.All the effects induced by OSDSE and OSDSS were equivalent.The inhibitory effect of spontaneous activity was enhanced following repeated administration,while the effect of diazepam was reduced.Conclusion Both oils of spine date seeds can induce sedative and hypnotic effects without tolerance.They have equivalent effects on sedation and hypnosis.
4.Study on establishing type Ⅱ collagen induced arthritis model in rats

Objective:Research on establishing a better experimental arthritis rat model of cold-dampness arthraligia on rheumatoid arthritis(RA) research.Methods:In order to induce arthritis,the rat's feet were injected with different concentrations of type Ⅱ collagen mixed with Freund's adjuvant;measure the feet swelling degrees of rats,then calculated swelling rate;detect the level of serum sialic acid(SA);observ the synovial histopathology and ultrastructural of rats.Results:The rat joints swell of the group with low concentration of type Ⅱ collagen and completed Freund's adjuvant was rapid and severe,while that of the group with high concentration of type Ⅱ collagen and incompleted Freund's adjuvant was moderate and lasting.The level of serum SA of model groups was higher than the normal group.The histopathology changes of rat's synovium conform to RA.Conclusion:The group injected with high concentration of type Ⅱ collagen and incompleted Freund's adjuvant is the ideal RA model.It is belonged to the symptom of cold-dampness arthralgia,compared with this model,the AA approaches the symptom of heat-dampness arthralgia.
5.Cloning of hTERT promoter and its specific transcriptional activity in MCF7 breast cancer cell

Background and purpose:Gene therapy is a novel approach for the treatment of the patients with breast cancer. One of the effective ways is to direct transgenic expression to specific tissues or tumors with the use of tissue-specific-promoters (TSP). hTERT (human telomerase reverse transcriptase) is highly expressed in many types of cancers including breast cancer. Thus, we hypothesized that the hTERT promoter targeting with gene therapy vectors could be exploited for breast cancer. In this study, we amplified hTERT gene promoter and cloned it into the reporter vector pEGFP and pGL3-Basic. Afterwards, the specific transcription of hTERT promoter in MCF7 cells was evaluated. Methods:hTERT gene minimal promoter was PCR amplified and cloned into the reporter plasmid pEGFP-1 and pGL3-Basic.The constructs pEGFP/TERT and pGL3/TERT were transfected into MCF7 breast cancer cells and HBL100 human epithelial cells, respectively.The expression of EGFP and luciferase were investigated, respectively..Results:pEGFP/TERT and pGL3 /TERT bearing hTERT gene promoter were constructed. The specific expression of EGFP was detected in MCF7 cells while little expression of EGFP was seen in HBL100 cells.In accordance with EGFP, luciferase driven by hTERT also showed specific and high activity in MCF7 cell (RLU/U: 33784), which is 15 times higher than in HBL100 (RLU/U: 2400).Conclusions:The high transcriptional activity of hTERT gene promoter in MCF7 cell indicates its potential utility as a novel candidate for transcriptional targeting of breast cancer.
6.Regulation of MCP-1 in Osteoblasts by PTHrP in Breast Cancer Cells

Objective:To identify the regulation of monoeyte chemoattractant protein-1(MCP-1)in osteoblasts by parathyroid hormone related protein(PTHrP) in breast cancer cells.Methods:Osteoblast-like UMR106 cells were treated with 10~(-8)mol/L PTHrP (1-34) for different times and the expression of MCP-1 was studied by semiquantitative RT-PCR.Recombinant shRNA plsmid PTH1R/pRNAT was constructed and transfected into UMR106 cells.Stable PTH1R/UMR106 cells were established,in which PTH1R expression was knocked-down.The conditioned medium(CM) of breast cancer MDA-MB231 was added to UMR 106 and PTH1R/UMR106 cultures respectively.Thereafter,mRNA of MCP-1 was examined by semiquantitative RT-PCR.Results:PTHrP induced expression of MCP-1 in UMR106 cells and the induction began at 3 h and reached the maximum at 6 h.Treatment of UMR106 cells with CM of MDA-MB231 also significantly induced mRNA of MCP-1 at 6 h(1.238 1±o.115 5 and 0.598 4±0.036 4,P<0.05).This effect was partly blocked by the knockdown of PTH1R in UMR106 cells(0.867 2±0.045 7,P<0.05).Conclusion:The osteoblastic MCP-1 is up-regulated in breast cancer through expression of PTHrP.
7.Sodium Phenylacetate Enhances HLA Molecule Expression on the Surface of Tumor Cells

Sodium phenylacetate can induce differentiation of tumor cells and has been approved as a drug for the treatment of adults with cancer. In order to explore the immunological mechanism of its antitumor effect, the influence of sodium phenylacetate on HLA class I and II molecule expression in various human tumor cell lines, including breast adenocarcinoma (MCF-7.MUA-453), gastrocarcinoma(MKN-28,MKN-45), ovarian cancer(3AO) and cervical cancer(Hela), was studied with ELISA. The result showed that HLA class II molecule was absent from the surface of MCF-7 cells, but they could be induced after 7 days of continued treatment with sodium phenylacetate. Sodium phenylacetate was found to increase HLA class I molecule expression on the surface of MCF-7 cells, HLA class 1 and II molecule expression on the surface of MDA-453、 MKN-28、 MKN-45、 Hela and 3AO cells. The effect of sodium phenylacetate on HLA class I molecule expression in tumor cells is dose-and time-dependent.
8.Anergy and Characteristics of Human T Cells Induced by Combination with B7-1 mAb and CsA

To study the induced condition and characteristics of T cell anergy in vitro. Methods: Anergic Tcell was induced by combination of B7-1 mAb and cyclosporin A (CsA) in vitro, cytokine gene of anergic T cells was detected by RT-PCR. Results: T cell anergy was antigen-specific. The state of T cell anergy can be reversed by PHA, CD3 mAb and PMA plus A23187. IL-2 can prevent the induction of T cell anergy, but it can not reverse the state of un-responsiveness. IL-2 and IFN mRNA can not express in anergic T cells. In contrast, IL-4 and IL-10 mRNA were detectable. Conclusion: T cell anergy can be induced in vitro , cytokine profile of anergic T cells deviated to Th2-like phe-norype.

Based on an analysis of the current condition of emergency department facilities in domestic urban hospitals and lessons drawn during the SARS outbreak,the paper argues that due attention ought to be paid to the spread of infectious diseases resulting from the inherent drawbacks in the construction of emergency departments.It presents constructive suggestions on ways in which construction of and medical procedures in emergency departments could meet the requirements for infectious disease prevention and treatment.
10.The change of morphology and airflow dynamics in upper airway by the use of oral appliance in OSAHS patients

Objective:To analyse the change of morphology and internal air flow in upper airway by the use of oral appliance(OA)in patients with obstructive sleep apnea hypopnea syndrome (OSAHS).Methods:A 46-year-old male patient with OSAHS accepted CT scan before and three months after use of OA.Computational fluid dynamics(CFD)model was built on the base of CT scans by Mimics 10.01 and ANSYS ICEMCFD14.0.The internal flow of upper respiratory tract was simulated by ANSYS-FLUENT 14.0 and the re-sults was analyzed by ANSYS-CFD-Post14.0.Results:The most narrow area of upper airway was located in the lower bound of pha-ryngopalatiae,and it augmented from 0.119 4 cm2 to 0.409 9 cm2 after wearing OA;the maximum air velocity was decreased from 11 . 087 m/s to 8.204 m/s,the minimum negative pressure was decreased from -83 Pa to -59 Pa,the resistance of cavum pharyngis de-creased from 250 Pas/L to 145 Pas/L.Conclusion:Application of OA may expanse the upper respiratory tract,decrease the negative pressure and resistance of the upper respiratory tract in narrow area,reduce the collapsibility of the upper airway and maintain the pa-tency of the airflow.
