2.Effect of India buead and coix seed powder on phlegm-dampness constitution dyslipidemia in population studies

Objective To explore the anomalous effects of India buead and coix seed powder on phlegm-dampness constitution dyslipidemia through population-based intervention studies. Methods According to the experimental results of the model group with dyslipidemia, the phlegm-dampness population meeting the inclusion criteria were chosen as the samples for the randomized controlled trial, who were divided into test meal (21 males and 39 females) and blank control (23 males and 37 females) groups with 60 cases in each, aged 18 to 65 y and the total of 120 cases completed the study. Statistical analyses were performed with SPSS 18.0 statistical software. The self comparison wass conducted by the method of paired-sample t test and the comparison between groups was conducted by using independent sample t test. The categorical statistics were described as frequency and compared throughc2 test, P<0.05 was considered as statistically significant. Without changing the original way of life, the meal group having taken the India buead and coix seed powder was compared independently and with the control group for TC, TG, HDL-C and LDL-C to detect the trends and degrees of the level changes. Meanwhile, according to efficacy tests and standards, the changes in the level of TC, TG and HDL-C have been evaluated and the efficiency and total effective rates of all the variables have been calculated. Results After the intervention, there was a declining tendency in each variable, 8.9%in TC, 21.4%in TG, and 27.2%in LDL-C, except for an increase of 0.13 mmol/L in HDL. There were significant differences in the variables before and after the intervention (P<0.05 in each). As for the test meal group after the intervention, the effectiveness of TC has been obtained in 25 cases and inefficacy in 35 cases, with efficiency of 41.7%;the effectiveness of TG has been shown in 53 cases and inefficacy in 7 cases, with efficiency of 88.3%;the effectiveness of HDL-C has been gained in26 cases and inefficacy in 34 cases, with efficiency of 43.3%; and the total effective rate was 57.8%. Conclusions There are significant changes and improvements in TC, TG, HDL-C and LDL-C in the test meal group after the intervention with the powder, which demonstrate that the powder is effective in the reduction of TG for animals with dyslipidemia and populations with phlegm-dampness constitution.
3.Expression of?-catenin in hepatic carcinoma and in human HepG2 and SMCC-7721 cells

Objective To study the expression and cellular distribution of?-catenin in hepatocellular carcinoma and in HepG2 and SMCC-7721 cells.Methods Immunohistochemistry,immunocytochemistry techniques and Western blotting analysis were used to examine the expression and distribution of?-catenin in the cell membrane,cytoplasm and nucleus of bepatocellular carcinoma tissues,HepG2 and SMCC-7721 cells.Results Immunohistochemical staining showed that the?-catenin poorly,unevenly,even not distributed on the mem- brane of hepatocellular carcinoma cells.The expression of?-catenin on the membranes of low differentiated liver cancer cells was lower than that in the highly differentiated liver cancer cells,but it was strongly expressed in the cytoplasm of hepatocellular carcinoma cells,The ex- pression of?-catenin in the membranes of hepatocellular carcinoma cells was found to be reduced obviously in 18 out of 20 patients(90%).?-catenin was found to be poorly,unevenly,even not distributed in the membrane cells in 10 patients(50%).An obvious accumulation, but with poor-distribution,of?-catenin was observed in the cytoplasm of 17 cases(85%).Significant differences in the?-catenin expres- sion in both membrane and cytoplasm were found among differently differentiated stages of liver cancer cells.In 7 of 9 cases(77.8%)with highly differentiated liver cancer cells,?-catenin was continuously and evenly distributed in both the membrane and cytoplasm,while in 4 of 5(80%)cases with poorly differentiated liver cancer cells the expression of?-catenin in the membrane was found to be significantly poor and showed a uneven distribution in the cytoplasm.The?-catenin expression was positive in the cytoplasm of 5 of 6(83.3%)cases with moderatdy differentiated liver cancer cells.No expression of?-catenin was found in the membrane of HepG2 and SMCC-7721 cells,while it was highly expressed in both cytoplasm and nucleus.Western blotting showed that the?-catenin expressed in both cytoplasm and nucleus of all the liver cancer tissue,and also in HepG2 and SM(3C-7721 cells.Conclusions The expression of?-catenin was down-regulated in the membrane of hepatocellular carcinoma cells and HepG2 and SMCC-7721 cells,while?-catenin was strongly expressed in cytoplasm,and it was obviously accumulated in the nucleus.The distribution of?-catenin was related to the stage of differentiation of cancer cells.

Objective:To explore the effect of parental rearing behavior and related factors of personality dysfunction in adolescents Method:By means of stratified cluster sampling, 9574 students from 25 representative senior high schools in urban and rural areas of Beijing were investigated by PDQ-4 (personality diagnostic questionnaire-4), EMBU and general information questionnaire A case control study based on community was carried out afterwards The cases were the students whose total PDQ-4 score was 42 (means?1 645s) or more The controls were students whose total PDQ-4 score was 25 (means) or less Cases of personality dysfunction (n=531) were individually matched with 1062 controls by age and gender in ratio of 1:2 The single variable and multivariate conditional logistic regression analyses were performed to screen out the risk factors of personality dysfunction Result: The single variable conditional logistic regression analysis revealed that parental rejection, overprotection, favoring subject, rural area, type of school, poor parental relationship and single parent family increased risk of personality dysfunction, while parental emotional warmth was a protective factor The multivariate conditional logistic regression analysis gave the same results with OR=11 43 for parental rejection, OR=7 68 for overprotection, OR=13 94 for rural area, OR=4 15 for poor parental relationship, and OR=0 50 for parental emotional warmth There was no association between personality dysfunction and nationality, family income, single child family or level of parental education Conclusion:Personality dysfunction in adolescents is closely related to family environment
5.Studies on the induction and differentiation of rat's hepatic stem cells in vitro

Objective To isolate hepatic stem cells from adult rats,and to examine the process and morphologic changes of isolated hepatic stem cells differentiated into hepatic cells and bile duct epithelial cells under in vitro culturing conditions.Methods AAF/PH was used to stimulate the oval cells.Selective enzymatic digestion and density gradient centrifugation were employed to isolate and purify these cells from heterogeneous liver cell population.The hepatic stem cells were induced to proliferate and differentiate into hepatic parenchymal cells by the addition of dexamethasone,DMSO and several growth factors.The morphologic changes of hepatic oval cells to that of hepatic cells and bile duct epithelial cells were examined.Immunofluorescence method was used to detect the hepatic stem cells c-kit and the expression of OV6 antigen.The albumin of hepatic stem cells and the CK19 expression were identified by RT-PCR analysis.Results The activity of freshly isolated oval cells were over 90%,with different colors and size.The diameter of oval cells was about 1/4~1/6 that of hepatocytes.Large and round hepatocytes appeared first during differentiation,and the cell morphology changed when the oval cells differentiated to hepatocytes.Freshly isolated oval cells was positive for c-kit and OV6 with immunofluorescence staining,and RT-PCR analysis indicated that the albumin and CK19 mRNA were expressed in the new isolated cells.Conclusion The oval cells could be induced to differentiate into a variety of cell lineages including hepatocytes and bile duct epithelial cells in the presence of DMSO and several growth factors.Several antigens such as c-kit,OV6,CK19 and albumin may be expressed in the freshly isolated hepatic stem cells,and the course of morphologic changes may be observed under in vitro conditions of induction and differentiation.
6.Change of inflammatory factors and its effect on prognosis in patients undergoing acute myocardial infarction thrombolysis treatment

ObjectiveTo explore the inflammatory factors C-reactive protein (CRP),interleukin (IL)-6,tumor necrosis factor (TNF)-α and white blood cell (WBC) count differences between acute myocardial infarction(AMI ) thrombolysis treatment and unthormbolysis treatment,and find out the relevance between the inflammatory factors and the prognosis.MethodsAccording to the condition of accepting AMI thrombolysis treatment,the 229 patients of AMI were divided into the thrombolysis group( 131 cases) and the unthrombolysis group(98 cases).The levels of myocardial troponin I (cTnI),creatine kinase(CK),creatine kinase isoenzyme-MB (CK-MB) were detected at the time of patients sent into the hospital for the immediate,6-hour later and 24-hour later.After 6-month's follow-up,prognosis was compared between two groups.ResultsTwenty-seven cases lost in the thrombolysis group.One case died within 6 months and the mortality was 1.0%(1/104) in the thrombolysis group,and 6 cases died within 6 months and the mortality was 6.1%(6/98 ) in the unthrombolysis group.There was significant difference between two groups (P < 0.05 ).The levels of CK,CK-MB in the thrombolysis group advanced,and compared with that in the unthrombolysis group,there were significant differences (P < 0.05 ).The levels of TNF-α,IL-6 in the thrombolysis group were significantly higher than those in the unthrombolysis group (P< 0.05),and CRP and WBC count had no significant differences between two groups (P > 0.05).The repatency rate was 79.4%( 104/131 ) in the thrombolysis group,the levels of TNF- α,IL-6 in repatency patients were higher than those in non-repatency patients.There were significant differences(P < 0.05 ).ConclusionsThe thrombolysis is an effective way to cure AMI.The increase of TNF- α,IL-6 after the thrombolysis is considered to be related to reperfusion injury,and CRP,IL-6,TNF-α and WBC count can forecast the inflammation of myocardial necrosis and take an impotant role in predicting the prognosis of the AMI.The antiinflammatory and antioxidation treatment is significant to improve the prognosis of the AMI.

Objective To investigate the relationship between ? catenin expression and colorectal adenoma with canceration. Methods The expression levels of ? catenin in 25 cases of normal colorectal mucosa (NCM), 42 cases of colorectal adenoma (CA), and 19 cases of colorectal adenoma with canceration (CAC) were detected by immunohistochemistry. Results ? catenin expression was detected on the cell membrane in normal colorectal mucosa. Reduced membrane expression, cytoplasmic and nuclear expression were detected in the colorectal adenoma and adenoma with canceration. 8.Washing Effect of Surgical Instruments after Three Different Methods to Keep Moist

OBJECTIVE To study the washing effect of different methods to keep the surgical instruments moist. METHODS The instruments which were selected from the routine surgical instruments within a week were washed after three different methods to keep moist. The washing effect was compared by eyes, without equipment and with 5 times magnifying glass. RESULTS The multienzymatic detergent was the best one to keep moist, the next was the water, and the chlorine releasing agent was the worst. CONCLUSIONS Hospitals should select the suitable method to keep moist by considering the actuality.
9.Laparoscopic procedures in the resection of colorectal cancers

Objective To investigate the use of laparoscopy in the resection of colorectal cancers. Methods A total of 17 cases of colorectal cancer resection had been made by laparoscopy, including 3 cases of right hemicolectomy, 3 cases of sigmoid colon cancer radical resection, 7 cases of Dixon’s procedure, 2 cases of Mile’s procedure and 2 cases of Park’s procedure. The operation was accomplished totally laparoscopically in 6 cases, while the laparoscopic-assisted and hand-assisted excision were conducted in 8 cases and 3cases, respectively. Results All the cases were cured to be discharged from the hospital. There were no deaths or complications or conversions to open surgery.The operation time was 150~240 min (mean, 190 min). The intraoperative blood loss was 20~300 ml (mean, 55 ml). The average length of postoperative hospital stay was 9.6 days. Analgesic was required in 6 cases. Follow-up for 1~6 months found no recurrence. Conclusions Laparoscopic resection of colorectal cancer is safe and feasible,with a satisfactory short-term effect.
