Objective To study the effect of Vincristine-loaded intact human erythrocytes on tumor cell in vitro. Methods VCR was loaded in intact human erythrocytes using a modified hypotonic preswelling technique. The proliferation of K562 cells after incubation with VCR-loaded erythrocytes was detected by MTT. Cellular cycle and apoptosis of K562 cells were analyzed through FACS staining with PI/Rhodamin123, and morphology of apoptotic K562 cells and their nucleus were observed through staining with Hoechest33258. Results VCR-loaded erythrocytes could efficiently inhibit the proliferation of co-cultured K562 cells and induce the cells′ apoptosis in a dosage-dependent manner. Cellular cycle analysis demonstrated that proportion of K562 cells in G 0/G 1 decreased whereas in S+G 2/M increased after incubation with VCR-loaded erythrocytes. Objective To explore the optimal method and condition for controlling release of vincristine (VCR) from VCR-loaded erythrocytes and enhancing the stability of carrier erythrocytes. Methods Erythrocytes were loaded into human erythrocytes using the modified hypotonic pre-swelling and isotonic resealing technique, and then with VCR, and they were treated with glutaraldehyde in concentration of 0.16% and 0.25% respectively. After storing at 4℃ for indicated time, the amount of VCR was determined with HPLC to compare their drug-release rate, and the degree of hemolysis in the supernatants was observed to compare the fragility of VCR-loaded erythrocytes treated with 0.16% glutaraldehyde with that with 0.25%, VCR-loaded erythrocytes treated with PBS, and VCR-loaded erythrocytes exposed to different hypotonic sodium chloride in different concentrations. Results The VCR-accumulated release rate increased in glutaraldehyde-treated VCR-loaded erythrocytes as well as untreated group during preservation. The VCR-accumulated release rate of 0.25% glutaraldehyde-treated group decreased by 71.67?4.20%, which was significantly slower than untreated VCR-loaded erythrocytes and 0.16% glutaraldehyde-treated group. After storing at 4℃ for 21 days, no significant changes at erythrocyte morphology was found in VCR-loaded erythrocytes treated with 0.25% glutaraldehyde, whereas untreated VCR-loaded erythrocytes and those treated with 0.16% glutaraldehyde can be stored only for 5d and 7.5d respectively. As compared to un-treated VCR-loaded erythrocytes, no significant change of osmotic fragility was seen in 0.25% glutaraldehyde-treated group, but significant increase in osmotic fragility was seen in 0.16% glutaraldehyde-treated group. Conclusion 0.25% glutaraldehyde could optimally block the membrane of VCR-loaded human erythrocytes and enhance their stability.

Objective To observe the change of the sera lipid profiles in patients with chronic hepatitis B Virus (HBV)infec-tion.Methods Sera from 254 patients suffered from liver disease were collected and divided into four groups according to the severity of the disease (71 of Cirrhosis;80,46,51 of Light,Moderate and Highly severity of the chronic HBV infection, respectively).Sera of 59 healthy patients were collected as control.The concentrations of their sera lipids (CHOL and TG), lipoprotein (HDL and LDL)and apolipoproteins (ApoA-I and ApoB)were determined and compared.Results Compared to the control group,the levels of the CHOL,TG,HDL,LDL,ApoA-I and ApoB were varied in degree in the chronic HBV in-fection group and the cirrhosis group.For the highly severity group and the cirrhosis group,statistics analysis showed de-creased and significantly different lipid and apolipoprotein results when compared to the control group.For the light and moderate severity group,when compared to the control group,levels of TG,HDL and ApoA-I were decreased with remarka-ble difference.The ApoB/ApoA-I ratio of the chronic HBV infection (Highly severity group)was 2.10±1.44,which was significantly higher than that of any other group (P<0.05).Conclusion The ApoB/ApoA-I ratio can be a potential marker for the evaluation of the severity of the chronic HBV infection.

Objective To investigate the safety of treatment of symptomatic middle cerebral artery stenosis with Gateway-Wingspan system by reviewing the data on complications.Methods Forty-seven patients with symptomatic middle cerebral artery stenosis treated with Wingspan system were respectively analyzed and emphasis was paid to cases with complications.Relevant data was collected such as characteristics of patients and lesions,selection of ballon and stent and radiographic information.Results Complications were found in 4 patients (8.5％),which were due to cerebral vasospasm,heperperfution syndrome,artery branch rupture and subacute stent thrombosis respectively.Two patients experienced symptomatic intracranial hemorrhage (4.3％)and one of them died (2.1％).The morbidity of severe complications was 6.4％.The success rate of stent implantation was 100％.Mean artery stenosis reduced from (83.5 ± 10.0)％ to (19.8 ± 9.2)％ after Wingspan stent implantation.Conclusion Middle cerebral artery stenting angioplsty greatly improved the stenosis with high technical success rate.However,the rates of cerebral vascular complications and symptomatic intracranial hemorrhage were relatively high in this study,which needs to be widely verified in practice.

Objective To explore the effects of videofluoroscopic swallowing study (VFSS) in the assessment of dysphagia in post-stroke patients. To study the representation and rehabilitative outcome of dysphagia following stroke. Methods Seventy post-stroke patients and 80 normal adults were assigned to study group and control group and underwent VFSS. Abnormal signs on VFSS were compared between 2 groups. Aspiration patients received swallowing therapy for 4 weeks followed by VFSS again. Abnormal signs on VFSS were compared with those of pre-treatment. Results Penetration/aspiration, oral retention and pharyngeal retention occurred respectively at a frequency of 5.0%, 13.4% and 25.3% in control group in mild status, 45.0%,46.5%, 48.9% in study group in moderate to severer status. 10 times of silent aspiration were seen, accounting for 24.4% of all aspiration. Objective To study the effect of lipopolysaccharide ( LPS ) on the activity of primary cultured macrophages and the distribution of divalent metal transporter 1 ( DMT1 ) and ferroportin 1 ( FPN1 ) .Methods Primary cell culture , MTT chromotest , cytochemistry chromotest and cell immunofluorescence techniques were used in this work . Results DMT1 was mainly distributed in the cytoplasm , which illuminates that DMT1 mediates the macrophage intracellular transit of iron from phagolysosome to cytoplasm .FPN1 was distributed in the cytoplasm and membrane , and the cytoplasm was the main site of FPN 1 distribution in macrophages .Conclusion Iron liberation from heme inside the phagolysosome occurs after erythrophagocytosis and it is possible that FPN 1 mediates intracellular transit of iron released by heme catabolism .The study found that LPS promoted the cell growth and this effect was reached to the peak in the 10 -5μg/L LPS group, but the cell growth was blocked with the increase of LPS concentration .

Objective To evaluate the relation of appearance of contrast-enhanced ultrasonography (CEUS) to Ki 67 and C-erbB-2 in breast invasive ductal carcinoma and ductal carcinoma in situ.Methods The appearance of CEUS in 29 patients with breast invasive ductal carcinoma and 12 patients with ductal carcinoma in situ,which was diagnosed by surgery or biopsy,was analysed retrospectively.The Ki-67 and CerbB-2 were measured by immunohistochemical test.The relation of the above two factors to appearance of contrast ultrasonography was analysed.Results The positive rate of Ki-67 was 68.97％ (20/29) in patients with breast invasive ductal carcinoma and 16.67％ (2/12) in patients with ductal carcinoma in situ.The positive rate of C-erbB-2 was 48.2％ (14/29) in patients with breast invasive ductal carcinoma and 42.6％ (5/12) in patients with ductal carcinoma in situ.The masses had a common appearance of high enhance in patients with breast invasive ductal carcinoma and ductal carcinoma in situ,which was not correlated to C-erbB-2.The Ki-67 was significantly higher in patients with ductal carcinoma in situ than that in patients with breast invasive ductal carcinoma,the appearance of CEUS was correlated to Ki 67.Conclusions The features of micro-vessels by CEUS were correlated to Ki-67 in patients with breast invasive ductal carcinoma and ductal carcinoma in situ.

Objective To evaluate the clinical efficacy ofZi Wu Liu Zhu Na Jia needling (acupuncture based on midnight-noon ebb-flow theory) in treating spastic hemiplegia after cerebral stroke by using surface electromyography (sEMG).Method Fifty-two patients with spastic hemiplegia due to cerebral stroke were randomized into a treatment group and a control group, 26 cases each. The two groups were both intervened by conventional rehabilitation training. In addition, the treatment group was givenZi Wu Liu Zhu Na Jia needling and the control group was given ordinary acupuncture. The two groups were treated once a day, 15 sessions as a course. The ambulation ability, muscle tension, neurological deficit score (NDS), Fugl-Meyer Assessment Scale (FMA), and Berg Balance Scale (BBS) were evaluated before the intervention and after 2 treatment courses, and the sEMG signals were also collected and analyzed.Result The Holden's Functional Ambulation Classification (FAC) and Modified Ashworth Scale scores were significantly changed in the two groups after the intervention (P<0.01). After the intervention, the FAC and MAS scores in the treatment group were significantly different from those in the control group (P<0.05). The NDS, FMA and BBS scores were significantly changed in the two groups after the treatment (P<0.05). The NDS, FMA and BBS scores in the treatment group were significantly different from those in the control group after the treatment (P<0.05). Of the sEMG signals, H/M max in the treatment group was significantly different from that in the control group after the intervention (P<0.01).Zi Wu Liu Zhu Na Jia needling with rehabilitation training can significantly mitigate the muscle tension and promote the activities of daily living in patients with spastic hemiplegia after cerebral stroke.

Objective To evaluate the curative effect,safety,and medium-term effect of intracranial artery stenosis stent (ICASS) for the treatment of the symptomatic intracranial artery stenosis.Methods ICASS was performed in 21 cases with the symptomatic intracranial artery stenosis.The stenting success rate,the improvement rate of stenosis post-stenting and the incidence of complications were observed.The change of the scores of the Modified Rankin Scale (MRS)and National Institutes of Health Stroke Scale (NIHSS)between pre-and post-stenting (1 month,3 months,6 months,12 months and 2 years)and the occurrence of ischemic cerebrovascular events after stenting were compared and analyzed respectively.Results Except one case failed because of intracranial hemorrhage during the operation,other 20 patients were treated with endovascular stent plasty successfully,the total technical achievement ratio was 95.2%.The aver-age stenosis ratio decreased from (73.4±7.9)% to (13.7±9.2)%.The complications incidence was 4.8% within the peroperative. 9 patients were reviewed with DSA in 6 months,the average ratio of stenosis was (29.5 ±12.2)%,no patient was found with the restenosis.Compared with pre-stenting,the score of MRS and NIHSS at the follow-up duration of 3 months,6 months,12 months and 2 years post-stenting of patients were significant improved (all P < 0.05 ).During the follow-up period,2 patients were found with new transient ischemia attack (TIA),1 patient had non-targeted vascular CI without other target vascular therapy-related com-plications.Univariate analysis indicated that patients with elderly (≥ 75 years old),or hypertension,or diabetes,or dyslipidemia,or in the set of events to endovascular treatment for longer than 1 month,or with more than two independent risk factors for patients with underlying diseases treatment,or without systemic therapy,whom would be much more easily to reoccur the cerebrovascular events in post-operation(all P <0.05 ).Conclusion ICASS should be an effective and safe therapeutic method to the symptomatic intracra-nial arterial stenosis.The medium-term efficacy is significant.Serious complications might still be emerge with endovascular stent plasty,so close attention must be paid with.
