1.Expression of recombinant human interleukin 8 in E.coll and its functional identification

By using genetic engineering techniques, we had successed in the expression of recombinant human interleukin 8 (rhIL-8)in E. col(?) IL-8 cDNA insert into the plasmid pGEM-IL-8 was subcloned into expression vector pMS316, and IL-8 fusion protein was produced in E.coli with high efficiency, The purified recombinant protein exhibited chemotactic activity for mouse neutrophils both in vive and in vitro. It showed that a fragment of the human IL-8 had no influence on the chemotactic activity of IL-8.
2.Effect of interleukin-8 on rat intestinal ischemia-reperfusion injury

cDNA of human interleukin-8 was expressed in E. coli bacteria, and recombi-nant human interleukin-8 ( rhIL- 8 ) was produced. The latter was used for treatment of rat intestinal ischemia-reperfusion(I-R) injury, and it was found that rhIL-8 significantly improved the hypotensive state of the animals,and it also inhibited the elevation of plasma endothelin level , decreased the intestinal myeloperoxidase activity, and obviously prolonged the survival timeof the animals. Perfusion of isolated rat superior mesenteric artery with rhIL-8 markedly inhibited endothelin synthesis or (and) release induced by anoxia. The above results suggest that rhIL- 8 may be an effective preventive and therapeutic agent for I-R injury through its inhibition on leukocyte adhesion and endothelin synthesis or (and) release.

4.Piperacillin/tazobactam Treatment for Severe Community-acquired Pneumonia among Elderly

OBJECTIVE To evaluate the clinical efficacy of piperacillin/tazobactam for severe community acquired pneumonia(CAP) among elderly.METHODS Seventy-six elderly cases of severe CAP were randomized into piperacillin/tazobactam group and ceftriazone group,and the sputum culture was underwent for each case before and after treatment.RESULTS The total efficacy rates and bacterial clearance rates in piperacillin/tazobactam group and ceftriazone group were 92.11% and 88.89%,and 73.68% and 58.82%,respectively,and the differences between the two groups were statistically significant.CONCLUSIONS It suggestes that piperacillin/tazobactam be a more effective drug for severe CAP among elderly in old age.
5.Decitabine for treatment of myelodysplastic syndrome in an elderly patient and review of literature.

Findings in epigenetic changes in meylodysplastic syndromes (MDS) and the development of demethylating drugs provide a new approach to the treatment of MDS. We used standard-dose decitabine for treatment of MDS in an elderly patient with an International Prostate Symptom Score (IPSS) of moderate risk group 2, and achieved a complete response in the first course. We report our experience with this case and review the relevant literatures.

6.The microsurgical anatomic research of the internal auditory canal area on the retrosigmold approach.

OBJECTIVE:
To evaluate the safety of the circular round window and discus anatomic landmarks of posterior wall of internal auditory canal by investigating the microscopic anatomy of internal auditory canal area of the retrosigmold approach, which can provide the anatomical basis for acoustic neutrinomas surgery.
METHOD:
Fifteen adult cadaver heads (30 sides) fixed with formalin were used in the study. The retrosigmold approach operations were imitated to dissect the blood vessels and nerves in internal auditory canal area by opening round bony window and removing posterior wall of internal auditory canal.
RESULT:
Fifteen specimens of 30 sides circular bone window were opened without injury with transverse sinus and sigmoid sinus. The vertical distance between the highest point of bone window margo superior and the lowest point of transverse sinus margo inferior was (4.02 ± 0.32) mm. The vertical distance from the most anterior point of bone window leading edge to the most posterior point of sigmoid sinus trailing edge was (6.31 ± 0.43) mm. The internal auditory canal tubercle located in the anterior superior position of internal auditory canal. The vertical distance from the highest point of internal auditory canal tubercle to the upper margin of internal auditory canal was (2.31 ± 0.32) mm. To expose the whole internal auditory canal, the length and width of the internal auditory canal posterior wall removal was (7.29 ± 0.32) mm, (4.12 ± 0.29) mm. Within this removal range, no case of cochlea, semicircular canal or venous was injured in 30 specimens.
CONCLUSION
The method of opening round window through retrosigmold approach is simple, practial and convenient. With little variation and easiness of location, the sinternal auditory canal tubercle can be used in the identification of the internal auditory canal. When exposing the whole internal auditory canal, the removal scope of the posterior wall should be paid more attention to, in order to avoid the damage of cochlea, semicircular canal and jugular bulb.

7.Survey on Knowledge of Stroke Rehabilitation among Traditional Chinese Medicine Physicians in 14 Tertiary A- level Hospitals

Objective To investigate the current situation of knowledge of stroke rehabilitation among Traditional Chinese Medicine (TCM) physicians in 14 tertiary A-level hospitals. Methods A cross section survey was conducted in 14 tertiary A-level hospital in China in May, 2012. A total of 128 TCM physicians completed the questionnaire, which was created according to Medical Care Guideline of Stroke Rehabilitation in China. Results Overall accuracy of stroke rehabilitation basic knowledge was 66.83%, 88.77% of stroke rehabilitation evaluation, 75.12% of stroke rehabilitation treatment. Conclusion The knowledge of stroke rehabilitation is insufficient among TCM physicians in tertiary A-level hospitals, and further training is needed.
8.Suitability assessment of domestic Kegel exercise online health education written materials

Objective To adopt Suitability Assessment of Materials for suitability assessment of domestic Kegel exercise online health education written materials. Methods After Searching Kegel exercise online health education written materials in search engines, four health education professionals assessed the suitability of the collecting materials.One professional evaluated all collecting materials,and all materials were divided into three parts. The other three professionals evaluated one part agian independently.Excel and SPSS 19.0 were used to analyze the results. Results 38 Kegel exercise online health education written materials were collected.2(5.3%)of the materials were assessed as"Superior",23 (60.5%) of the materials were "Adequate", and 13 (34.2%) of the materials were evaluated as "Not suitable". Average percentage score was 44.05% (24% ~74% ), and the overall assessment was"Adequate". Conclusion Most domestic Kegel exercise online health education written materials′quality should be certified and suitability were adequate,which need some improvement.

Objective To investigate the relationship between maternal body composition in first trimester and gestational diabetes mellitus (GDM). Methods In this nested case-control study based on a prospective cohort study, we enrolled gravidas between 8 and 14 weeks of gestation, who received prenatal care and voluntary nutrition evaluation in Gansu Provincial Maternity and Children Health Care Hospital, from July 2016 to January 2017. Body mass index (BMI) of each gravida was recorded and the maternal body composition including body fat, body fat percentage and fat-free mass was measured by bioelectrical impedance analysis. Pregnancy outcomes were followed up. A total of 70 patients diagnosed with GDM were allocated to the GDM group and 140 healthy gravidas matching for age and pre-pregnancy BMI were selected as the control group. Differences in body composition between two groups and their relationships with GDM were analyzed by Chi-square test and multivariate logistic regression. ResuLts Maternal BMI ≥ 30 kg/m2 (OR=1.973, 95%CI:1.095-7.664, P=0.024) and body fat percentage≥30%,≥35% and≥40% in first trimester (OR=1.261, 95%CI:1.021-2.982, P=0.010; OR=4.020, 95%CI: 1.341-7.950, P<0.001; OR=8.311, 95%CI: 5.018-42.771, P<0.001) were the risk factors of GDM. ConcLusions BMI ≥ 30 kg/m2 and body fat percentage ≥ 30% in first trimester are risk factors for GDM and excessive adipose tissue may play an important role in the development of GDM.
10.Preparation,characterization,and application of acylated collagen with anhydride

BACKGROUND:Collagen is a biomedical material with good biocompatibility,low toxicity,low immunoactivity,and high cellular affinity.However,the defects such as hydrophilicity and poor thermal stability are the key technical problems that need to be solved urgently in biomedical,tissue engineering,and other applications. OBJECTIVE:To elaborate on the preparation method,characterization,and application progress of acylated collagen. METHODS:PubMed,X-mol,and CNKI databases were used to search the literature on acylated collagen preparation methods,characterization,and application.The search time was from January 1992 to May 2023.The English search terms were"acylated collagen,modified collagen,water-soluble collagen,acetic anhydride".Chinese search terms were"acylated collagen,modified collagen,water-soluble collagen".After an initial screening of all articles according to inclusion and exclusion criteria,53 articles with high relevance were retained for review. RESULTS AND CONCLUSION:(1)The preparation of acylated collagen is mainly obtained by the acylation reaction of acid anhydride,and the reaction sites are mainly ε-amino and N-terminus-amino of collagen,and the reaction environment of acylation reaction needs to be carried out in an alkaline environment.(2)Succinic anhydride is the preferred reactant of acylated collagen.Carbonic anhydride,citric anhydride,and bifunctional modifiers with acylation ability have been excavated in recent years to meet various requirements,but it is still limited to laboratory preparation;complex preparation process is difficult to industrialize,and follow-up research is needed to continuously improve.(3)The characterization methods of acylated collagen are similar to those of collagen,but there is still a lack of standards for the water-soluble characterization of acylated collagen.(4)In recent years,acylated collagen has been gradually used in the preparation of lenses,hydrogels,and dressings,as well as cosmetic raw materials.However,there is still little research on the application of acylated collagen,and there is a lack of complete in vivo experiments to verify the practicality of acylated collagen.
