1.Serum Pharmacological Study of Baihe Zhimu Decoction in Protecting Rat Ovarian Granular Cells

Objective To observe the effect of serum containing Baihe Zhimu Decoction(BZD) on ovarian granular cells(GC) injury induced by cadmium chloride(CdCl2) in rats.Methods Totally fifteen female SD rats aged 23 days,3 months and 12 months(5 rats in each age group) were enrolled into the study.The number of ovary GC was counted.The growth of GC within 10 days was observed continuously for the selection of the well-growth ovarian GC.On the third day of the culturing,the screened ovary GC was cultured for another 48 hours in 10,20,40,60 and 80?mol/L CdCl2 solution respectively at 37℃ with 5% CO2 added.The optical density(OD) at 490nm was determined by methyl thiazolyl tetrazolium(MTT) assay for the screen of the optimal concentration of CdCl2 solution.Then the ovary GC were cultured in 96-well plate for 72 hours with serum containing BZD(at the final concentration of 1.25%,2.5%,5%,10% and 20% respectively) added.Meanwhile,the blank control group(cultured with 10% DMEM culture medium),model group(cultured with 10% DMEM culture medium and optimal concentration of CdCl2 solution),and blank serum group(cultured with blank serum and optimal concentration of CdCl2 solution)were set up.After the culturing for 48 hours,the OD value of all the groups was detected to evaluate the effect of BZD-containing serum on CdCl2-induced ovarian GC injury in rats.Results The number of GC in 23-day rats was the biggest and their growth was the best in the manner of exponential growth,indicating that GC in 23-day rats was suitable for the modeling.Ovarian injury was increased with the concentration of CdCl2,and 40?mol/L was the optimal concentration for CdCl2,which induced the shrinkage of GC edge.Serum containing BZD promoted the growth of GC,and counteracted the CdCl2-induced GC injury.Conclusion The protective mechanism of BZD-containing serum on CdCl2-induced GC injury is probably related with the counteraction of CdCl2-induced injury and with the increase of ovarian GC activity and quantity.
2.Effect of Baihe Zhimu Decoction on serum sex hormone and uterus index of ovariectomized rats with kidney-yin deficiency

Objective: To observe the effect of Baihe Zhimu Decoction on serum sex hormone and uterus index of ovariectomized rats with kidney-yin deficiency. Methods: 3-month years old femal SD rats which were ovariectomized were divided randomLy into 6 groups: ovariectomized, Baihe Zhimu DecoctionⅠ, Baihe Zhimu DecoctionⅡ, Baihe Zhimu Decoction Ⅲ, Baihe Zhimu Decoction Ⅳ, then the normal group was set. The rats were induced to kidney-yin deficiency by using etraiodothyronine, then ig Baihe Zhimu Decoction for 4 weeks. The levels of sex hormone E2, FSH, LH in serum were detected and the indexes of uterus were caculated to observe the effect of Baihe Zhimu Decoction on sexual hormone and sexual organ in ovariectomized rats with kidney-yin deficiency. 3.Preparation of Curcumin-Lysine Cocrystal and Solubility Comparison of Different Crystal Forms

Objective:To develop a new cocrystal form to improve bioavailability and druggability of curcumin. Methods:Solvent crystallization was used to prepare curcumin-lysine cocrystal, and two crystal forms ( a and b) were obtained. The two cocrystal forms were characterized by elemental analysis, differential scanning calorimetry and X-ray diffraction. The dissolution profiles of the two crystal forms were detected and compared with that of curcumin. Results:Curcumin combined with lysine at a stoichiometry of 1: 1 to form the cocrystal with much better solubility than curcumin. Conclusion: Curcumin-lysine cocrystal has ideal druggability, which means the cocrystal can be a new chemical entity used for subsequent development.
4.Determination of the Plasma Protein Binding Rate of Plasma Brucine in Rats by HPLC

Objective To study the protein binding rate of brucine in plasma of rats.Methods The plasma balance dialysis was used.HPLC was performed for the quantitative determination of brucine,and then the protein binding rate of brucine in rats plasma was calculated.Results The protein binding rate was 59.679 %,59.935 %,and 56.387 %when the concentration of plasma brucine was 2.5,1,and 0.4 ?g?mL-1 respectivley.Conclusion The HPLC method used to determine brucine is simple,rapid,and sensitive,and with good specificity,precision and accuracy.Brucine has medium capacity in binding to plasma protein.
5.Tissue distribution of strychnos nux-vomica alkaloids in rats

AIM To study the tissue distribution of strychnos nux vomica alkaloids in rats. METHODS Rats were given strychnos nux vomica alkaloids intravenously and strychnine(S), brucine(B), strychnine N oxide(SNO), brucine N oxide(BNO) in every tissue was determined by HPLC timely. RESULTS The concentration of S, B, SNO and BNO were high in brain and spinal marrow. CONCLUSION S, B, SNO and BNO all have the ability to permeate blood brain barrier.
6.Study on Extraction Process of Polysaccharide in Ophiopogon japonicus

Objective To optimize the extraction process and to establish a method for the content determination of the polysaccharides in Ophiopogonjaponicus.Methods With extracts rate of polysaccharide as the index,an orthogonal de- sign test was adopted to optimize the extraction process.And phenol-sulphuric acid colorimetric method was used to de- termine the content of polysaccharides in Ophiopogonjaponicus.Results The optimum extract conditions were as follows: refluxing twice with eight times amount of water,thirty minutes each time,and precipitation with 80 % alcohol.A good linearity of polysaccharides was in range of 0.02032~0.10008 mg(r=0.9998),and the average recovery rate was 102.41%,RSD was 1.78 %.Conclusion The polysaccharide can be fully extracted under these conditions.
7.Factors related to the failure of weaning off ventilator

Objective To analyze the failure rate and failure factors related to ventilator weaning off in children. Methods Clinical data of 214 patients who received mechanical ventilation in pediatric intensive care unit of Tianjin Children's Hospital from Jan 2005 to Dec 2008 were analyzed retrospectively. Results There were 141 planned extubation events,122 of which were successful and 19 of which were unsuccessful.The failure rate of planned extubation was 13.5%. The failure rate of infants less than 6 months was higher than that of infants more than 6 months, but there was no significant difference ( 15.0% vs 9. 8%, P >0.05). The success rate of extubation in pneumonia cases (90. 7% )was higher than that in others (P <0. 05 ). The duration of mechanical ventilation had no effect on success rate of extubation ( (7. 64 ± 5.68 ) d vs (6. 95 ± 3. 14) d, P > 0. 05 ). The duration of corticosteroid treatment after extubation in the successful group was less than that in the failure group ( ( 12. 35 ±9. 69) h vs ( 18.63 ± 12. 17) h,P <0. 05). According to the result of multiple linear regression analysis ( R2 = 0. 093), airway obstruction was the high risk factor of the extubation failure( F = 14. 256, P < 0. 001 ). Conclusion Currently,weaning from mechanical ventilation in PICU depends on the combination of pediatricians' experience and objective indicators. The key point for improving the success rate of exubation is that we should explore reasonable and feasible extubation protocol and discover and exclude the related factors of exubation failure as earlier as possible.
8.Effect of Muscle-region Alignment Needling plus Dermal Needle Therapy on the Long-term Therapeutic Efficacy for Post-stroke Upper-limb Spasticity

ObjectiveTo evaluate the long-term therapeutic efficacy of muscle-region alignment needling plus dermalneedle therapy in treating post-stroke upper-limb spasticity.MethodTotally 488 patients with post-stroke upper-limb spasticity were randomized into a treatment group and a control group, 244 in each group. Besides theessentialrehabilitation treatment, the treatment group was intervened by muscle-region alignment needling plus dermal needle therapy, while the control group was given regular Western medication.Thetwo groups were intervened for 3 weeks and were followed up for 6 months. The neurologic function, Functional Comprehensive Assessment (FCA), and Stroke Speciality-Quality of Life (SS-QOL) were observed for the follow-up study.ResultThe total effective rate was 93.4% in the treatment group versus 61.5% in the control group, and the difference was statistically significant (P<0.05); the neurologic function, FCA score, and SS-QOL score in the treatment group were significantly superior to that in the control group at the end of the follow-up study (P<0.05).ConclusionMuscle-region alignment needling plus dermal needle therapycan produce a content long-term therapeutic efficacy in treating post-stroke upper-limb spasticity.
9.Effect of Cognitive Therapy on the Insomniac

ObjectiveTo explore the cognitive psychological characteristics of the insomniac and the effect of cognitive therapy.Methods93 insomniac in-patients were divided randomly into the trial group (n=47) and control group (n=46). The trial group was treated by cognitive therapy plus medicine. The control group was only treated with medicines. The treatment lasted for 8 weeks. Before and after treatment, patients of all two groups were examined by Beliefs and Attitudes about Sleep Scale (DBAS). 45 healthy individuals were also examined at the same time. The therapeutic effect was evaluated with Pittsburgh Sleep Quality Index (PSQI).ResultsBefore treatment, DBAS scores of insomniacs were significantly different from the healthy (P<0.001). After treatment, scores of DBAS and PSQI of the trial group were significantly different from that of the control group ( P<0.001).ConclusionThe cognitive treatment can change erroneous cognitions related to sleeping in the insomniac and improve the therapeutic effect.

Beijing has been one of the epicenters attacked most severely by the SARS-CoV (severe acute respiratory syndrome-associated coronavirus) since the first patient was diagnosed in one of the city's hospitals. We now report complete genome sequences of the BJ Group, including four isolates (Isolates BJ01, BJ02, BJ03, and BJ04) of the SARS-CoV. It is remarkable that all members of the BJ Group share a common haplotype, consisting of seven loci that differentiate the group from other isolates published to date. Among 42 substitutions uniquely identified from the BJ group, 32 are non-synonymous changes at the amino acid level. Rooted phylogenetic trees, proposed on the basis of haplotypes and other sequence variations of SARS-CoV isolates from Canada, USA, Singapore, and China, gave rise to different paradigms but positioned the BJ Group, together with the newly discovered GD01 (GD-Ins29) in the same clade, followed by the H-U Group (from Hong Kong to USA) and the H-T Group (from Hong Kong to Toronto), leaving the SP Group (Singapore) more distant. This result appears to suggest a possible transmission path from Guangdong to Beijing/Hong Kong, then to other countries and regions.
