microRNAs(miRNAs)are a class of 18～26 nt small non-coding single strand RNA molecules,which negatively regulate the expression of a variety of genes at post-transcriptional level by binding to complementary sites on target mRNA.Mutation,deletion or high expression of miRNA was found to correlate with various human cancers.miRNAs involved in tumor cell proliferation,differentiation and apoptosis processes as function as oncogenes or tumor supressors It is predictable that microRNA might be applied in the diagnosis and treatment of malignant tumors.

Background Aberrant proliferation of residual lens epithelial cells (LECs) is one of main causes of posterior capsular opacification (PCO).Researches indicated that Wnt3a signaling pathway promote proliferation of epithelial cells,but its effect on LECs is still unclear. Objective The present study was to investigate the effects of Wnt3a on proliferation of human LECs and its mechanism and to provide a new gene target in the prevention and treatment of PCO. Methods Human LECs line (SRA01/04 cells ) was cultured and then incubated to 6-well plate at the density of 4×105/well.A human Wnt3a cDNA expressing vector targeted human LECs was constructed to increase the Wnt3a expression in SRA01/04 cells,and pcDNA3-HA expression vector was used as the control group.The expression of Wnt3a was identified by Western blot assay after transfected.The growth and proliferation of SRA01/04 cells were detected by MTT and flow cytometry (FCM).The expressions of β-catenin,cyclin D1 and c-myc in the cells were detected by Western blot assay.β-Catenin expression was localized using immunofluorescence assay,and the expression and localization of proliferating cell nuclear antigen ( PCNA ) were analyzed by immunocytochemistry for the exploration of the active mechanism of Wnt3a to proliferation of LECs.Results Human Wnt3a cDNA expression vector was designed successfully and transiently transfected to SRA01/04 cells,and Wnt3a/SRA01/04 cells and pcDNA3-HA/SRA01/04 cells were obtained.The expression of Wnt3a was verified in the Wnt3a transfected group compared with the control group.MTT indicated that the cell proliferating rate was significantly different between the Wnt3a transfected group and the control group ( Fgroup =15.235,P =0.005 ;Ftime =369.677,P =0.000),and that in various time points after transfected was significantly different (t =20.843,P=0.001 ;t =26.214,P＜0.001 ;t=25.177,P=0.001 ;t =35.516,P＜0.001 ;t =615.056,P＜0.001 ).The proportion of SRA01/04 cells in G1 phase was 51.74％ in the Wnt3a cDNA transfected group,with a significantly decrease in comparison with 79.44％ of the control group.However,the proportion of SRA01/04 cells in S phase in the Wnt3a cDNA transfected group was higher than that of the control group (36.23％ versus 12.34％ ).The positive expression rate of PCNA protein in SRA01/04 was (47.00％ ±7.58％ ) in the Wnt3a cDNA transfected group and ( 16.00％ ±3.61％ ) in the control group with a significant difference between them (t =8.256,P＜0.01 ).After 48 hours of transfection of the Wnt3a cDNA,the expression amount of β-catenin proteins was higher and the immunofluorescence was stronger in cell nucleus,and the expressions of cyclin D1 and c-myc proteins were elevated in Wnt3a/SRA01/04 cells. Conclusions The overexpression of Wnt3a activates the Wnt/β-catenin signaling pathway and downregulates the expression of a subset of target genes,including cyclin D1 and c-myc,which plays an important role in promoting the proliferation of human LECs.

Objective To investigate the role of the chemokine receptor CXCR3 and its ligands in the migration of lymphocytes and acute hepatic failure. Methods BALB/cJ mice (6-8 weeks, female) were intraperitoneally injected with 100 PFU mouse hepatitis virus-3(MHV-3). The proportions and numbers of T cells and NK cells in liver, spleen, and blood as well as the expression of CXCR3 in T cells, and NK cells post MHV-3 infection was analyzed by flow cytometry. The hepatic mRNA level of the CXCR3-associated chemokines(CXCL9 and CXCL10) was detected by real-time PCR. A transwell migration assay was used to assess the chemotactic effect of MHV-3-infected hepatocytes and CXCL10 on the splenic lymphocytes. Results Following MHV-3 infection, the number of hepatic NK cells and T cells and the frequencies of hepatic NK cells and T cells expressing CXCR3 increased markedly; however, in the spleen and peripheral blood, they both decreased significantly. Moreover, the hepatic mRNAs levels of CXCL9 and CXCL10 were significantly elevated post infection. The transwell migration assay demonstrated that MHV-3-infected hepatocytes have the capacity to attract and recruit the splenic NK cells and T cells, and CXCL10 plays a key role in lymphocyte mobilization from the spleen. Conclusion Interactions between CXCR3 and its ligands (CXCL9 and CXCL10),especially CXCL10 may play a key role in the recruitment of intrahepatic lymphocytes and subsequent necroinflammation and acute hepatic failure in MHV-3 infection.

Objective:To observe the clinical effects of integrated disease and TCM syndrome on patients with second degree atrioventricular block.Methods:Because of the lentor cardiac rhythm characteristics of econd degree atrioventricular block,the herbs which had the effect in increasing cardiac rhythm were selected and were used in combination of clinical syndrome differentiation to treat 35 cases of second degree atrioventricular block.Results:The total effective rate was 88.6%.Conclusion:Integrated differentiation of disease and TCM syndrome had significant effects on second degree atrioventricular block.

Objective To investigate the effect of propofol anesthesia on hemodynamic changes and dose requirement of sodium nitroprusside (SNP) during SNP-induced hypotension.Methods Sixteen patients undergoing elective microneurosurgery were randomly assigned to one of two groups: propofol group and isoflurane group. Objective To observe the clinical effect of acupuncture combined with traditional Chinese medicine Tongluo huayu decoction on white matter damage in patients with occlusive cerebral infarction.MethodsA total of 76 patients with cerebral infarction with cerebral hemorrhage were enrolled in this study from April 2013 to January 2015.The random number table was divided into observation group and control group (38 cases).The control group was given routine western medicine treatment, the observation group on the basis of the control group plus traditional Chinese medicine tongluo huayu decoction, and with acupuncture treatment, the two groups were treated for 2 months, compare the effectiveness of treatment, analysis of the two groups before and after treatment platelet aggregation rate MAR (CDCA) and platelet-activated granule membrane glycoprotein 62P (CD62P), and the prognosis was analyzed by NIHSS, Montreal Metabolic Assessment Scale (MoCA) and Barthel Index (BI) Observe adverse reactions.ResultsThe effective rate of the observation group was 94.74% higher than that of the control group (78.95%, P< 0.05).After treatment, the expression of CD62P (21.87±1.55)%, MAR (35.23±1.62)% and NIHSS score (10.27±0.78) (P< 0.05).The incidence of adverse events was 13.16% in the observation group (25.04±1.11) and the BI index (88.15±1.20) were significantly higher than those in the control group (P< 0.05);There was no significant difference in the prevalence of adverse effects between 13.16% and 7.89%.ConclusionAcupuncture combined with traditional Chinese medicine Tongluo huayu decoction can effectively protect the white matter of patients with massive arterial occlusive cerebral infarction, improve its platelet CD62P, MAR and neurological function, cognitive function, improve the quality of life, and mild adverse reactions, it is worth in the clinical promotion application.
