Objective To observe the antimutagenic action of sodium selenite and vitamin E on condensates of cooking oil fumes(COF) Methods SCE (sister chromatid exchanges) were observed and contrasted before and after addition of sodium selenite or vitamin E at a certain dose to the culture of human peripheral blood lymphocytes treated by COF Results Sodium selenite added to lymphocytic culture treated by COF at 37 ℃ for 4 hour incubation significantly lowered the frequencies SCEs of lymphocytes and very significant synergistic effects on lowering frequencies of SCEs were observed during adding both sodium selenite(terminal concentration 0 9?10 -7 mol/L) and vitamin E(torminal concentration 0 1% (v/v) to lymphocytic culture treated ty COF for 4 hour incubation Conclusion Combined sodium selenite and vitamin E at certain concentrations in cultured lymphocytes for 4 hour incubation showed very significant synergistic effects on lowering the mutagenicity of lymphocytes induced ty COF.Sodium selenite alone was also shown to be effective in lowering the frequencies of SCEs of human peripheral blood lymphocytes
2.Application of invasive intracranial pressure monitoring in treatment of traumatic brain injury

Objective To discuss the effect of invasive intracranial pressure (ICP) monitoring in treating traumatic brain injury (TBI).Methods A total of 116 patients with TBI (including 112 with severe TBI,3 with moderate TBI and 1 with slight TBI) were monitored by using invasive ICP monitoring device in Tiantan Hospital from July 2003 to December 2007.All patients underwent ICP monitoring within first 24 hours after admission and treated with corresponding therapy including drug therapy and surgical treatment.Results Of all,74 patients survived but 42 with severe TBI died.Of 75 patients with GCS 3-5,33 died,with morbidity rate of 44%.Of 37 patieats with GCS 6-8,nine died,with morbidity rate of 24%.Four patients with GCS 9-15 survived.Conclusions Continuous ICP monitoring can help timely understanding of ICP changes for early diagnosis and correct treatment of TBI and is useful for judgment of prognosis.Low GCS and high ICP are predictors for bad outcome.
3.Investigation on status quo of metabolic syndrome among populations in west Jiangxi area

Objective To understand the incidence of metabolic syndrome (MS) among populations in the west Jiangxi area and its distribution characteristics.Methods A total of 6 714 people in west Jiangxi area were performed the cross-sectional survey for analyzing the prevalence situation of MS among different age groups and its components related factors.Results The MS incidence rate in west Jiangxi area was 14.6% (male 10.4%,female 19.8%),which in young,middle-aged and elderly groups were 9.4%,14.5% and 9.4% respectively,in which the prevalence rate of various age groups in the youth group were 2.3% fro the group A,7.5% for the group B and 13.8% for the group C respectively.The statistic analysis showed that MS was related with the related risk factors such as age,sex,blood pressure,body mass index,blood sugar and blood lipids.Conclusion The incidence rate of MS in the west Jiangxi area is positively correlated with the age,the risk factors are dominated by obesity,and the MS onset shows the rejuvenation trend.
4.Reflections on the Construction of Laboratory of Medical Test

To accomplish laboratory construction for qualify education,experiment items were changed,laboratories were opened to students after school,administraion for experiment teaching was accomplished.Apart from these,quality of laboratory staff were improved significantly.All the mentioned above are constructive in experiment teaching for students' quality.
5. Effects of dexmedetomidine in protective effects of propofol on newborn rats hippocampal synapses and the influence of the Bcl-2 expression

Objective: To evaluate the role of dexmedetomidine in protective effects of propofol on newborn rats hippocampal synapses and the influence of the Bcl-2 expression. Methods: Neurons were separated and purificated and identificated by microtubule associated protein-2 (MAP-2) antibody. Rats were divided into normal control group, propofol group(add 12 tng/L propofol), and propofol + dexmedetomidine group (add 12 mg/L propofol and 0.25 mg/L dexmedetomidine). The expression of Bcl-2 test by Real-time PCR and Western blotting; cell synapse was detected by transmission electron microscope. Results: Rat hippocampal neurons were separated successfuly. Hippocampal nerve cells in control group were given priority to with multiple processes of pyramidal cell, the cell body was triangular or elliptic, large cell body bright and pervious to light quality, had a halo around, abundant cytoplasm, nuclear. Compared with the control group, in propofol group the Bcl-2 expression decreased obviously. Compared with the propofol group, the expression of Bcl-2 increased obviously (P<0.05). Conclusion: Dexmedetomidine can influence the expression of Bcl-2 so as to play a protective role of hippocampal neurons.
6.PI3K/Akt signal transduction pathway and apoptosis after cerebral ischemia

Apoptosis is one of the important forms during cerebral ischemia.Phosphoinositide-3 kinase (PI3K)/serine/threonine kinase (Akt) is the important cell survival signaling pathway,while c-jun N-terminal kinase (JNK) is the important pro-apoptotic signaling pathway.The dynamic equilibrium of the two signal transduction pathways maintains cell survival and apoptosis under the physiological state.Stimulation during cerebral ischemia breaks this physiological balance and results in the apoptosis of massive neurons.A variety of proved neuroprotective factors are associated with the amplification of enhancement of cell survival signal or inhibition of apoptosis signal,and thrus maintain the balance between the two signal pathways.
7.Establishment of Drug-Resistant Cell Lines of Acute Myeloid Leukemia and Correlation of Sirt1 and PGC-1α Expression Levels with Drug Resistance.

OBJECTIVE:
To construct cytarabine-resistant acute myeloid leukemia (AML) cell lines, and explore the correlation between Sirt1, PGC-1α expression levels and drug resistance.
METHODS:
Human acute promyelocytic leukemia Kasumi-1 cells were induced by the method of gradually increasing the concentration of Ara-C drug. The IC50 value of Kasumi-1 cells before and after drug addition was detected by CCK-8 method, so as to construct Ara-C resistant cell lines. The expression levels of Sirt1 and PGC-1α mRNA in Kasumi-1 drug-resistant cell lines and their parental cell lines were detected by real-time fluorescence quantitative PCR, and the expression levels of Sirt1 and PGC-1α protein in kasumi-1 drug-resistant cell lines and their parental cell lines were detected by Western blot.
RESULTS:
The constructed Kasumi-1 cell line had common morphological characteristics of drug-resistant cell lines under microscope, and the drug resistance index was greater than 5, indicating that Kasumi-1 drug-resistant cells had good drug resistance after the construction. The RT-qPCR and Western blot assays showed that the expression levels of Sirt1 and PGC-1α mRNA and protein in the drug-resistant cell lines were higher than those of the parental cell lines (P<0.001).
CONCLUSION
AML cell lines resistant to Ara-C can be successfully induced by the method of gradually increasing the concentration, and the co-high expression of Sirt1 and PGC-1α may mediate the drug resistance of AML cells.

8.Minimally invasive percutaneous nephrolithotomy under ultrasound and X-ray guidance for treatment of complicated renal calculi.

OBJECTIVETo evaluate the therapeutic effect of minimally invasive percutaneous nephrolithotomy (MPCNL) under ultrasonic and X-ray guidance in the treatment of complicated renal calculi.
METHODSA retrospective analysis was conducted among 36 patients undergoing MPCNL under ultrasonic and X-ray guidance for complicated renal calculi. The general clinical data, stone size, operative time, blood loss, complications and stone clearance rate were analyzed.
RESULTSDouble working channel was established accurately under ultrasonic and X-ray guidance in the 36 cases. The stone clearance rate in one treatment session was 80.56%, with an average operation time of 100 min and a mean hospital stay of 13.5 days (13-15 days) after operation. A second lithotripsy was performed in 5 cases and no residual renal calculi were found 1 month after the operation. Sixteen out of the 18 patients with preoperative elevation of serum creatinine recovered normal creatinine level after the operation.
CONCLUSIONMPCNL under ultrasonic and X-ray guidance is safe and effective for treatment of complicated renal calculi and is associated with few postoperative complications.

9.Formulation Optimization of the Volatile Oil Microcapsules of Cinnamomi Ramulus and Angelicae Sinensis Radix and Their Pharmacokinetics in Vivo

OBJECTIVE To optimize the preparation technology of the volatile oil microcapsules of Cinnamomi Ramulus and Angelicae Sinensis Radix, characterize the microcapsules prepared after the formulation optimization and study pharmacokinetics characteristics in rats. METHODS Spray drying was used to prepare the volatile oil microcapsules of Cinnamomi Ramulus and Angelicae Sinensis Radix. The preparation process was optimized by Box-Behnken response surface method with the drug loading and encapsulation efficiency as indexes. The microcapsules were characterized by Fourier transform infrared spectroscopy(FT-IR) and scanning electron microscopy(SEM). Blood samples were collected after gastric administration at a dose of 100 mg·kg-1. HPLC method was adopted in the plasma concentration determination, and pharmacokinetics behavior in vivo was also compared. RESULTS The optimal formulation: core material-capsules material ratio was 1∶1.7, capsules material concentration was 10.25%, sodium starch octenyl succinate∶maltodextrin was 3.8∶1, high pressure homogenization pressure was 20 MPa and the inlet temperature of spray drying was 185 ℃. Under the optimized conditions, the drug loading was (18.94±1.09)% and the encapsulation rate was (96.03±2.91)%, respectively. The results of FT-IR and SEM showed that the essential oils had been successfully coated in the capsule wall material. The microcapsules were basically spherical in shape, with concave surface but no obvious cracks. The main pharmacokinetic parameters such as tmax, t1/2, CL, AUC0-t, k10,k12, and k21 of microcapsules had significant difference of the volatile oil(P<0.05 or P<0.01). Microcapsules effectively prolonged the circulation time of volatile oil in the body, promoted the absorption of drugs in the body and the oral bioavailability was enhanced to 2.62 times. CONCLUSION The model established by the Box-Behnken design-response surface method can be used to optimize the formulation of volatile oil microcapsules of Cinnamomi Ramulus and Angelicae Sinensis Radix with great prediction effect. The microcapsules prepared by the optimized process has good drug loading properties and the bioavailability of volatile oil is increased.
10.Relationship between classroom lighting and poor vision of primary and middle school students

Objective:
To investigate the relationship between classroom lighting and poor vision of primary and middle school students of poor vision with classroom natural light selecting and artificial lighting, so as to provide reference and basis for the prevention and control of eyesight of primary and middle school students.
Methods:
A total of 1 734 students from 45 classrooms in 7 primary and secondary schools (2 in primary school, 2 in junior high school, and 1 in vocational school) in Baiyun District, Guangzhou were selected by stratified cluster sampling method for research. The classroom lighting environment was monitored by the illuminometer, the naked eye vision of students was detected by 5 m standard logarithmic vision light box, and the basic information and myopia-related behaviors of students were investigated by questionnaire. And the correlation between poor vision of primary and middle school students and classroom lighting was analyzed.
Results:
The poor vision rate of primary and middle school students in Baiyun District of Guangzhou was 74.2%(1 286), the girls’ rate(79.7%) was higher than boys’(69.4%), the rate of senior high school students(63.4%) was higher than that of middle school students(81.1%), the rate of vocational school students(82.8%) was higher that of primary school students(60.2%), the rate of resident students(78.5%) was higher than that of non-resident students(69.6%). The results of multivariate analysis after controlling for confangulation factors showed that average illumination on the blackboard, and uneven illumination on the desk were associated with higher risk of poor vision[OR(OR95%CI)=1.51(1.01-2.25), 1.42(1.02-1.98),P<0.05)].
Conclusion
Poor eyesight of primary and middle school students in Baiyun District of Guangzhou city is serious, especially that of female students, senior high school students and resident students. There is a significant correlation between classroom lighting and poor vision in primary and middle school students. The blackboard and desk lighting are associated with higher risk of poor vision in primary and middle school students.
