1.Clinical significance and influence factor of PS_2 protein expression in breast cancer

Objectives:To study the relationship between the expression of PS2 protein and estrogen receptor(ER) and progesterone receptor(PR) ,c-erbB2 status and prognosis in breast cancer.Methods:PS2 protein expressions in 52 cases of breast cancer were detected by immunohistochemical method.Results:The positive rate of PS2 protein was 59.6 %(31/52) in 52 patients.There was significant correlation between PS2 protein and menopausal status,ER status,PR status respectively.Tumor size,clinical stage and lymph node status were not significantly related with the expression of PS2 protein.Patients with PS2 protein positive expression had higher 5-year-survival rate than those with PS2 protein negative expression.Conclusion:PS2 protein appears to be an independent and valid prognostic factor of breast cancer,and detection of PS2 protein expression was useful for treatment of breast cancer.

3.Biological ingredient analysis of traditional Chinese medicines utilizing metagenomic approach based on high-throughput-sequencing and big-data-mining.

The quality of traditional Chinese medicines (TCMs) has been mainly evaluated based on chemical ingredients, yet recently more attentions have been paid on biological ingredients, especially for pill-based preparations. It is a key approach to establish a fast, accurate and systematic method of biological ingredient analysis for realization of modernization, industrialization and internationalization of TCMs. The biological ingredient analysis of TCM preparations could be abstracted as the identification of multiple species from a biological mixture. The metagenomic approach based on high-throughput-sequencing (HTS) and big-data-mining has been considered as one of the most effective methods for multiple species analysis of a biological mixture, which would also be helpful for the analysis of biological ingredients in TCMs. Simultaneous identification of diverse species, including the prescribed species, adulterants, toxic species, protected species and even the biological impurities introduced through production process, could be achieved by selecting appropriate DNA biomarkers, as well as applying large-scale sequence comparison and data mining. By this approach, it is prospective to offer an evaluation basis for the effectiveness, safety and legality of TCM preparations.

5.Study on the Drug Resistance Related Genes and Drug Resistance of Acinetobacter Baumannii from 2 0 1 1 to 2 0 1 5

Objective To investigate the changes of drug resistance related genes and drug resistance of Acinetobacter Bau-mannii in hospital,and provide evidence for rational use of antibiotics in clinic.Methods A retrospective analysis of Shaanxi Corps Hospital of Chinese Peoples’s Armed Police Forces from 2011 to 2015 from clinical samples of 11 521 specimens were isolated and cultured 1 861 strains of Acinetobacter Baumannii,drug susceptibility test by K-B method.The antimicrobial minimum inhibitory concentration (MIC)according to the 2014 edition of the CLSI judgment criteria of judgment.Results 1 861 strains of Acinetobacter Baumannii were detected drug resistance genes gyrA,parC,OXA-51,OXA-23,int on the 1,I TEM,AAC (6’)-1,AAC (3’)-1,ant (3)-1,ant (2)-1 and Caro gene detection rates were 50.4%,72.1%,70.9%,55.7%, 56.2%,65.6%,12.9% and 89.9% respectively,and SHV,IMP and VIM genes were not detected.The sequencing results showed that gyrA and parC gene mutation was the main cause of quinolone resistance.The antibiotic resistance of Acineto-bacterBaumannii was an increasing trend year by year,and 1 861 strains has 1 419 strains showed multiple drug resistance, all strains were sensitive to polymyxin.In 2015,quinolones aminoglycosides resistance rate was more than 65%.To imipen-em,meropenem and Cefoperazone/sulbactam were 35.17%,36.01% and 42.40%.Conclusion The detection rate of Acine-tobacter Baumannii is increasing year by year,and the drug resistance and multi drug resistance is increasing year by year.It is necessary to strengthen the clinical rational drug use andcontrol the hospital infection.
6.Perioperative nursing of 83 patients with sialolithiasis treated by sialoendoscope

This paper introduces the perioperative nursing of 83 sialolithiasis patients undergoing sialolithectomy by sialoendoscope. The key points of successful nursing care were careful preoperative psychological nursing and equipment preparation, correct and skilled intraoperative cooperation, and early postoperative health instruction. As a result, all the patients underwent the surgery succesfully and recovered well except 3 cases who needed gland removal later.
7.Over-expression of GSTP1 increases oxaliplatin-induced apoptosis in hu-man hepatoma HepG2 cells

AIM: To investigate the effect of GSTP1 over-expression on the sensitivity of human hepatoma HepG2 cells to oxaliplatin (OXA).METHODS: Adenovirus carrying GSTP1 (Ad-GSTP1) was used to infect HepG2 cells for establishing the cell line over-expressing GSTP1.The cells were randomly divided into 6 groups: control, vehicle, Ad-GSTP1, OXA, OXA +vehicle and OXA +Ad-GSTP1.The cell survival rates were examined by CCK-8 assay, and the apoptotic rates were analyzed by flow cytometry.The protein levels of GSTP1, Akt, mTOR, p-Akt and p-mTOR were deter-mined by Western blot.RESULTS: OXA decreased the cell survival rate in a dose-dependent manner (P <0.05).The protein expression of GSTP1 increased after transfection with adenovirus.At basal level, up-regulation of GSTP1 signifi-cantly decreased the cell survival rate, increased the cell apoptosis, and inhibited the phosphorylation levels of Akt and mTOR (P <0.05).Moreover, GSTP1 over-expression enhanced the effect of OXA on the cell viability, cell apoptosis, and further inhibited the phosphorylation levels of Akt and mTOR ( P <0.05).CONCLUSION: Over-expression of GSTP1 augments the enhanced effect of OXA on HepG2 cell apoptosis, which may be related to the inactivation of Akt/mTOR signaling pathway.
8.Inhibition effects of complement depletion with CVF on cardiac allografts rejection in inbred strain rats

Objective To study the efficacy of complement depletion with CVF in preventing a-cute rejection after heart allotransplantation in inbred strain rats. Methods Inbred male Lewis rats were used as recipients and Brown-Norway rats as donors, and the heterotopic heart transplantation model was established. The allografts were divided into 2 groups (n=8 in each group). After infusing low-dose CVF 20?g/kg to the CVF-treated group, cardiac allograft survival time was observed on 4 rats of each group. The remaining 4 rats in each group were killed respectively at day 1,3,5 and 6, the pathological grade for acute rejection, the complement activity in serum, the deposition of C3 on tissue, and the extent of infiltration by CD3+ T cells were compared. Objective To evaluate the effects of dexmedetomidine used to supplement analgesia with sufentanil on the stress response and inflammatory response after cardiac valve replacement with cardiopulmonary bypass (CPB).Methods Forty-four patients of both sexes,aged 18-64 yr,with body mass index of 18-30 kg/m2,of American Society of Anesthesiologists physical status Ⅱ or Ⅲ (New York Heart Association Ⅱ or Ⅲ),scheduled for elective cardiac valve replacement with CPB,were randomly divided into sufentanil analgesia group (group S,n =20) and dexmedetomidine supplementing sufentanil analgesia group (group DS,n=24) using a random number table.After induction of general anesthesia,the patients were endotracheally intubated and mechanically ventilated.Combined intravenous-inhalational anesthesia was used to maintain anesthesia.Dexmedetomidine was infused at 0.3 μg · kg-1 · h-1 from closure of the chest to the end of surgery in group DS,while the equal volume of normal saline was given instead in group S.The patient-controlled intravenous analgesia (PCIA) was used for postoperative analgesia.PCIA solution contained sufentanil 2 μg/kg in 100 ml of normal saline in group S,or sufentanil 2 μg/kg plus dexmedetomidine 3 μg/kg in 100 ml of normal saline in group DS.The PCIA pump was set up with a 0.5 ml bolus dose,a 5 min lockout interval and background infusion at a rate of 1.5 ml/h.Visual analogue scale score was maintained ≤ 3.When visual analogue scale score ≥ 4,morphine 0.1 mg/kg was injected intravenously.The occurrence of analgesics-related respiratory depression,bradycardia and hypotension was recorded.At the end of surgery (T0),and 12,24,48 and 72 h after surgery (T1-4),blood samples were collected from the central vein for determination of serum cortisol,β-endorphin,C-reactive protein,interleukin-2 (IL-2),and IL-6 concentrations by enzyme-linked immunosorbent assay.Results No analgesics-related adverse events were detected in the two groups.Compared with group S,the requirement for morphine was significantly decreased,the serum cortisol concentration was decreased at T2-4 and the serum β-endorphin,C-reactive protein concentrations were decreased at T1-4,the serum IL-2 concentrations were increased at T1-4,and the serum IL-6 concentrations were increased at T3 in group DS (P<0.05).Conclusion Dexmedetomidine used to supplement analgesia with sufentanil can alleviate the stress response and inflammatory response after cardiac valve replacement with CPB.
10.Efficacy and Safety of Autologous Adipose-derived Stem Cells Transplantation in Patients with Type 2 Diabetes Mellitus

Objective to evaluate the clinical efficacy and safety of ADSCs infusion in patients with type 2 diabetes mellitus(t2DM). Methods Autologous ADSCs were cultured and identified. totally 48 cases of type 2 diabetes patients were divided into two groups:24 cases in A group re-ceived ADSCs and conventional treatment,while the other 24 cases in B group received conventional treatment. With 15 months′follow-up,FPG,2h-PGh,HbA1c,FC-P,AUCC,insulin medication changes and security on post-treatment 3,6,12 and 15 months were calculated. Results the AD-SCs which cultured met the clinical application of standards. FPG,2h-PGh and HbA1c in both groups were significantly decreased,and the values in A group were more impressive than B. there were significant improvement in FC-P,AUCC and insulin of A group compared with B group. there was not treatment related adverse events. Conclusion the clinical data demonstrate that the treatment with ADSCs can improve metabolic control and beta cell function in patients with t2DM. Moreover,the safety of the treatment method was considerable.
