Chinese Journal of Tissue Engineering Research 2015;19(19):3096-3101
BACKGROUND:Type 1 diabetes melitus is an autoimmune attack characterized by the selective destruction of pancreatic cels. Patients need to rely on external insulin to survive, and there is a lack of specific treatment. OBJECTIVE:To review the source of al kinds of stem cels, differentiation capacity, self-exclusion and existing problems, and to provide the basis for regeneration treatment of type 1 diabetes melitus. METHODS: In order to search relevant article about stem cel therapies for type 1 diabetes melitus from PubMed database (from 2003 to 2015), a computer-based search was performed using the key words of type 1 diabetes melitus, stem cel, stem cel therapy, regeneration treatment, stem-cel differentiation, stem-cel transplantation, immune suppression, insulin secreting cels, islet β-cels, therapeutic effect in English. Finaly, 42 articles were chosen for further analysis. RESULTS AND CONCLUSION:Stem cel therapy for type 1 diabetes melitus has attracted quite a lot of attentions. Restoration of damaged β-cels by exogenous stem cel transplantation is the currently ideal therapeutic options, that is, stem cels cultured under suitable conditions can be induced into islet β-cels that have endocrine function. Through al kinds of research, this paper expounds the source of stem cels, directional differentiation, purification, immune suppression characteristics and existing problems, and it also has a discussion for the occurrence and development of type 1 diabetes melitus so as to select the best scheme of stem cels, which helps to cure the disease.

7.Optimization on formulation of sinomenine hydrochloride pulsatile drug delivery tablets by central composite design-response surface method
Chinese Traditional and Herbal Drugs 1994;0(07):-
Objective To optimize the formulation of sinomenine hydrochloride pulsatile drug delivery tablets by central composite design-response surface method. Methods The tablets containing sinomenine hydrochloride were prepared by dry-compression coating technique. The influence factors included the amount of sodium carboxymethyl starch in core tablets, the ratio of HPMC/carrrageenan,and the amount of matrix materials in coating film. The evaluation parameter was lag time.Experiments were done on the central composite design, and the data were simulated using multi-linear equation and second-order polynomial equation. The possibly optimal formulation was predicted by response surface method. The lag time of the tablets prepared under the optimum condition was compared with the predicted. Results The lag time was simulated using second-order polynomial equation and the regression coefficient was 0.993 7.The lag time in vitro of the tablets prepared under the optimum conditions was about 6 h,then drug released in pulsatilerelease character.Bias between the observed and predicted values of the lag time was within ?4.43%.Conclusion The sinomenine hydrochloride pulsatile tablets could release drug quickly in vitro at the predetermined time.Central composite design-response surface method can be used to optimize the formulation and the model developed in this study proves to be highly predictable.
8.The expression and significance of lysyl oxidase like-2 in pulmonary fibrosis mice
Chinese Journal of Rheumatology 2015;19(9):606-610,后插1
Objective To assess whether lysyl oxidase like-2 (LOXL2) has a predictive or prognostic value in pulmonary fibrosis mice, and identify the relationship between LOXL2 and transforming growth factor (TGF)-β 1.Methods Male C57BL/6 mice (n=50) were randomly divided into the experimental group (n=40) and controls (n=10).Each mouse in the experimental group was induced by injecting bleomycin into trachea and mice were sacrificed on day 3,7,14 and 28,respectively.The concentrations of LOXL2,TGF-β 1 of the serum and lung homogenates were measured by enzyme-linked immunosorbent assay.One-factor analysis of variance (ANOVA) and nonparametric rank sum test was used for the comparisons between the two groups.Associations between these factors were analyzed by Spearman's test. Results ① The levels of serum and lung homogenates LOXL2 in the experimental group were obviously increased as compared to those in the controls [serum:(56±48) vs (25±23),P<0.05;lung homogenates:(61±32) vs (22±16),P<0.05].And in the experimental group,a positive correlation was found between the level of LOXL2 and the radiological index, the pathological score of inflammation and fibrosis.(Serum:r=0.435,0.533,0.335,P<0.05;lung homogenates:0.675,0.736,0.526,P<0.05).② The levels of lung homogenates TGF-β 1 in the experimental group were obviously increased as compared to those in the controls [(302±197) vs (64±16),P<0.05].A positive correlation was found between the level of lung homogenates LOXL2 and the level of lung homogenates TGF-β1 (r=0.520,P<0.05).Conclusion LOXL2 participates in the pathological process of bleomycin induced pulmonary fibrosis in mice.It predicts pulmonary fibrosis in the preclinical stage and is associated with increased risk for disease progression. The elevated levels of LOXL2 and TGF-β 1 may have some relationship. They both are involved in the disease progression of pulmonary fibrosis.
