1.Biological ingredient analysis of traditional Chinese medicines utilizing metagenomic approach based on high-throughput-sequencing and big-data-mining.

The quality of traditional Chinese medicines (TCMs) has been mainly evaluated based on chemical ingredients, yet recently more attentions have been paid on biological ingredients, especially for pill-based preparations. It is a key approach to establish a fast, accurate and systematic method of biological ingredient analysis for realization of modernization, industrialization and internationalization of TCMs. The biological ingredient analysis of TCM preparations could be abstracted as the identification of multiple species from a biological mixture. The metagenomic approach based on high-throughput-sequencing (HTS) and big-data-mining has been considered as one of the most effective methods for multiple species analysis of a biological mixture, which would also be helpful for the analysis of biological ingredients in TCMs. Simultaneous identification of diverse species, including the prescribed species, adulterants, toxic species, protected species and even the biological impurities introduced through production process, could be achieved by selecting appropriate DNA biomarkers, as well as applying large-scale sequence comparison and data mining. By this approach, it is prospective to offer an evaluation basis for the effectiveness, safety and legality of TCM preparations.

2.Effects of Shuxuetong on Hemodynamics and Serum sICAM-1, IL-6, TNF-α in Patients with Cerebral Infarction

Objective To investigate the effects of Shuxuetong on hemodynamics and serum sICAM-1,IL-6,TNF-α in patients with cerebral infarction.Methods A hundred patients with cerebral infarction were randomly divided into observation group and control group from Jan 2011 to Jan 2015.Fifty patients in control group were treated with conventional therapy alone and 50 patients in observation group were treated with conventional therapy combined with intravenous injection with Shuxuetong.The changes of hemodynamics were observed before and after treatment and the levels of serum sICAM-1,IL-6,TNF-α were detected by enzyme linked immunosorbent assay (ELISA).Results No statistical significance was found in hemodynamics of control group before and after treatment (P >0.05).The indexes of hemodynamics in observation group were significantly improved compared with those in observation group before treatment and those in control group after treatment (P< 0.05).The levels of serum sICAM-1,IL-6,TNF-α were significantly reduced in two groups after treatment (P <0.01) and the levels of serum sICAM-1,IL-6,TNF-α after treatment were significantly lower in observation group than those in control group (P<0.01).Conclusion Shuxuetong can effectively improve the indexes ofhemodynamics and reduce the levels of serum sICAM-1,IL-6 and TNF-α.It has a positive effect on the treatment of cerevral infarction.

4.Effects of Antihypertensive Protein from Pheretima on Angiotensin Ⅱ and Angiotensin Ⅱ AT_1 Receptor in Spontaneously Hypertensive Rats

OBJECTIVE:To investigate the effects of the antihypertensive protein from Pheretima on blood pressure, angiotensinⅡ and angiotensin Ⅱ AT1 receptor in spontaneously hypertensive rats (SHR).METHODS: The dynamic change of blood pressure in SHR after singel intravenous injection of antihypertensive protein from Pheretima was observed. After intervention with antihypertensive protein from earthworm for 28 d,the levels of blood pressure, angiotensinⅡ and expression of angiotensin Ⅱ AT1 receptor in SHR were detected. RESULTS: Either single intravenous injecion or multiple dosing of the antihypertensive protein from Pheretima could significantly reduce the blood pressure in SHR (P0.05).The expression of angiotensin Ⅱ AT1 receptor in kidney of SHR model increased significanlty compared the normal control (wistar rats), but decreased after the intervention of the antihypertensive protein from Pheretima.CONCLUSION: The antihypertensive protein from Pheretima has antihypertensive effect on SHR. The mechanism may be related to the reduction of angiotensinⅡ level and lowering of the expression of a angiotensin Ⅱ AT1 receptor in kidney.
5.Effects of Metformin on Serum Sex Hormone and Leptin in Ovariectormized Female Rats

OBJECTIVE:To study the effects of metformin (MF)on serum sex hormone and leptin contents in ovariectormized female rats. METHODS: 40 SD rats were randomly divided into four groups: control group(normal saline 5 mL? kg-1?d-1,NS group),OVX group(normal saline 5 mL?kg-1?d-1),OVX+low and high dosage MF group(135,270 mg? kg-1?d-1,MFlow and MFhigh group),all group were ig drugs for 30 days on end.The serum sex hormone indexes including E2,P,LH,LSH and leptin content were measured with RIA. RESULTS:Compared with NS group,the serum E2 and P content were significantly lower(P0.05). CONCLUSIONS: MF can significantly improve sex hormone secretion and lower serum content of LEP in OVX rats.
6.Inhibition effects of complement depletion with CVF on cardiac allografts rejection in inbred strain rats

Objective To study the efficacy of complement depletion with CVF in preventing a-cute rejection after heart allotransplantation in inbred strain rats. Methods Inbred male Lewis rats were used as recipients and Brown-Norway rats as donors, and the heterotopic heart transplantation model was established. The allografts were divided into 2 groups (n=8 in each group). After infusing low-dose CVF 20?g/kg to the CVF-treated group, cardiac allograft survival time was observed on 4 rats of each group. The remaining 4 rats in each group were killed respectively at day 1,3,5 and 6, the pathological grade for acute rejection, the complement activity in serum, the deposition of C3 on tissue, and the extent of infiltration by CD3+ T cells were compared. 7.Acquisition of Pulse Characteritics Based on Wavelet Module Maximum Principle.

Objective To study a wavelet method for acquiring the characters of pulse wave based on the principle of wavelet transform which can effectively solve the problem of inaccuracy of the conventional methods. Method The method of wavelet module maximum was used to divide the pulse wave according to periods and basically decomposed it, by arranging in time order and symbolizing the module maximum character points of pulse wave signal at the ascending and descending edges as well as the crest and trough. Result Through the method, the unobvious dicrotism, trail wave and the anomalistic wave appearing at the ascending and descending edge of the main wave, and multi-scale characters and all kinds of characters in time domain of pulse signal were further acquired more accurately. Conclusion Simplicity, quickness and accuracy are achieved by focusing on the character points of pulse signals. It provides a new means for further studies of classification and identification of pulse signals.
8.Inhibitory effect of LIGHT-Fc gene transfection on human esophageal squamous carcinoma cell line Eca109

Objective To investigate the in vitro inhibitory effect of LIGHT on human esophageal carcinoma cells. Methods LIGHT Fc expression vector was transfected into human esophageal squamous carcinoma cell line, Eca109. The inhibitory effect of LIGHT gene on cell growth was detected by MTT and cell growth curve. The expressions of LT?R and HVEM were detected by RT PCR. Results Expression of LIGHT Fc gene could inhibit Eca109 cell proliferation. The growth curve of Eca109/LIGHT was significantly lower than that of the control group in the culture medium containing 1% FCS. 9.The effect of ecoimmunonutrition enteral nutrition on systemic inflammatory response and prognosis in severe acute pancreatitis

Objective To evaluate the effect of ecoimmunonutrition enteral nutrition on systemic inflammatory response and prognosis in patients with acute severe acute pancreatitis. Methods Fifty-seven patients with severe acute pancreatitis were randomly divided into two groups: parenteral nutrition (PN) group(28 cases), and ecoimmunonutrition(EIN) group (29 cases), each of the two nutrition formulas was given to each group respectively for 7 days. The level of plasma endotoxin and serum concentrations of TNF-α ,IL-1β ,IL-6 and IL-10 and monocyte NF-κB were tested at admission,1 d,3 d,5 d,7 d after nutrition support. Statistics the two groups results of clinical prognosis. Results The levels of endotoxin, TNF-α ,IL-1β ,IL-6 and IL-10 , NF-κB activities in EIN group were no significant difference compared with PN group at admission. After nutrition support 7 d, the levels of endotoxin, TNF-α ,IL-1β ,IL-6 and IL-10 , NF-κB activities in EIN group were (2.70 ± 0.13) ng/L, (30.13 ± 8.12) ng/L, (20.17 ± 8.04) ng/L, (36.43 ± 8.24)ng/L, (86.45 ± 14.54) ng/L, (70.4 ± 3.2)% respectively, which all were significantly lower than those in PN group [(3.25 ±0.32) ng/L, (313.42 ± 144.35) ng/L,( 155.29 ±32.78) ng/L, (324.15 ± 31.47) ng/L, (472.72 ±48.55) ng/L, (88.4±53)% ](P<0.05 or <0.01). Conclusion EIN can attenuate endotoxemia.decrease NF-κB activities and concentrations of cytokines, maintain the balance of pro-and anti-inflammation, improve patients' condition and prognosis.
10.Clinical features and early management of combined thoracoabdominal injury

Objective To evaluate the clinical features and early management of combined thoracoabdominal injury (CTI).Methods A retrospective study was carried out in 29 cases with CTI treated surgically.There were 21 cases with penetrating injuries and 8 cases with blunt injuries.All cases were associated with penumothorax and/or hernothorax at various degrees and 13 cases complicated with shock.In this study thoracotomy was performed in 17 cases,laparotomy in 6 cases,thoracotomy plus laparotomy in 5 cases and combined thoraco-laparotomy in 1 case.Results The injury severity score in this series was (40.16±15.23) scores.Four cases were died from hemorrhagic shock (2 cases),pericardial tampenade(1 case) and multiple organ failure(1 case).Conclusions CTI has high mortality rate,and the most frequent cause of death is hypovolemic shock.The operative approach is based on individual injury condition.Early diagnosis and timely surgical intervention are key to improving the cure rate and reducing mortality in severe CTI.
