1.Association of peroxisome proliferator-activated receptor-α gene polymorphism and the risk of metabolic syndrome in Southern Chinese

Objective To investigate the association between the peroxisome proliferator-activated receptor-or (PPARα) polymorphism rs1800206 (c.484C > G,L162V) and the risk of metabolic syndrome (MES)Methods There were 1184 subjects aged 40-60 years recruited from our hospital between March 2010 and March 2011.The PPARα polymorphism 484C > G was genotyped using TAQMAN assay by real-time PCR. The relationship between PPARα polymorphism and MES risk were investigated.Results The genotype frequencies were 91.4%,8.4% and 0.2% for the PPARα CC,CG and GG,respectively.This SNP was in Hardy-Weinberg equilibrium(P =0.845).Compared with the most common CC genotype,the variant genotypes(CG + GG) had higher fasting glucose((5.82 ± 1.59) mmol/L vs (5.49 ± 1.17) mmol/L,t =2.630,P =0.009) and LDL-C levels((3.53 ± 1.03) mmol/L vs (3.36 ± 0.65) mmol/L,t =2.376,P =0.018) and lower HDL-C levels ((1.56 ± 0.35) mmol/L vs (1.65 ± 0.43) mmol/L,t =2.430,P =0.015).Furthermore,the 484G variant genotypes(CG + GG)was associated with the risk of MES after adjusting for age,gender,education,BMI and lifestyle (smoking and alcohol status) (adjusted OR =1.89 ;95% CI =1.09-3.23,P =0.012).Condusion PPARα polymorphism rs1800206 is a significant independent predictor of the MES in Southern Chinese.
3.Influence of percutaneous coronary intervention on brain natriuretic peptide and ventricular remodeling ;in patients with acute ST elevation myocardial infarction

Objective:To explore influence of percutaneous coronary intervention (PCI)on brain natriuretic peptide (BNP)level and ventricular remodeling in patients With acute ST elevation myocardial infarction (STEMI).Meth-ods:According to their therapeutic methods,a total of 151 patients diagnosed as STEMI Were divided into group A (n=52,received emergency PCI treatment),group B (n=49,received selective PCI treatment)and group C (n=50,received standard medication).BNP level and left ventricular remodeling Were compared among three groups before treatment and after treatment.Results:There Was no significant difference in BNP level among three groups before treatment (F=0.01,P>0.05),BNP level significantly decreased among three groups after treatment,com-pared With group B and C,there Was significant decrease in BNP level [(466.2±113.5)ng/L vs.(601.4±126.7) ng/L vs.(242.7±101.3)ng/L]in group A (P<0.01 all).There Were no significant difference in left ventricular end-diastolic volume (LVEDV),left ventricular end-systolic volume (LVESV),left ventricular ejection fraction (LVEF)and left ventricular fractional shortening (LVFS)among three groups before treatment (P>0.05 all);af-ter treatment,there Were significant increase in above ventricular indexes in three groups (P<0.05 all),compared With group C,there Were significant increase in LVFS [(22.6±6.9)% vs.(28.9±7.6)% vs.(27.5±7.3)%]in group A and B,compared With group B and C,there Was significant increase in LVEF [(53.1±6.5)% vs.(50.9± 5.7)% vs.(57.9±6.2)%]in group A (P<0.01 all).Conclusion:PCI could effectively improve ventricular re-modeling and decrease BNP level in STEMI patients,and emergency PCI shoWs more significant therapeutic effects than selective PCI.
4.Association of EPHX2 polymorphism G860A with hypertension in middle ages

Objective To investigate the association between the EPHX2 polymorphism G860A and the risk of hypertension in mild ages. Methods In a hospital based case-control study, one common polymorphism C860A in EPHX2 gene in a case-control study of 100 hypertension and 300 age- and sex frequency - matched disease-free controls were genotyped in a Southern Chinese population. SAS 9. 13 was used toanalysis the polymorphism and hypertension risk. Results Genotype frequencies of EPHX2 G860Alocus between the cases and the controls were significantly different (P =0. 01). Compared with the most common 860GG genotype, the 860GA heterozygote had an increased risk of hypertension (adjusted OR =1.98; 95%CI = 1.19 -3.51), the AA homozygote had a further increased risk of hypertension (adjusted OR =2. 84; 95%CI = 1.01 -6. 11). There was a significant trend for an allele dose effect on risk of hypertension (P trend = 0. 03). Conclusions EPHX2 polymorphism G860A is associated with an increased risk of hypertension, and the 860A variant may be a marker for susceptibility to hypertension.
5.Effect of catgut embedding in the Liumai acupuncture point on myocardial potential, myoelectric potential and tension in the rumen of Tibet sheep

One of the purposes for the improvement of forages efficient utilization in rumens was to adjust the physiological function of ruminant in Tibet,and to improve the disadvantage influences under the altitude environment.The electrocardiography,rumen electromyography and tension in the rumen were drawn the before and after catgut embedding in the liumai acupuncture point to the Tibet sheep.The result showed that catgut embedding in the liumai acupuncture point could promoted the blood circulation,alleviated the overload of myocardial and remedied the right deviation of heart electrical axis up to 40°.The VPP and RA of rumen were significantly lower and the tension of rumen decreased 1/3 after catgut embedding.The result indicated that it could relaxed the right ventricular hypertrophy,reduced the tension of rumen,improved the function of gastrointestinal tract and blood circulation,ensured sufficient time to digest the forage in rumen.
6.Expression of melanoma antigen-a mRNA in coal tar pitch-induced lung cancer tissues in mice

Objective: To investigate the expression of mage-a mRNA in lung cancer tissues of mice induced by coal tar pitch(CTP) fume and to discuss the above model for lung cancer immunotherapy with mage-a. Methods: Tumor tissue samples of lung cancer and paired non-tumor tissues were obtained from 8 lung cancer mice. Total RNA was extracted and cDNA was synthesized. Nested polymerase chain reaction amplification using mage-a specific primer was performed to detect the expression of mage-a. The 2 clones of mage-a mRNA positive PCR products were sequenced by DNAs sequencer (PE-377). Results: Of 29 mice in the experimental group, 8 were induced to lung cancer.Among which 5 (5/8) expressed mage-a mRNA. The expression of mage-a gene was not found in adjacent lung tissues. The DNA sequencing confirmed that the target gene fragments in 2 samples of PCR products were mage-a cDNA. Conclusion: The mage-a gene is highly expressed in lung cancer in mice induced by CTP fume, suggesting that CTP-induced lung cancer in mice may be an ideal animal model for lung cancer therapeutic experiment with MAGE-A.
7.Factors influencing disinfection efficacy of ultraviolet ray

Objective To investigate the effect of various fa ctors on disinfection efficacy of ultraviolet ray. Methods The germicidal exper iment was carried out in a purified chamber. Standard bacteria sprayed in the p urified chamber was irradiated by using an 100 μw*cm-2*s-1ultrav iolet lamp under different c onditions. Results The disinfection efficacy showed correlation with tempe rature, humidity, intensity, way of radiation, electric fan running or not and different microorganisms, among which temperature showed the strongest effect on killing rate. Under the same condition, the efficacy of ultraviolet l amp in killing staphylococcus albus is superior to that in killing escheri chia coli, and the efficacy of killing the spore of bacillus subtilis is the l owest. Conclusions The way of air disinfection by a high intensity and low ozone ultraviolet lamp irradiation is reliable, safe, practical and feasi ble.
8.Analysis of oprD gene in imipenem-intermediate clinical isolates of Pseudomonas aeruginosa

Objective To analyze the mechanism of imipenem resistance in Pseudomonas aeruginosa clinical isolates. Methods? Antibiotic?resistance?was?analyzed?using?VITEK32?system.?Metallo?β-lactamase?activity?was?determined?by?double-disc?synergy?test.?Amp?C?β-lactamase?activity?was?determined?by?Kirby-Bauer?disc?method.?OprD?protein?was?analyzed?by?sodium?dodecyl sulphate-polyacrylamide gel electrophoresis. PCR was performed to amplify gene oprD. The amplified products were subject?to?sequencing?analysis.?The?phylogenetic?relationship?was?determined?using?random?amplified?polymorphic?DNA?(RAPD)?method. Results Membrane protein OprD was analyzed in 7 clinical isolates of imipenem-intermediate P. aeruginosa. Two strains were devoid of OprD proteins and the corresponding oprD genes were found disrupted by the insertion element ISRP10 in the coding regions. Five strains had OprD proteins with different sizes. Sequence analysis showed that the peptides ranged from 427 to 443 amino acids. Multiple amino acid substitutions and / or deletions were found within the Loop 1 through Loop 8 of the OprD secondary structures. Conclusions ISRP10 inactivation and amino acid substitutions in oprD gene confer imipenem resistance in the clinical isolates of P. aeruginosa.
9.Variation characteristics of peripheral blood β-catenin and DKK1 in patients with rheumatoid arthritis and the correlation with bone and joint damage

Objective To study the variation characteristics of peripheral blood β-catenin and DKK1 levels in patients with rheumatoid arthritis (RA) and the correlation with bone and joint damage.Methods One hundred and eight patients with RA (observation group) and 110 healthy individuals (control group) were selected.The bone mineral density(B MD) in the femur (femur neck,Ward area,greater trochanter,total femur area) and lumbar spine (L2,3,4) were measured by dual-energy X-ray absorptiometry instrument.X-ray for two hands were evaluated according to the Sharp score.The peripheral blood β-catenin and DKK1 levels were detected by enzyme-linked immunosorbent assay (ELISA).Multiple analysis was completed by multiple linear regression and Logistic regression.Results The peripheral blood DKK1 level in observation group was significantly higher than that in control group [(8.2 ±5.0) mg/L vs.(6.1 ±4.2)mg/L],there was statistical difference (t =2.452,P =0.025),but there was no statistical difference in β-catenin level between the 2 groups (t =15.947,P =0.142).Compared to control group,patients with RA had lower BMD at femur and lumbar spine (P < 0.01).The incidence of osteoporosis in observation group was significantly higher than that in control group [31.5% (34/108) vs.15.5% (17/110)],there was statistical difference (x2 =9.289,P =0.005).The correlation analysis results showed that the peripheral blood DKK1 was positive correlation with erythrocyte sedimentation rate (ESR),28-jonit disease activity score (DAS28),alkaline phosphatase (AKP) and two hands X-ray joint narrowing space score in patient with RA (P < 0.01 or < 0.05) ;the peripheral blood β-catenin level was positively correlated with ESR and AKP (P < 0.01 or < 0.05).The multiple linear regression results showed that the disease duration,health assessment questionnaires (HAQ),DKK1 influenced two hands X-ray joint narrowing space score; but the age,disease duration,HAQ influenced two hands X-ray bony erosion score.The multiple Logistic regression results showed that the two hands X-ray Sharp score was the risk factor of osteoporosis at femur in patients with RA; but the age was the risk factor of osteoporosis at lumbar in patients with RA.Conclusions Peripheral blood DKK1 level in patients with RA increases significantly,while there is no apparent alteration in β-catenin level.Peripheral blood DKK1 level is correlated with disease activity and joint space narrow score.
