1.Effect of X-ray on amino-acid transmitters in brain of epileptic rat

Objective To investigate the effective mechanism, time and rational dose of radiotherapy of epileptic rat. Methods To make rat model of epilepsy, and give 0, 24,12 or 6Gy X ray irradiation, and then detect the amino-acid contents in frontal lobe of those rats in 1 hour, 24 hours or 1 week after radiotherapy. Results Glu and GABA are the key amino-acids in radio effection. After 12Gy or 24Gy irradiation there were low Glu, high GABA and low Glu/GABA in frontal lobe of rat. After 1h, 24h and 1 week irradiation, the contents of GABA raised up while the ratio of Glu/GABA declined with a remarkable decline at 24h. Conclusions Radiotherapy to epileptic rat may set off a rapid and enduring change of GABA and Glu transmitters, and 12Gy may be the rational radio dose for epileptic rat.
2.Operative Treatment for Schwannoma in the Spinal Canal

Purpose: To retrospectively analyse the characteristics of diagnosis and operative treatment of 66 cases of scwannoma in the spinal canal. Methods: Sixty-six cases, 27 of the cervical region, 13 the thoracic and 26 of the lumbar and sacral regions were treated from 1983 to 1996. Clinical manifestation and types, and results of spinal radiography, myelography, CT scannig, MR imaging, operative treatment were separately described. Results: All of the 66 cases obtained timely and accurate diagnosis and treatment. The tumors in 60 patients were completely resected; and in remaining 6 patients subtotal excision was performed. The duration of follow-up averaged two years and eleven months and excellent and good results were achieved in 90.9% of the patients. Conclusions: 1) Schwannoma in the spinal canal is not uncommon; four in this series are malignant or with malignant tendency. 2) The forms of the tumor are with lots of variations. 3)An approximate position of the lesion can be determined by clinical neurological examination. Accurate diagnosis can be made by a procedure combining clinical and imaging examinations, operative exploration and pathological examination. 4) Total resection of the tumor may result in good restoration although it is sometimes difficult and risky.
3.Plasma visfatin levels in subjects with different states of glyeemia and its related factors

Fasting plasma visfatin was determined in subjects with various types of glucose tolerance. Visfatin level was (17.64±1.37)μg/L in subjects with normal glucose tolerance,and rose to (27.42±1.34) μg/L(P<0.05) in those with impaired glucose tolerance or impaired fasting glucose, and to(26.55 ± 1.64) μg/L(P< 0.05)in diabetic patients. No difference was found between obese and non-obese subjects. Positive correlation existed between visfatin and HbA1c(r=0. 317,P<0.01). Multiple linear step-wise regression showed that HbA1c and fasting plasma glucose had independent relation with the visfatin level. The result suggests that plasma visfatin may be related to glueometabolism and may play a role in the development of type 2 diabetes.
4.Enhancement of tendon-bone healing of rotator cuff with recombinant human bone morphogenetic protein-2 in rabbits:a histological and biomechanical study

[Objective]To evaluate the effectiveness of recombinant human bone morphogenetic protein-2 in the repair of rabbit rotator cuff injury.[Method]Forty-eight male New Zealand rabbits,aged 8 months,received an rotator cuff acute injury and reconstruction of the insertion of supraspinatus tendon on greater tuberosity of humerus.The rabbits were randomly divided into 3 groups postoperatively:(1) rhBMP-2 group:fibrin sealant(FS) containing rhBMP-2 was applied to the interface between the bone-tendon interface; (2)FS control group:only FS was applied;(3)blank control group:untreated after the surgery.Harvested 36 specimens underwent biomechanical analysis at the 2nd,4th,8th postoperative weeks respectively.Harvested 12 specimens underwent histological analysis at the 8th postoperative week.[Result]Histological examination showed that Sharpey's fibers were found in the interface with the formation of four-layer indirect insertion in the rhBMP-2 group at the 8th postoperative week.In the FS control group and blank group,the tendon-bone interface was filled with granulation tissue and part of Sharpey's fibers and the newly generated bone tissue.Biomechanical analysis displayed that the tensile strength and stiffness of bone-tendon interface increased time-dependently in all groups,and it in the rhBMP-2 group was significantly higher than those in the other two groups at any time-points (both P
5.Curative effect of oxaliplatin combined with S-1 in treatment of advanced colorectal cancer

Objective To analyze the curative effect of oxaliplatin combined with S-1 in the treatment of advanced colorectal cancer.Methods Totally 90 cases of patients with advanced colorectal cancer in Shangluo No.2 Provincial People's Hospital from January 2013 to August 2015 were divided into observation group and control group,45 cases in each group.Patients in control group were treated with oxaliplatin combined with fluorouracil,4 weeks for one cycle of treatment,and patients in observation group were treated with oxaliplatin combined with S-l,21 d for one cycle.The curative effects,adverse reactions,life quality,and one year survival rate were compared between two groups.Results After treatment,the total remission rate of observation group was higher than that of control group,and the disease control rate was lower than that of the control group,but there was no significant difference between the two groups.During the treatment,there were no significant differences in Ⅰ degree and Ⅱ degree adverse reactions between two groups;But the adverse reactions of level Ⅲ and level Ⅳ of gastrointestinal reaction,liver function damage,reduce the incidence of white blood cells in observation group were significantly lower than that of control group (P < 0.05).There was no significant difference between the two groups in the SF-36 scale scores.The one year survival rate of observation group was (21/45),and the survival rate of the control group after treatment was 42.22% (19/45) in the control group,and there was no significant difference between the two groups in the one year survival rate.Conclusion Compared with oxaliplatin combined with fluorouracil,oxaliplatin combined with S-1 has equivalent efficacy and life quality,but the adverse reactions in gastrointestinal tract,liver,and marrow were better.
6.Culture of rabbit osteoblasts digested by collagenase in DMEM containing fetal bovine serum

BACKGROUND: The skill to culture osteoblasts primarily has been well developed, however, variousdigestive enzymes can affect membrane protein of osteoblasts when they are used on primary tissue separately.OBJECTIVE: To avoid the damage from enzyme to cell as best possible,digest cranial osseous tissue with collagenase in DMEM containing fetal bovine serum and carry out in vitro culture of osteoblasts, then observe the effect of collagenase in DMEM containing fetal bovine serum on the digestion of osteoblasts.DESIGN: Control experiment.SETTING: Department of Orthopaedic Surgery, Tongji Hospital Affiliated to Huazhong University of Science and Technology.MATERIALS: This experiment was carried out in the Department of Orthopaedic Surgery, Tongji Hospital Affiliated to Huazhong University of Science and Technology from March to December 2004. Two Japanese white rabbits with big ears that were pregnant for 28 days were used in the experiment.METHODS: Type I collagenase with the same batch number was prepared into 0.1% collagenase with fetal bovine serum and 0.1% collagenase without fetal bovine serum by using culture medium containing 0.15 volume fraction of fetal bovine serum and DMEM solution respectively, serving as the enzyme digestive juice A and B for experimental group and control group respectively. Abdominal delivery was performed to take ont the fetal rabbit which was at embryonic 28 days under aseptic condition.Then, craniums of fetal rabbits were dissected, rinsed and chipped into pieces. 5 mL solution A and 5 mL solution B were added into the experimental group and control group at 37℃, respectively. Culture solution containing 0.15 volume fraction of fetal bovine serum was gradiently diluted into 0,1,2 and 4 folds in each group, and continued to digest and culture osteoblasts. The cellular survival rate was measured with trypan blue staining, and the osteoblast and its purity were identified with alkaline phosphatase(ALP) staining. Cellular growth was observed under the microscope.MAIN OUTCOME MEASURES: ①Cellular survival rate of two groups was detected with trypan blue staining. ② Cellular purity of two groups was detected with ALP staining in the cells. ③Cellular growth was distinguished by observing cellular morphology under the microscope.RESULTS: ① Results of trypan blue staining: Through cell counting, the rate of cells, which refused to be stained, of the experimental group reached 98%, and the cellular survival rate was high; that of control group reached 95%, and the cellular survival rate was also very high. ② Results of ALP staining in the cells: The area of ALP staining positive of experimental group was not less than 95%, and cellular purity was very high; but that of control group was not more than 75%, and cellular purity was very low. ③ Observation results under the microscope: Cells of experimental group were well stacked, convex and stereoscopic, they grew prosperously and had enough nutrition; while those of control group were not significantly in comparison with experimental group.CONCLUSION: Collagenase containing fetal bovine serum is used to digest the osseous tissue of cranium of fetal rabbits, and the cultured osteoblasts have typical properties of osteoblasts with simple component and high survival rate.
7.TRANSCAROTID ARTERY CHEMOTHERAPY FOR GLIOMAS: AN ANALYSIS OF 212 CASES

Objectives To evaluate the effectiveness of transcarotid artery chemotherapy for gliomas after surgery, and selection of drug, avenue of administration, and optional time for the therapy. Methods Beginning from 4 to 30 days after operation, Nimustine (ACNU, Japan) 2.5mg/kg was injected per carotid artery, once every week for three times as one course. A second course of treatment was given after an interval of 4 to 6 weeks. Results With the above regime, the effect was marked in 39 cases (18.8%), fairly effective in 44 cases (20.8%), only slightly effective in 59 cases (27.8%), no effect in 61 cases (28.8%), and failure in 5 cases (2.4%), the mean survival time was nearly 100 weeks. Conclusion Transcarotid artery chemotherapy for gliomas is helpful in prolonging survival period, with little side effects, easy to carry out, less expensive, and better accepted by the patients.

Objective:Effect of the low molecular weight natural tumor suppressor (LMW NTS) of human fetal hepatocyte and that of embryo calf hepatocyte on the growth of murine S 180 cells was observed.Methods:LMW NTS was isolated respectively from human fetal hepatocyte and embryo calf hepatocyte.(1)In vitro two layers agar plate culture method was used to detect the suppressive effect of the human fetal hepatocyte LMW NTS and the emtryo calf hepatocyte LMW NTS on the growth of murine S 180 cells.(2)Surviral of the mice bearing S 180 cells and treated respectively with intraperitoneal injections of the human fetal hepatocyte LMW NTS or the embryo calf hepatocyte LMW NTS(0.5 mg/one mouse/day for 21 days)was surveyed.Results:Both the human fetal hepatocyte LMW NTS and the embryo calf hepatocyte LMW NTS can obviously inhibit the growth of murine S 180 cell,and the suppressive effect was enhanced while the concentration of LMW NTS was increased.Survival of the mice bearing tumor that treated with intraperitoneal injections of the human fetal hepatocyte LMW NTS or the embryo calf hepatocyte LMW NTS was remarkably prolonged.Conclusion:The LMW NTS of embryo calf hepatocyte is similar to that of human fetal hepatocyte,and it possesses marked anti tumor activity both in vitro and in vivo.
9.Effects of propofol, etomidate and ethanol on GPCR mRNA expression in Daphnia pulex .

The mechanisms of general anesthesia, which was introduced about 170 years ago, remain poorly under- stood. Even less well understood are the effects of general anesthesia on the human body. Recently we identified 18 G-protein coupled receptor (GPCR) genes of Daphnia pulex, an invertebrate model organism. Phylogenetic analysis identified these genes to be the homologs of the human γ-aminobutyric acid, type B (GABAB) receptor, metabotropic glutamate receptors (mGluR), adrenergic receptor, serotonin (5-HT) receptor, dopamine receptor and muscarinic acetylcholine receptor (mAChR). Using reverse transcription and quantitative PCR techniques, we systematically measured the effects of propofol, etomidate and ethanol on these 18 GPCR mRNA expressions in Daphnia pulex.

10.Effects of continuous passive motion on early healing of rabbit rotator cuff bone-tendon junction:MRI verification at varied time points

BACKGROUND: Supraspinatus tendon injury is common in rotator cuff injury, the repair difficulty of which is reconstructing the bone-tendon interface. Currently, there are no effective therapeutics and suitable experimental animal models.OBJECTIVE: To establish rabbit rotator cuff injury prosthetic experimental animal model and to observe the effect of continuous passive motion (CPM) on early healing of rabbit rotator cuff bone-tendon interface by MRI, which can provide guidance for preparing an optimal rehabilitation strategy after rotator cuff injury.METHODS: Sixteen male New Zealand rabbits, aged 8 months, were received a rotator cuff acute injury and reconstructing insertion of supraspinatus tendon on greater tuberosity of humerus. The rabbits were randomly divided into cage activity group (n=4) and CPM group (n=12). At 2 days after operation, rabbits were treated by rabbit shoulder joint continuous passive motion apparatus of 75°~75° flexion-extension with various CPM speeds[2 (°)/s, 4 (°)/s, and 10 (°)/s)]. General state and MRI changes of rabbit rotator cuff bone-tendon interface was observed prior to and at weeks 1,2, and 4 after operation.RESULTS AND CONCLUSION: Gross observation showed that the incisions healed very well at 2 weeks after operation. There was no infection, haematoma or disruption after removing the suture. Compared to the cage activity group, the tendon-bone interface of CPM group heeled better at weeks 1, 2 and 4, especially at the speed of 4°/s with 75°-75° flexion-extension. Results demonstrated that this animal model can be satisfied to experimental study. From the following day after operation, various CPM speeds can accelerate the healing of tendon-bone interface, and the optimal speed of CPM was 4 (°)/s.
