1.Influence of skin-derived mesenchymal stem cells from psoriasis lesions on T cell proliferation

BACKGROUND:Studies have found abnormalities in the biological activity and cytokine levels of bone marrow mesenchymal stem cels from psoriasis patients, while the biological activity of skin-derived mesenchymal stem cels from psoriasis lesions is rarely reported.
OBJECTIVE:To explore the influences of skin-derived mesenchymal stem cels from psoriasis lesions on T cel proliferation
METHODS:After isolation and culture, skin-derived mesenchymal stem cels from psoriasis lesions and normal controls were separately co-cultured with peripheral blood T cels. T cel proliferation was detected using MTT assay,andlevels of epidermal growth factor and transforming growth factor β1 in cel supernatant determined using ELISA method.
RESULTS AND CONCLUSION:Skin-derived mesenchymal stem cels frombothpsoriasis lesions and normal controls significantly inhibited T cel proliferation (P< 0.05). Furthermore, skin-derived mesenchymal stem cels from psoriasis lesions exhibited a weaker inhibitory effect on T cel proliferation than normal skin-derived mesenchymal stem cels (P< 0.05). Skin-derived mesenchymal stem cels from psoriasis lesions significantly increased epidermal growth factor but reduced transforming growth factor β1 compared with the normal skin-derived mesenchymal stem cels (P< 0.05). In conclusion, maybe the imbalances of growth factor levels due to skin injury elicit a decrease in the inhibitory effect of skin-derived mesenchymal stem cels from psoriasis lesions on T lymphocytes.
2.Expressions of T-bet and GATA-3 mRNA in peripheral blood mononuclear cells of patients with pityriasis rosea

Objective To investigate the significance of T-bet and GATA-3 in the pathogenesis of pityriasis rosea.Methods SYBR Green Ⅰ real-time fluorescence quantitative PCR was performed to detect the expressions of T-bet and GATA-3 mRNA in peripheral blood mononuclear cells (PBMCs) from 17 patients with pityriasis rosea and 20 normal human controls.Results The average delta Ct value of T-bet mRNA and GATA-3 mRNA was 3.33 ± 0.94 and 5.22 ± 0.69 respectively in the patients,4.31 ± 1.11 and 4.36 ± 1.02respectively in the controls.There was a higher expression of T-bet mRNA but a lower expression of GATA-3 mRNA in the patients compared with the controls (both P < 0.05).Conclusions Patients with pityriasis rosea show an increased ratio of T-bet to GATA3 expression and predominant expression of Th1-type cytokines.Abnormal cellular immunity may play an important role in the pathogenesis of pityriasis rosea.
3.Evaluation of Three Dimensional Electro-anatomical Mapping System in Treating the Patients With Paroxysmal Supra-ventricular Tachycardia by Radio Frequency Catheter Ablation

Objective: To explore the feasibility and safety of three dimensional (3D) electro-anatomical mapping system (Carto3) in treating the patients with paroxysmal supra-ventricular tachycardia (PSVT) by radiofrequency catheter ablation (RCFA).
Methods: A total of 180 PSVT patients were divided into 2 groups, n=90 in each group. 3-D group, the patients received RCFA with 3-D reconstructed valve ring model under Carto3 guidance. 2-D group, the patients received RCFA under conventional X-ray guidance. The procedural and X-ray exposure times, rates of success and complications, tachycardia recurrence at 6 months after procedure and the cost were observed and compared between 2 group.
Results: The procedural time was similar between 2 groups, P=0.1403. The patients in 3-D group had the lower X-ray exposure time (2.1 ± 0.7 vs 7.8 ± 3.6) min, particularly in those with right-sided accessory pathway (3.4 ± 0.7 vs 20.2 ± 7.1) min, and dual atrio-ventricular (A-V) nodal pathways (1.1 ± 0.3 vs 5.5 ±1.7) min, all P<0.0001. There was 1 patient in 3-D group without RCFA and all the others were successes. 2-D group had 3 patients with failed RFCA including 2 of right-sided accessory pathway, 1 of dual A-V nodal pathways and received 2nd RCFA under Carto3 guidance. 3-D group had no complication, no recurrence. In 2-D group, 1 patient suffered from complete A-V block (AVB) during ablation and 1-year later, the Holter showed II° to III° AVB;2 patients with recurrence including 1 of dual A-V nodal pathways and had successful 2nd ablation. The cost was higher in 3-D treatment.
Conclusion: RFCA was feasible for treating PSVT patients under Carto3 guidance, which had the higher success rate with lower X-ray exposure and complication.
4.Evaluation of the effect of modified method of fixation and withdrawal of needles in intravenous infusion

Objective To evaluate the effects of modified method of fixation and withdrawal of needles in intravenous infusion.Methods The modified method of two-step fixation and withdrawal of needles was adopted in emergency transfusion room of our hospital from September 2013.In January 2015,during every day's low peak period 12:00-14:00,246 emergency transfusion patients were chosen and divided into the traditional group (122 cases,using three-step fixation and withdrawal of needles) and the modified group (124 cases,using two-step fixation and withdrawal of needles) according to transfusion order.The pain degree caused by withdrawal of needles using Numeric Rating Scale (NRS),time consumption of fixation and withdrawal of needles and rate of adhesive pastes abscission was assessed by nurses and compared between two groups.Results The incidence of pain in the modified group was lower than that of the traditional group [48.39% (60/124) vs.81.97% (100/122)],x2=30.49,P<0.05.The time consumption of fixation and withdrawal of needles in the modified group was shorter than that of the control group [(7.55 ±2.01) seconds vs.(10.88 ±2.72) seconds;(2.44 ±0.84) seconds vs.(11.55 ± 4.62) seconds],Z=8.70,13.55,P<0.05.Therate of adhesive pastes abscission in the modified group was lower than that of the control group [4.0%(5/124) vs.18.9% (23/122)],x2 =13.39,P<0.05.All the difference between two groups was statistically significant.Conclusions The modified method of fixation and withdrawal of needles can relieve the pain caused by withdrawal of needles.Nurses can operate easily,adhesive pastes is fixed sturdily,which is popular among nurses and patients.
5.Establishment of multicultural nursing health education program for coronary disease patients based on Delphi technique

Objective To establish multicultural nursing health education program for coronary disease patients based on Delphi technique. Methods By referring to document literatures, conducting semi-structured interviews and clinical pre-survey on patients, the frame contents of multicultural nursing health education program for coronary disease patients were preliminarily established. Delphi technique was used two-round consultation of 17 experts. Results After the two-round consultation, the active coefficient of experts was 100%, and the degree of specialist authority was 0. 840 ( >0. 7). It was finally determined that in the multicultural nursing health education program, 10 first-class indexes, 34 second-class indexes and 96 third-class indexes were included. Conclusions The research result is reliable and scientific. The program established is innovative and practicable, which may provide basis and references for performing health education that is both individualized and consistent with the proper culture for coronary disease patients.
6.Correlation analysis of self-efficacy and quality of life among patients after coronary artery bypass grafting

Objective To address the correlation between self-efficacy and quality of life in patients after coronary artery bypass graft. Methods A total of 88 patients in our hospital were investigated by using general data questionnaire, Barnason self-efficacy scale and concise health questionnaire ( MOS-SF 36 ) at 2 weeks, 6 weeks and 3 months after coronary artery bypass graft. Results The self-efficacy scores were (42. 8 ± 5. 4) at 2 weeks after operation, (44. 5 ± 4. 2) at 6 weeks after operation and (47. 6 ± 5. 6) at 3 months after operation, and the scores of quality of life were (73. 7 ± 11. 4) at 2 weeks after operation, (85. 9 ± 10. 6) at 6 weeks afteroperationand(93.9±16.2)at3monthsafteroperation(F=3.452,P<0.05). Afteroperation,theself-efficacy increase gradually, and the difference was statistically significant. The scores of quality of life were (73. 7 ± 11. 4) at 2 weeks, (85. 9 ± 10. 6) at 6 weeks, and (93. 9 ± 16. 2) at 3 months after operation, and there was a positive correlation between self-efficacy and quality of life ( r=0. 411, 0. 447, 0. 307,respectively;P<0. 01). Conclusions The quality of life and self-efficacy of patients after coronary artery bypass surgery are improved, and the two are positively correlated. Nursing staff should be based on principles of self-efficacy theory to strengthen the nursing work which may improve the patient′s self-efficacy and the quality of life.
7.Effects of motivational interview on self-efficacy and health related behaviors in patients after coronary artery bypass grafting

Objective To discuss the impacts of motivational interviewing health education on the self-efficacy and the impacts of health-related behaviors in patients after coronary artery bypass grafting (CABG). Methods A total of 84 patients after the first CABG were selected in Department of Cardiac Surgery of Shanghai East Hospital. These patients were divided into 42 cases of intervention group and 42 cases of control group. Conventional methods of health education were used in the control group, and the patients of intervention group received motivational interview plans. After 6 months, self-efficacy and healthy behavior were evaluated in these patients. Results Before the intervention, the scores of self-efficacy and health related behavior′s difference were not statistically significant (P>0. 05); after 6 months intervention, the score of self-efficacy, diet, medication management, movement, stress coping and health related behavior score respectively (47. 5 ± 6. 3), (33. 5 ± 3. 3), (39. 2 ± 4. 3), (14. 0 ± 4. 2), (15. 7 ± 3. 8) (93. 1 ± 6. 2) in the intervention group were higher than that of the control group, except exercise and stress coping dimensions, there were significant differences between the two groups (P <0. 05). Conclusions Motivational interview health education can increase the self-efficacy and health-related behaviors in patients after CABG.
8.Effect of self-management education based on nurse-leading on the quality of life in patients with chronic heart failure

Objective To evaluate the effect of self-management education based on nurse-leading on the quality of life in patients with chronic heart failure (CHF).Methods Eighty patients with CHF in a tertiary hospital were enrolled in this study,and were divided into the intervention group and the control group randomly,with 40 cases in each group. Patients in the control group received conventional health education, while patients in the intervention group received the self-management education based on nurse-leading. The score of self-management behavior and the quality of life in two groups were evaluated and compared after six months.Results After six months,the score of self-management in the intervention group was (70.83±12.02), and was significantly higher than that in the control group (55.58±6.46) (t=9.675,P<0.05). The score of total quality of life and self-management in the intervention group was significantly higher than that in the control group (P<0.05).Conclusions Self-management education based on nurse-leading can improve the self-management and the quality of life of patients with CHF.
9.Generation of MCM2 gene inducible knockout cervical cancer HeLa cells and its effect on DNA replication

Objective To generate minichromosome maintenance protein 2(MCM2)gene knockout cervi-cal cancer HeLa cell lines using inducible CRISPR/Cas9 technology,and to explore the effect of MCM2 on DNA replication and replication stress.Methods The inducible CRISPR/Cas9 system,TLCV2,was used to construct MCM2 knockout HeLa cell lines.And the cell lines were divided into control group(Control),knockout group 1(KO1),and knockout group 2(KO2).Western blot,Edu incorporation experiment,real-time quantitative PCR(qPCR),immunofluorescence and MTT assay were used to analyze the effects of MCM2 knockout on DNA replica-tion and replication stress induced by hydroxyurea.Results The CRISPR/Cas9 system successfully knocked out the MCM2 gene after induction,and MCM2 knockout affected the stability of MCM2-7 complex.Compared with the control cells,MCM2 knockout cells had a dramatic decrease in the capacity of DNA replication,and the mRNA levels of Cyclin A1,Cyclin E1 and CDK4.Under DNA replication stress,MCM2 knockout cells decreased cell viability,DNA damage repair capacity,and increased genomic instability compared with control cells.Conclusion Knockout of MCM2 gene reduces the DNA replication capacity of HeLa cells under normal conditions and cell viability under replication stress.This study successfully generates MCM2 gene inducible knockout HeLa cell lines,laying the foundation for further research on the role and biological function of MCM2 gene in the occurrence and progression of cervical cancer.
