1.PIT CELLS IN LIVER AND EXTRAHEPATIC ORGANS AND THEIR SIGNIFICANCE

By electron microscopic observation,we found few pit cells in liver,spleen.lungs,and small intestine in Wistar rat.The pit cells show several morphologicalcharacteristics as follows:1.electron dense granules in cytoplasm,which aresurrounded by a limiting membrane;2.vesicles with rod shaped bodies incytoplasm;3.eccentric nucleus with marginal heterochromatin;4.well developedpseudopodia.In addition,the pit cells have also been found in breast carcinoma in3 patients.According to their distribution and morphology,we preliminarilydeduced they probably belong to natural killer cells and may act as a cytotoxicagent and an immunological defender against carcinoma.
2.A case of asymptomatic process of hepatic artery thrombosis in the patients following orthotopic liver transplantation

Objective To investigate the possible causes for asymptomatic process of hepatic artery thrombosis in the patients following orthotopic liver transplantation (OLT). Methods Hepatic functional tests, color Doppler ultrasonic observation, hepatic arteriographic examinations were done during the course of the development of the hepatic arteriothrombosis. All possible factors leading to the asymptomatic process were comprehensively analyzed. Results At the 4th postoperative day, thrombosis started to form along the anastomosis line. At the 21st postoperative day, the artery became completely thromboses. During this course, serial liver biopsy revealed introhepatic cholestasis, hydropic degeneration of hepatocytes, atrophy of the biliary epithelium and fibrosis in the portal area. Serial monitoring of liver function gradually revealed normal with the exception of elevation of r-GT and ALP. At the 71st day after OLT, arteriography showed that hepatic artery remained completely occluded, while the collateral circulation and compensatory expansion of the portal vein were established. The patient didn't have any symptoms of arterial thrombosis. Conclusions In this case, it was the collateral circulation and the compensation of the portal vein that made the allograft survive and helped to prevent the patient from retransplantation after hepatic artery thrombosis. Color Doppler ultrasonic examinations within 2 weeks following OLT were helpful to the early diagnosis of hepatic arteriothrombosis.
3.Efficacy of transurenthral resection of muscle-invasive bladder cancer in the elderly

Objective To investigate the clinical effects of transurenthral resection of muscleinvasive bladder tumor (TURBT) in senile. Methods This study was clinical prospective random research.46 senile cases with muscle-invasive bladder cancer treated by TURBT was divided into 2 groups:control group and observed group.The observed group was treated with TURBT combined with intravesical instillation of pirarubi 30 mg immediately after TURBT,the bladder wall around tumor was resected thoroughly.The control group underwent radical cystectomy and pelvic lymph node dissection. Results The clinical improvement rate was higher in observed group than control group (85.7% s.63.6%,X2 =5.3008,P=0.021).There was not difference in 3 years recurrence rate between observed group(25.0%) and control group(13.6%) (x2 =S.3008,P>0.05).The 1 year survival rate were 95.8%,90.9% and 3 years survival rate were 79.2%,72.7% in observed group and control group,respectively,and there were no differences between the two groups(X2 =0.262,P=0.609). Conclusions TURBT combined with intravesical instillation of pirarubi can enhance clinical effect and reduce recurrence rate in the treatment of the elderly with muscle-invasive bladder cancer.
4.Formulation Optimization of Propranolol Hydrochloride Gels by Central Composite Design-Response Surface Methodology

Objective To optimize the matrix formula of propranolol hydrochloride gels. Methods On the basis of single factor experiment, central composite design-response surface method was used to optimize the formula. Addition levels of glycerol (A), PEG-400 (B) and HPMC (C) were evaluated as the independent variables.Eight-hour accumulative penetration amount per unit area measured by HPLC was used as the index. Quadratic polynomial was used to estimate the relationship between the index and the independent variables, and to delineate response surface and overlay contour plots in order to select the optimal formulations.Finally, predicted responses were verified. Results The optimized formula consisted of 18.53% glycerol, 8.54% PEG-400 and 2.35% HPMC.The quadratic polynomial regression model of 8-h accumulative penetration amount per unit area:R1=-7 415. 69+306. 10A+167. 47B+4 820. 59C-8. 26A2-9. 81B2-1 025. 75C2 , and the cumulative transmittance was 49.6%. Conclusion A credible model is established by using central composite design-response surface method and the formula of propranolol hydrochloride gels is optimized and the gel is stable and controllable.
5.STUDY ON THE ANTIFATIGUE EFFECT OF TEA CONTAINING ACANTHOPANAX SENTICOSUS

Objective: To study the antifatigue effect of tea containing Acanthopanax senticosus.Methods: Forty eight mice were randomized into 4 groups:three groups of tea containing Acanthopanax senticosus at doses of 50, 75, 100 ml/(kg?d), and the saline group.After 4 w, muscle and liver glycogen, serum LDH activity, BUN and blood LAC were measured.Results: Three tea groups showed increase of muscle and liver glycogen,and serum LDH activity, and decrease of serum BUN and LAC in swimming mice.The 75-100 ml/(kg?d) group was the best.Conclusion: Tea containing Acanthopanax senticosus has antifatigue effect.
6.Stepwise Regression Analysis of Morphologicai Parameters of Cell-Apparatus of Breast Carcinoma Cells in the Late Stage

Cell morphometry method was applied in courting a nd analyzing 12 ultrastruc-ture morphological parameters of the rough endoplasmic reticulum and ribosome from 10 female patients of the simple breast carcinoma in the late stage by pathological diagnosis. The best equation was set up with stepwise regression analysis was as follows: y=0.22+0.503N6-013N7. Due to above equation, selecting the best parameters were the surface density of free ribosome N6 and specific surface of rough endoplasmic reticulum N7. They could be used to distinguish the normal cell-apparatus from carcinoma cell-apparatus. These results show that N6 and N7 in varying degrees could provide fixed quantity basis for the prognosis of breast carcinoma.
7.The stereologic analysis of morphological structure in mid stage and late stage of the simple carcinoma of breast

In this article,stereologic method was applied in analyzing 17 morphological parametera of cell, nucleus and nucleolus in 20 cases of mid stage and late stage of the simple carcinoma of breast (the stylebooks of EM photos were 320). We analyzed the difference of each sort of cell with T test and Q test. Compared with the normal cells, the cancer cells of mid stage were found obviously not only decreased in their nucleus surface density Svn and nucleus contrast surface density Sn but also increased in their nucleus average section An and nucleus average perimeter Bn. Compared with the cancer cells of the mid stage the cancer cell of the late stage were found obviously increased in their nucleus average volume Vn and Bn. Compared with the normal cells, the cancer cells of the late stage were found obviously not only decreased in their Svn and Sn but also increased in their Vn, An, Bn and nucleolus average perimeter Bu.
8.Determination of astragaloside Ⅳ in Qixiu Granules by HPLC-ELSD

AIM: To determine astragaloside Ⅳ in Qixiu Granule (Radix Astragali seu Hedysari, Radix Salviae Miltiorrhizae, Radix Paeoniae Rubra, etc.) by HPLC ELSD. METHODS: HPLC ELSD was used in the quantitative analysis by using Alltima C 18 chromatography column and acetonitril water (34∶66) as a mobile phase. The flow rate of mobile phase was 1mL?min -1 . The tube temperature of the detector was 100?C. The flow rate of pure air was 2.7L?min -1 . RESULTS: The regressione quation was Y=-4.40?105+2.07?105X, r=0.9992 (3.73~13.59?g). The average recovery of astragaloside Ⅳ was 100.73%, RSD was 3.97%. CONCLUSION: The method is simple, reliable, accurate and can be regard as the quantity control method of Qixiu Granule.

OBJECTIVE:To observe the effects of felodipine on blood vessel endothelial function in patients with benign prostatic hyperplasia(BPH) complicating essential hypertension(EH).METHODS:Fifty five patients with BPH complicating EH were assigned to receive 5mg felodipine orally qd for 6 weeks,with levels of endothelin-1(ET-1),nitrous oxide(NO) and von Wilebrand factor(vWF),brachial artery flow-mediated dilatation(FMD) function,the maximum urine flow rate,the average urine flow rate and International Prostate Symptom Score(I-PSS) determined before and after treatment.Meanwhile levels of ET-1,NO and vWF and FMD in 37 normal healthy males were determined.RESULTS:In the treatment group compared with normal control group there were significant differences in levels of ET-1,NO and vWF and FMD(P0.05).There were significant differences in ET-1,NO and vWF levels and FMD after treatment as compared with before treatment(P
10.Transumbilical laparotomy: a cosmetic and minimally invasive operative route in pediatric patients

Objective:The feasibility of a minimally invasive cosmetic transbilical laparotomy was studied in fifty-seven pediatric patients.The advantages of this technique will be discussed.Methods:The circumferential incision was made following the ring fold of the right umblilicus,which was less than 2700.The peritoneum was opened in the midline via rectus abdominis.Intra-peritoneal lesions were detected and explored.The surgical procedures were able to be carried out either inside or outside of the peritoneum.After closure of peritoneum,the umbilical incision was closed subcutaneously with an absorbable suture.Results:From January 2001 to January 2005,58 procedures via transumbilical route have been done in 57 pediatric patients.None of those cases required conversion to a traditional abdominal incision.The duration of surgery was from 20 to 170 minutes.All patients were discharged five days postoperatively and follow-up was conducted from 3 months to 4 years.No complications were observed in all but one patient.This patient had a leaking anastamosis.No visible scars were present and the cosmetic results were excellent.All patients had shorter hospital stay and lower operating cost.Conclusion:Transumbilical approach is a feasible,safe,economical,minimally invasive procedure with excellent cosmetic results which appears to be superior to traditional procedures.It suggests that the transumbilical route should be widely used clinically and might be applicable for elder children and possibly would be of benefit in the treatment of other conditions.
