1.Effect of neck rotation and lateral bending on the blood flow of the vertebral arteries

Objective To study the effect of the neck rotation and lateral bending on the vertebral artery blood flow, and to analyze the relationship between the artery blood flow and cervical vertigo. Methods Thirty patients with cervical vertigo were studied with exclusion of auditory vertigo and severe cardio-cerebrovascular disease, which involved 18 males and 12 females, whose average age was 52.13 years with a range of 42-73 years. They all had cervical degenerative change in different degree and neck-shoulder-back pain history. The study also included 15 volunteers without vertigo, 8 males and 7 females, the average age was 49.47 years with range, 38-70 years, they also had some cervical degenerative change. They were selected to detect the blood flow of bilateral intracranial segments of the vertebral arteries using TCD with the neck in neutral, bilateral rotation 15?,30?,45?,60? and bilateral bending. 2.REGULATION OF MMP_(13) GENE EXPRESSION IN RAT HEPATIC STELLATE CELLS BY IL-1? AND TNF-?

In order to investigate the influence of interleukin 1?(IL 1?) and tumor necrosis factor ? (TNF ?) on gene expression of rat interstitial collagenase (MMP 13 ) in hepatic stellate cells, cells of a hepatic stellate cell line (HSC T6) were cultured in medium containing IL 1? or TNF ? for different incubation periods. Total RNA of HSC was extracted and MMP 13 gene expression levels were measured by reverse transcription polymerase chain reaction. The results showed that MMP 13 gene expression level of the IL 1? group was much higher than that of the control group at 8h, 24h and 48h after exposure to IL 1?. The maximal value was 3 fold of the control group and was at 24h . MMP 13 gene expression level of the TNF ? group was also significantly higher than that of control group at 8h, 24h, 48h, 72h, and the maximum nalue was 4 fold of the control group at 48h. These results indicated that IL 1? and TNF ? might increase the gene expression of MMP 13 in hepatic stellate cells.
3.Medication Monitoring of Anti-infection Drugs in a Patient with Lupus Nephritis Complicated with Com-munity Acquired Pneumonia

Objective:To explore the value of clinical pharmacists in the anti-infection practice for a patient with lupus nephritis complicated with community-acquired pneumonia(severe). Methods:Clinical pharmacists carried out medication monitoring in the entire treatment process of a patient with lupus nephritis complicated with community-acquired pneumonia( severe)in the clinical front line. Depending on the pharmaceutical knowledge,clinical pharmacists assisted physicians to make out the optimal medication and ad-justed the medication according to the patient′s conditions. Results:Moxifloxacin and piperacillin sodium /tazobactam sodium could ef-fectively control the severe pulmonary infection of the patient with lupus nephritis and avoid the potential risk of recurrence or aggrava-tion of lupus nephritis. Conclusion:The choice of antimicrobial agents for the special populations should consider not only the antibac-terial spectrum and antibacterial activity,but also the individual characteristics of patients and pharmacokinetics of drugs in order to im-prove the safety and effectiveness of medication.
4.Effect of Pulse Microwave Combined with Ofloxoacin on Chronic Prostatitis

Objective To observe the effect of pulse microwave combined with antibiotics on chronic prostatitis.Methods100 outpatients with chronic prostatitis were randomly divided into the trail group and control group with 50 cases in each group. The patients of the trail group were treated with pulse microwave combined with Ofloxoacin, and those of the control group were treated only with Ofloxoacin. The treatment course was 5 weeks. The evaluation with National Institutes of Health-Chronic Prostatitis Symptom Index (NIH-CPSI) and test of white blood cell (WBC) count in prostate secretion were performed before and after treatment for all patients of two groups.ResultsIn the trail group, the mean symptom degree score of NIH-CPSI was 9.3±2.6 and living quality score was 3.4±1.2, the total score was 12.4±4.1, and WBC count was 13.7±5.4/HP. In the control group, these indexes were 13.3±3.5, 4.5±1.2, 17.5±4.6 and 17.1±5.2 respectively. The NIH-CPSI score and WBC count of the trail group were lower than that of the control group ( P<0.05), and cure rate and total effective rate were higher than that of the control group ( P<0.05).ConclusionThe treatment of pulse microwave combined with Ofloxoacin for chronic prostatitis can improve the cure ratio and efficiency.
5.Relationship of peritoneal transport patterns and the nutritional status in continuous ambulatory peritoneal dialysis patients

Objective To explore the relationship of peritoneal transport patterns and the nutritional status in continuous ambu-latory peritoneal dialysis(CAPD) patients .Methods 89 patients who took CAPD in 2010 January to December 2012 in the perito-neal dialysis center of our hospital were selected in this study .According to the ratio of creatinine in peritoneal dialysis solution and in blood(D/Pcr)of the peritoneal equilibrium test(PET) ,all CAPD patients were divided into HA group(high average transport) and LA group(low average transport) .Nutritional status including serum albumin(ALB) ,prealbumin(PA) ,transferrin(TF) ,hemo-globin(Hb) ,Lean body mass(LBM) ,subjective global assessment(SGA)and normalized Protein Equivalent of Nitrogen Appearance (nPNA) were compared in both groups .The relationship of D/Pcr and the nutritional status were analyzed .Results D/Pcr in HA group were significantly higher than in LA group(P<0 .01) .ALB ,PA ,TF ,Hb and SGA in HA group were significantly higher than in LA group(P<0 .01) ,but nPNA in two groups has no statistical differences (P>0 .05) .D/Pcr was negative related to ALB , TF ,Hb and SGA(P<0 .01) ,but not related to nPNA ,PA and LBM (P>0 .05) .Conclusion The nutritional status is related to peritoneal transport patterns in peritoneal HA patients ,it is necessary to strengthen nutritional guidance ,as the nutritional status is worse compared to LA ones .
6.Risk factors analysis of sudden death in patients suspected with pulmonary thromboembolism in emergency room

Objective To explore the correlative factors of sudden death in patients suspected with pulmonary thromboembolism (PTE) in emergency room (ER).Methods A retrospective analysis was conducted.The clinical data of 12 patients with sudden death suspected with PTE (sudden death group) in ER of the Air Force General Hospital from January 2011 to June 2014 were analyzed.The non-sudden death group included 35 patients during the same time period who were diagnosed with PTE based on findings of CT pulmonary arteriography (CTPA) and showed no sudden death in ER.Factors,including sex,age,previous operation,tumor,syncope,dyspnea,bilateral or unilateral edema of lower extremity,heart rate (HR),white blood cell count (WBC),D-dimer,arterial partial pressure of oxygen (PaO2),arterial partial pressure of carbon dioxide (PaCO2) and typical clinical manifestation of electrocardiogram (SⅠTⅢQⅢ),were compared between the two groups.The potential predictors of sudden death of PTE were analyzed by logistic regression analysis.Results Young age (years old:51.3±15.5 vs.62.3±14.4),lower PaO2 [mmHg (1 mmHg =0.133 kPa):49.9± 12.3 vs.62.7± 10.2],higher HR (bpm:122.0± 19.5 vs.89.1 ± 18.5) and higher WBC (× 109/L:13.8 ± 6.9 vs.7.2 ± 2.5) were found in sudden death group as compared with those in non-sudden death group (P < 0.05 or P < 0.01).There was no significant differences in D-dimer level and PaCO2 between sudden death group and non-sudden death group [D-dimer (pg/L):986 (891,3 230) vs.2089 (598,3 397),PaCO2 (mmHg):33.0 (28.6,43.4)vs.36.5 (32.9,41.0),both P > 0.05].The syncope,antineoplaston treatment or tumor metastasis within 6 months,operation in previous 4 months,bilateral asymmetrical edema in sudden death group were more than those of the non-sudden death group,and chest pain was less (P < 0.05 or P < 0.01).Difference in gender,dyspnea and typical SⅠTⅢQⅢ in electrocardiogram were not significant between the two groups (all P > 0.05).It was shown by multiple logistic regression analysis that higher HR [odds ratio (OR) =1.124,95% confidence interval (95%CI) =1.024-1.235,P =0.014] and higher WBC (OR =1.347,95%CI =1.043-1.738,P =0.022) were identified as independent risk factors of sudden death for PTE.Conclusions Gender,dyspnea,typical S Ⅰ TⅢQⅢ in electrocardiogram,PaCO2 and D-dimer seem unrelated to sudden death of patients with PTE.Young age,chest pain,syncope,bilateral asymmetrical edema,antineoplaston treatment or tumor metastasis within 6 months,operation in previous 4 months and low PaO2 were potential predictors of sudden death according to the univariate analysis.Higher WBC and higher HR are independent risk factors of sudden death for PTE patients.
7.L-hydroxyproline changes in rats' skin induced by ultraviolet C irradiation

ObjectiveTo investigate the L-hydroxyproline (L-Hyp) changes in rats' skin after exposed to different doses of ultraviolet C irradiation (UVC) and the effect of UVC radiation on collagen synthesis.MethodsAfter the animal model was set up, each male Wistar rat was made three fresh skin wounds, and three skin wounds of rats were divided into the 15 MED UVC irradiation (15mJ/cm2),60 MED UVC irradiation (60mJ/cm2) and control (without UVC irradiation). Then the chemistry method was utilized to research changes of L-Hyp contents of the granulation.ResultsFrom the 21st to the 28th day after UVC irradiation, contents of L-Hyp in the 15 MED group were higher than that in controls (P<0.05). While at the 28th day, L-Hyp level in the 60 MED group increased greatly and was higher than that in other two groups (P<0.01).ConclusionUVC irradiation increases L-Hyp level in rat's wound skin, so it accelerates the collagen synthesis and is helpful to promote wound healing, and the effect of 60 MED dose is superior to that of 15 MED.
8.Construction and identification of the RNA interference eukaryotic expression vector targeting the CD40 gene in rats

Objective: To construct and identify the eukaryotic expression vector targeting the CD40 gene in rats.Methods: Two sequences corresponding to the rat CD40 gene were designed on Ambion's Web.The two complementary oligonucleotide strands of DNA fragments were synthesized by chemosynthesis.After annealing of the complementary strands,the DNA fragments were connected to the polyclone sites of the pSilencer 4.1-CMV neo vector,followed by transformation,amplification,plasmid extraction and identification of the recombinant plasmids by BamH Ⅰ and Hind Ⅲ digestion and DNA sequence analysis.Results: The connections between the DNA fragments encoding CD40-targeted siRNA and the pSilencer 4.1-CMV neo vector were correct,as confirmed by agarose gel electrophoresis and DNA sequence analysis.Conclusion: The two recombinant plasmids expressing siRNA of the rat CD40 gene were successfully constructed,which prepared the groundwork for future research on the role of the CD40 gene in homograft rejection.
9.The VEGF production by dedifferentiated chondrocytes under synovial fluid stimulation from coxarthrosis and femoral neck fracture patients

Objective To investigate the vascular endothelial growth factor (VEGF) expression level by chondrocytes isolated from patients with osteoarthritis (OA) in hip or femoral neck fracture (FNF) and explore the effect of synovial fluid from OA or FNF on secretion of VEGF. Methods The cartilage tissues were collected from 12 patients with OA in hip and 8 patients with FNF. Cartilage was stained with HIM and Safranin O/Fast Green (S/F) method. The damage of cartilage was evaluated using Mankin scores.Cathepsin B which was selected for cell dedifferentiation monitoring marker and VEGF level was detected in the supernatant fluid. The synovial fluid from OA, FNF and DMEM were respectively added to the culture medium to explore their effects on regulating VEGF. Results Cartilage the Mankin scores of OA group were higher than that of FNF group. Chondrocytes gradually lost their original spherical appearance, with Cathepsin B upregulated while VEGF downregulated. The OA synovial fluid can stimulate chongdrocytes to secrete more VEGF than the one from patients with FNF. However, chondrocytes gradually produced less VEGF after passaging. Conclusion Mankin scores had good correlation with chondrocytes' VEGF production in the early stage of primary culture. Chondrocytes showed quick dedifferentiation characteristics in vitro. OA synovial fluid showed abig ger capability in stimulating chondrocytes to express more VEGF, which might indicate that OA synovial fluid participated in the pathological process of OA.
10.Optimal condition for preservation of three Achromobacter xylosoxidans bacteriophages

Objective To find simple and effective methods of preservation for Achromobacter xylosoxidans bacteriophages.Methods The plaque forming unit( PFU) of surviving phages under different preservation conditions and temperatures at different time points was determined.Results and Conclusion The titers of phiAxp-1 and phiAxp-2 main-tained at the initial 1010 PFU/ml, and that of phiAxp-3 decreased slightly from 1011 PFU/ml to 1010 PFU/ml 16 months after the three Achromobacter xylosoxidans bacteriophages were stored in glycerol and dimethyl sulfoxide at -80℃, -20℃and 4℃.When stored in chloroform at 4℃for 16 months, the titers of all the three phages decreased slightly but were higher than at other temperatures (-80℃, -20℃, room temperature, and 37℃) .Thus these methods can effectively preserve Achromobacter xylosoxidans bacteriophages.
