1.Analysis on the detection of Chlamydia trachomatis infection in 3 199 pregnant women

Objective To investigate the status of Chlamydia trachomatis(CT) infection in pregnant women in Wuxi ,and to ex‐plore the relationship between CT and abortion .Methods A total of 3 199 cases of pregnant women visiting this hospital from Jan‐uary 2013 to December 2014 were enrolled in this study and divided into the missed abortion group(2 633 cases) ,incomplete abor‐tion group(265 cases) ,the inevitable abortion group(137 cases) ,threatened abortion group(104 cases) and control group(60 cases , all women undergoing induced abortion) .The CT infection was detected by using the fluorescent quantitative polymerase chain reac‐tion(FQ‐PCR) .Results The positive rate of CT in missed abortion group ,incomplete abortion group ,the inevitable abortion group and threatened abortion group were 5 .62% ,4 .53% ,5 .11% and 4 .81% ,respectively ,all were higher than that in the control group (3 .33% ) ,without statistically significant differences (P>0 .05) .The positive rate of CT in women in 16- <21 years old age group and 21- <26 years old age group was higher than that in other age groups ,which was 7 .06% and 6 .20% respectively .However , there were no statistically significant differences in positive rate of CT between these age groups (P>0 .05) .Conclusion The posi‐tive rate in women undergoing spontaneous abortion is higher than that in women undergoing induced abortion ,and young women has a higher infection rate of CT .
2.EnCor vacuum assisted minimally invasive rotary cutting technology in the ultrasonic BIRADS 2~3 level analysis of diagnosis and treatment of breast diseases

Objective To study the EnCor vacuum negative pressure rotary system in ultrasonic BI-RADS 2~3 scale mammary gland disease application value.Methods Under the guidance of ultrasound, the application EnCor vacuum negative pressure rotary cutting system for 142 patients, 366 cases of breast lesion resection, specimens sent to pathological examination.Results A total of 366 breast lesions, 359 complete resection, the success rate was 98.1%, 2 transit operation, and transfer rate was 0.5%, 5 partial nephrectomy, residual rate was 1.4%.359 complete resection of the pathological results for breast malignant lesions in 4, the infiltrating ductal carcinoma 1, 3 ductal carcinoma in situ;breast benign lesions in 355, of which 132 breast fibroadenoma, adenosis with adenoma formation of 60, 99 breast adenosis, breast adenosis with duct expansion 43, 11 breast intraductal papilloma, one with four individual catheter atypical increase the living.Two transit operation pathological results were fibroadenoma, 5 disease of partial nephrectomy for adenosis with duct expansion.Conclusions The excision of breast neoplasm EnCor vacuum negative pres-sure rotary cutting system, not only small trauma, good cosmetic effect, and reduce misdiagnosis and im-prove the patients'satisfaction.
3.Present Situation Investigation of Trace Elements of 0~7 Years Old Children in Shenzhen District

Objective To understand the Shenzhen Dapeng new district and Longhua district 0~7 years old children in copper (Cu),zinc (Zn),calcium (Ca),magnesium (Mg),iron (Fc) and lead (Pb) 6 kinds of trace elements,in order to establish the reasonable dietary structure and provide a scientific basis for scientific and rational supplement of trace elements.Methods Collected 2 190 cases of 0~7 years old children for health physical examination in Dapeng New District Maternal and Child Health Care Hospital of Shenzhen form March to November 2015.According to the age,they were divided into the baby group (0~1 years old) of 637 cases,children group (2~4 years old) of 723 cases and preschool group (4~7 years old) of 830 cases of three groups.Used QL8000 automatic whole blood trace elements analyzer to test Fe,Zn,Ca,Cu,Mg and Pb 6 kinds of trace elements in whole blood,and the detection results were statistically analyzed and compared.Results Test results of 5 kinds of whole blood trace element(Fe,Zn,Ca,Cu,Mg) had different degrees lack in 2 190 cases of children aged 0 ~7,and lack of different level,lack of rate were 36.26%,32.83%,14.20%,0.64% and 0.50%,respectively.Fe and Ca deficiency in young children group was most serious,lack of Zn in infant group was most serious,other microelement lack rate had no statistically significant difference between different groups (P> 0.05).Pb poisoning rate was 15.71%,the Pb poisoning rate decreases along with the age growth,and boys than girls,the difference was statistically significant (x2=3.15,P <0.05),different gender children other microelement lack rate differences had no statistical significance between the results.Conclusion The whole blood trace element of 0~7 years old children had different lack,mainly by Fe,Zn and Ca deficiency in Shenzhen Dapeng New District and Longhua District.But Children's Pb poisoning problem could not be ignored.Therefore,to streng then the 0~ 7 years old children's whole blood trace elements level survey,microelement lack of prevention and treatment of children is of great significance.
4.Effects of anisodamine on the pressure of rats portal vein of liver fibrosis

Aim To investigate the effects of anisodamine on the pressure of portal vein of experimental liver fibrosis and its mechanisms of action. Methods The experimental liver fibrosis model was produced by CCl_4. The preventive group was treated ten weeks with anisodamine 7.0 mg?kg~(-1) ip once everyday. All therapeutic groups were treated six weeks with anisodamine 7.0 mg?kg~(-1) or 14.0mg?kg~(-1) ip once everyday. After CCl_4 injection for ten weeks, the pressure of portal vein,the content of NO in livers, and liver iNOS and eNOS mRNA expressions were detected with different methods.Results The pressure of portal vein was significantly reduced in anisodamine preventive group and anisodamine therapeutic groups. The liver content of NO and the expression of iNOS and eNOS were all inhibited by the treatment of anisodamine.Conclusion Anisodamine reduced the expressions of iNOS and eNOS to synthesis of NO in liver. As a result, the pressure of portal vein of fibrosis rats decreased. So Portal hypertension of liver fibrosis may be improved by anisodamine in patients.
5.Research in correlation between quality of life and acceptance of disability of colostomy patients

Objective To explore the relationship between quality of life and acceptance of disability of colostomy patients.Methods Using convenience sampling method to investigate 111 colostomy patients.General information questionnaire,QLQ-C30 scale,QLQ-CR38 scale and Acceptance of Disability Scale were used to investigate patients' general condition,quality of life and acceptance of disability.Data was analyzed by SPSS 17.0.Results The general health condition of colostomy patients was better than reference value and the score of ADS was at an average level.There was a relationship between general health condition,functioning dimensions,symptom dimensions and acceptance of disability.Conclusions There was a close relationship between quality of life and acceptance of disability.The acceptance of disability should be improved to help patients to obtain better quality of life.
6.Zebrafish as a model animal for the study of blood-brain barrier permeability by biomolecules.

Blood-brain barrier (BBB) is the major obstacle for drug delivery into the central nervous system (CNS). However, there is no ideal model animal for the study of BBB permeability till now. Currently zebrafish (Danio rerio) has emerged as a powerful model organism for the study of vertebrate biology. In this study, the feasibility of using zebrafish as model animal was investigated for BBB permeability by comparing the results of administration of BBB-penetrating peptide and protein to mouse and zebrafish. The results showed that the BBBs of mouse and zebrafish were similar in molecular permeability. Additionally, zebrafish has advantageous features as a model animal, such as small size, fertile and easy to breed. Therefore, it is suggested that zebrafish may be a favored model for the study of BBB permeability.
7.Effects of PDCA method on improving the accuracy of the Autar Deep Venous Thrombosis Scale

Objective:To investigate the effects of PDCA method on improving the accuracy of the Autar Deep Venous Thrombosis (DVT) Scale used by nurses to assess the risk of deep venous thrombosis in surgical patients.Methods:Applied the way of PDCA, namely, raising questions, analyzing reasons, implementing measures, feeding back effects to professional explanate the difficulty in using the scale, moreover, training the nurses used level education method between August 2017 and December2018.The accuracy and consistency of the scale were compared before and after interventions.Results:A total 396 patients were evaluated in the study. There were significant differences in the consistency of evaluation among hospitalized, post-operative and discharged patients after intervention compared with before intervention ( P< 0.0167).The consistency of assessment of team members and responsible nurses increased from 68 to 120 cases, and the consistency of assessment reached 90.91%. The Kappa consistency of assessment of responsible nurses and team members increased from 0.354 before intervention to 0.879 after intervention. The effect was remarkable. Conclusions:PDCA cycle method is based on the problems found in clinical practice. It clarifies the professional terms and rules used in the scale, improves the accuracy and consistency of the Autar DVT scale used by nurses, correctly reflects the risk of thrombosis, ensures the safety of patients, and is worthy of promotion and reference.
8.Construction of the eukaryotic expression vector PsecTaq2A-AMG for human amelogenin.

OBJECTIVEThe purpose of this study was to construct a eukaryotic expression vector for human amelogenin (AMG).
METHODSPCR was performed to amplify the AMG encoding region. Amplified fragments for human AMG were recovered and inserted into eukaryotic expression vectors PsecTaq2A. The recombinant plasmid PsecTaq2A-AMG was constructed and their positive clones were identified.
RESULTS1. Amplified products were checked by electrophoresis and the results were satisfactory. 2. The recombinant plasmid PsecTaq2A-AMG was analyzed by restriction endonuclease mapping and DNA sequencing. The results of sequencing were consistent with those from GenBank.
CONCLUSIONThe recombinant plasmid PsecTaq2A-AMG was successfully constructed with properly inserted DNA sequence encoding mature amelogenin.

9.Extraction and purification of porcine amelogenin and preparation for the polyclonal amelogenin antibody.

OBJECTIVETo prepare the polyclonal antibody to amelogenin.
METHODSThe fetal porcine dental enamel was collected. Enamel matrix protein was extracted in 4M guanidine HCl (pH 7.4) with protease inhibitors present. Polyacrylamide gel filtration was included to isolate amelogenin from the initial dissociated extraction. The purified amelogenin conjugated with or without complete/incomplete Freund's adjuvant was then used to immunize the rabbits subcutaneously or intravenously. The specific IgG antibody was further purified by DE-52 cellulose. The working concentration of IgG antibody was determined through ELISA test.
RESULTSThe Gel filtration showed that amelogenin components is at molecular weights of 15 kD and 13 kD apparently, which was consistent with those described before. The ELISA results showed that the working concentration for IgG was 1:1000.
CONCLUSIONThe antibody prepared in this study can be used for the detection of amelogenin.

10.Efficient evaluation of standardized specific immunotherapy in personalized maintenance phase to patients of allergic rhinitis.

OBJECTIVE:
To explore the efficacy of individualized subcutaneous immunotherapy (SCIT) in allergic rhinitis(AR) maintain phase.
METHOD:
Compare nasal symptom scores (VAS) and special disease scale--nasal conjunctivitis quality of life questionnaire(RQLQ) score after 3 years treatment to evaluate the therapeutic effect of each group and the level of improving patients quality of life. Take patients' blood to detect the serum level of IL-10 by enzyme linked immunosorbent test (enzyme linked immunosorbent assay).
RESULT:
After 3 years treatment, there was no difference of VAS between the conventional SCIT group and the individualized SCIT group. ELISA results showed that the level of IL-10 was significantly higher in the drug symptomatic treatment group than that in the healthy group, the levels of IL-10 were significantly lower in the conventional SCIT group and the individualized SCIT group than that in the healthy group, but there was no difference between the conventional SCIT and the individualized SCIT group.
CONCLUSION
After 3 years treatment, there was no difference between conventional and individualized SCIT groups. But the efficacy of the conventional and individualized SCIT groups were significantly better than that in the drug symptomatic treatment group.
