1.The separation of baicalin by aqueous two-phase extraction system

AIM:To explore the extraction mechanism of the separation-purification of baicalin in aqueous two-phase systems. METHODS:By experiment,nonionic surfactant PEG-K_2PHO_4-H_2O two aqueous phase system was selected to phase-forming conditions,and the effect of temperature,pH,salt composition on partition of baicalin PEG/salt system. RESULTS:The extraction rate of baicalin in aqueous two-phase system was up to 98.6%. CONCLUSION:The two aqueous phase system for baicalin extraction is easy to operate has good reproductivity,and can be applied in mass production.
2.Content Determination of Residual Organic Solvents in Artesunate by Capillary GC

OBJECTIVE:To establish capillary GC method for the determination of residual organic solvents in artesunate,such as methanol and ethyl acetate.METHODS:Capillary GC was adopted and the temperature of flame ionization detector was 250℃.RESULTS:The linear range were 0.03~0.605 mg?mL-1 for methanol(r=0.999 8) and 0.05~1.002 mg?mL-1 for ethyl acetate(r=0.999 7).The average recovery rates were 98.7% for methanol(RSD=3.0%) and 99.1% for ethyl acetate(RSD=2.1%).Only the ethyl acetate was detected in samples.CONCLUSION:Established method is simple and accurate for the content determination of residual organic solvents in artesunate.

Background and purpose:It is verifi ed that MCM2 is a specifi c marker in the cell cycle,and expressed in all cells entering into the cycle,however,no expression is found in the differentiated cells and static period cell.Therefore,in the abnormally proliferative developmental cells and mutagenic cells,MCM2 could be used to mark the status of the cells as a cellular proliferative marker,and then to be a diagnostic tool for some heterotypical pathological changes and tumors.Our study demonstrated that the expression and clinical significance of MCM2 in bladder transitional cell cancer(BTCC).Methods:The expression of MCM2 was examined by immunohistochemistry Streptavidin-Peroxidase method in 12 cases of normal bladder tissues and 42 cases of BTCC.Results:The positive expression of MCM2 in BTCC was 100%,whereas in normal tissues,no positive expression was found(P0.05).The expression of MCM2 was closely associated with tumor pathological grade(P
4.Study on leuprorelin acetate in treatment of uterine adenomyosis with infertility

Objective To study clinical efficacy of leuprorelin acetate in treatment of uterine adenomyosis with infertility.Methods From January 1,2011 to March 31,2012,166 cycles in 166 infertile patients combined with uterine adenomyosis undergoing in vitro fertilization embryo transplant (IVF-ET) with long protocol ovum induction by leuprorelin acetate in centre of medical reproduction,ningbo women and children's hospital were studied retrospectively.In the mean time,200 cycles in 200 infertile patients with tubal factors were enrolled as control group.The volume of uterus and outcome of IVF-ET were compared and studied between two groups.Results (1) Volume of uterus:in adenomyosis group,after 2-6 cycles of injecting leuprorelin acetate (3.75 mg/28 days),the mean uterine volume was shrinked from (180 ±73) cm3 to (86 ± 67) cm3 (P < 0.05).(2) Outcome of IVF-ET:the rate of embryo implantation was 39.1% in adenomyosis group and 35.8% in control group.The rate of clinical pregnancy was 54.2% in adenomyosis group and 53.7% in control group.The rate of abortion was 4.7% in adenomyosis group and 4.2% in control group.They all did not show statistical differences (P >0.05).(3) In adenomyosis group,the rate of fertilization,two pronuclear (2PN) and superior embryo were 67.2% (319/475),60.8%(289/475) and 52.9% (162/306) in patients with failed pregnancy and 74.2% (423/570),67.7%(386/570) and 62.1% (256/412) in patients with successful pregnancy after IVF-ET,which reached significant difference (P < 0.05).Conclusion Leuprorelin acetate could improve volume of uterine adenomyosis and outcome of pregnancy in patients undergoing IVF-ET.
6.The prediction role of interleukin-18 and vascular endothelial growth factor in the ovarian hyperstimulation syndrome development

Objective To determine the role of interleukin-18(IL-18) and vascular endothelial growth factor (VEGF) in the ovari-an hyperstimulation syndrome (OHSS) development. Methods Serum and follicular IL-18 and VEGF levels were measured with ELISA in 66 IVF patients, who were divided into three study groups according to OHSS risk factors on the day of ovum retrieval and whether will devel-op OHSS in future or not. 20 patients had no OHSS risk factors (normal group), 28 patients had OHSS risk factors without developing OHSS (OHSS risk group) and 18 patients had OHSS risk factors and developing OHSS in future (OHSS group). Results Serum and follicular IL-18 levels in OHSS group were significantly higher than those of other two groups(P<0.01). Serum and follicular VEGF levels in OHSS group were also significantly higher than those of other two groups(P<0.01). Conclusion Serum and follicular IL-18 and VEGF levels in OHSS group increased significantly, which suggested IL-18 and VEGF were positive correlated with OHSS development. And follicular IL-18 and VEGF levels in serum on the day of ovum retrieval have a prediction role in OHSS development.
7.Profile of pathology in rabbit unstable plaque with ~(18)F-FDG PET/CT detection

Objective To study the feasibility of noninvasive detection of unstable plaques with 18F-Fluorodeoxyglucose (18F-FDG) PET/CT imaging. Methods Atherosclerotic plaques were induced in male New Zealand white rabbits. Animals were injected with FDG labeled with 18F,then examined with PET/CT. Aorta was explanted for photography with digital camera,and 18F-FDG uptake analysis. Thirty unstable plaques and 30 stable plaques were choosed so as to compare the quantitativly 18F-FDG uptake.The number of macrophages and smooth muscle cells was detected by immunohistochemical technique. Results Experimental group showed inconsistent uptake of 18F-FDG in the abdominal aorta. The results were confirmed in the ex vivo digital photo of the explanted aorta. 8.Influence of postoperative pelvic floor function on different surgical procedures of hysterectomy

Objective To compare the influence of postoperative pelvic floor function after different surgical procedures of hysterectomy. Methods A total of 260 patients who underwent hysterectomy in Renmin hospital of Wuhan University from January 2012 to January 2014 were enrolled in the study, and divided into 5 groups by different surgical procedures, which were total abdominal hysterectomy (A-TH;46 cases), total laparoscopic hysterectomy (L-TH;59 cases), total vaginal hysterectomy (V-TH;42 cases), abdominal intrafascial hysterectomy (A-CISH;78 cases), laparoscopic intrafascial hysterectomy (L-CISH;35 cases). Pelvic examination, pelvic organ prolapse quantitation (POP-Q), test of pelvic muscle strength, pelvic floor distress inventory-short form 20 (PFDI-20) and the female sexual function index (FSFI) questionnaire were measured after 6 months and 12 months. Results The differences of pelvic organ prolapse incidence after 6 months, A-TH and A-CISH [7%(3/46) versus 3%(2/78)], A-TH and L-CISH [7%(3/46) versus 3% (1/35)] were statistically significance (all P<0.05).POP-Q grade after 6 months between A-TH and A-CISH was statistically different in degree (P<0.05). The differences of incidence of abnormal pelvic floor muscle fatigue after 6 months of A-TH and A-CISH [59% (27/46) versus 29% (23/78)], A-TH and L-CISH [59%(27/46) versus 26%(9/35)] were statistically significant (all P<0.05), after 12 months the difference of L-TH and A-CISH [61% (36/59) versus 29%(23/78)] was statistically different (P<0.05). The differences of incidence of abnormal pelvic floor muscle strength after 6 months of L-TH and A-CISH [53%(31/59) verus 24%(19/78)], V-TH and A-CISH [60%(25/42) verus 24%(19/78)], V-TH and L-CISH [60%(25/42) verus 23%(8/35)] were statistically significant (all P<0.05);after 12 months the difference of V-TH and A-CISH [57% (24/42) versus 26% (20/78)] was statistically significant (P<0.05). Stress urinary incontinence, abnormal bowel movements after 6 months and 12 months were no significant difference between groups (all P>0.05), PFDI-20 total score was not statistically significant (P>0.05). FSFI total score after 6 months and 12 months in A-TH and A-CISH, L-TH and A-CISH, A-CISH and L-CISH were statistically significant (all P<0.05). Conclusion The influences of different surgical procedures to pelvic floor function are no statistical difference; as to the surgical resection of hysterectomy, intrafascia hysterectomy compared with extrafascia hysterectomy, the former is more helpful to the protection of the structure and function of the pelvic floor.
9.Study on the Separation of Ketoprofen Enantiomer by Pre-column Derivation RP-HPLC

Objective:To establish a method for the separation and determination of ketoprofen enantiomer .Methods:A pre-col-umn derivation RP-HPLC method was used with L-alanine-β-naphthylamine ( L-Ala-β-NA) as the derivation reagent .The RP-HPLC conditions were as follows: a Hypersil ODS-2 column (250 mm ×4.6 mm,5 μm) was applied, the mobile phase was acetonitrile-0.025 mol· L-1 phosphate buffer solution (40∶60, v/v) and the flow rate was 1.0 ml· min-1 , the detection wavelength was set at 245 nm and the column temperature was 25℃.The injection volume was 10μl.Results:Base line separation was achieved for the sep-aration of enantiomer from ketoprofen , and the retention time for S-(+) -ketoprofen and the R-(-) -ketoprofen was 24.2 min and 26.0 min, respectively.Dexketoprofen within the range of 0.025-0.125 mg had a good linear relationship (r=0.998 1) and the aver-age recovery was 90.93%(RSD =4.10%, n=9 ).Conclusion:The method is simple, accurate and reliable, which can be applied in the separation and determination of ketoprofen .
10.Simultaneous Determination of Four Glycosides in Microctis Folium by UPLC

Objective:To develop a UPLC method for determining four glycosides in microctis folium.Methods:The UPLC deter-mination was performed on an Agilent SB -C18 (3.0 mm ×50 mm, 1.8 μm) colume with 0.05% acetonitrile -phosphate solution as the mobile phase.The detection wavelength was set at 320 nm.The flow rate was 0.3 ml· min-1.The column temperature was 25℃.Results:Narcissoside, isovitexin, kaempferol-3-rutinoside and isorhamnetin-3-O-glucoside all showed good linearity with the re-covery of 98.89%,99.03%, 97.00%and 97.41%, and RSD of 0.41%, 0.84%,0.44%and 0.80%, respectively (n=9).Con-clusion:The method is convenient with good stability , repeatability and sample recovery rate , which provides basis for the quality eval-uation and control of microctis folium.
