1.The clinical and laboratorial characteristics of eleven nonsecretory myeloma

Objective To get more understanding on the symptom,and laboratory characteristics of nonsecretory myeloma(NSM).Methods 11 patients with NSM were examined by immunoglobulins IgG,IgA,IgM,IgD,IgE and their light chains,electrophoresis(PE),immunofixation electrophoresis(IFE)and serum free light chain(FLC).Results All 11 patients had more serious bone pains.all had osteolytic bone lesions.All patients were negative in PE,IFE,urine FLC tests.2 newly diagnosed patients had increased serum FLC,abnormal k/λ ratio.1 patient had normal serum FLC and normal k/λ ratio.All 11 patients had more than 30%bone marrow plasma cells.Their overall survivals were same with the typical multiple myeloma(MM).Conclusion Most NSM have more serious bone pains,and osteolytic bone lesions.PE,IFE,urine FLC tests are negative.Newly diagnosed or refractory patients have increased serum FLC,abnormal k/λ ratio,plateau phase patients have normal or reduced serum FLC.Most NSM have more than 30%bone marrow plasma cells.The therapy and prognosis of NSM are same with typical MM.
2.Bone turnover markers in multiple myeloma

Objective To study the role of bone turnover markers in multiple myeloma(MM)bone disease.Methods Thirty-eight MM patients were studied.Serum and urine samples were taken before,after 3 months and 6 months therapy.Serum samples of tartrate resistant acid phosphatage isoform-5b(sTRACP-5b),bonespesific alkaline phosphatase(sBAP),osteocslcin(sOC),urine samples of N-telopeptides of type Ⅰ collagen(NTX)were measured.Results Urine NTX concentrations were significantly higher in newly diagnosed and relapsed or refractory patients than that in pleateum patients and controls.serum TRACP-5b concentrations were significantly higher in newly diagnosed and relapsed or refractory patients than that in controls.There were no statistical significance compared with pleateum patients.Serum BAP concentrations were significantly lower than that in pleateum patients and controls.Serum OC concentrations were not statistically significant among the newly diagnosed patients,relapsed or refractory patients,pleateum patients and control.In newly diagnosed patients.urine NTX levels were significantly higher in stage Ⅲ than that in stage Ⅰ/Ⅱ,serum BAP levels were significantly lower in stage Ⅲ than that in stage Ⅰ/Ⅱ.There was a positive relationship between urine NTX and serum TRACP-5b in newly diagnosed patients.Urine NTX levels were significantly decreased,and serum BAP levels were significantly increased after 3 cycles effective therapy.There was no change in serum TRACP-5b and OC.Urine NTX and serum TRACP-5b were significantly diseased,serum BAP and OC were significantly increased after 6 cycles of effective therapy.But bone lesions on X-ray did not diminish at that time.Conclusion There is a closely relationship between bone turnover markers and bone lesions in MM.The bone turnover markers may be useful in monitoring MM progress and therapy.

The paper reviewed the main methods for the determination of paralytic and diarrhoetic shellfish toxins,including mouse bioassay,immunoassay and high performance liquid chromatography coupled with multiple techniques and so on,and summarized the principles,advantages and disadvantages of these methods.Some newly developed techniques and the development tendency for the detection of paralytic and diarrhetic shellfish poisoning toxins were also described in this paper,in order to give some reference for the detection of marine food in our country.
4.Perspective on Chinese Drugs Dispensing

OBJECTIVE:To discuss the dispensing of prescriptions of traditional Chinese medicine.METHODS:According to the dispensing procedure of Chinese traditional medicine and the principle of drug use,the problems occurred during Chinese traditional medicine dispensing were pointed out and analyzed.RESULTS:The problems occurred in the dispensing of prescriptions of traditional Chinese medicine were mainly resulted from doctors,pharmacists and nurses' lack of basic rationale or they had no intimate knowledge of operation regulations.These problems can be avoided if with efforts.CONCLUSION:Only through strictly following the prescription of Chinese traditional medicine can the quality and efficacy of Chinese drugs dispensing be guaranteed.
5.Expression and Clinical Significance of ALDH1 and ABCG2 in Cervical Cancer Cells

Objective:To detect the expression of ALDH1 and ABCG2 in the specimens from different levels of cervical lesions.Methods:Use SP immunohistochemistry to detect the expression of ALDH1 and ABCG2 in 76 cases of cervical squamous cancers,52 cases of HSIL,and 37 cases of chronic cervicitis tissues,and analyze the results.Results:①The positive expression rates of ALDH1 and ABCG2 in chronic cervicitis tissues were 5.41% (2/37) and 8.11% (3/37),in HSIL tissues were 23.08% (12/52) and 28.85% (15/52),and in cervical cancers tissues were 53.95% (41/76) and 61.84% (47/76),respectively.The positive expression rates in cervical cancers tissues were significantly higher than in HSIL and Cc hronic cervicitis tissues,(P < 0.05).②The positive expression of ALDH1 in cervical cancer tissues had correlation with tumor grade and lymphnode metastasis (P <0.05),while the positive expression of ABCG2 in cervical cancer tissues had correlation with tumor grade,clinical stage and lymph-node metastasis(P <0.05).③The expressions of ALDH1 and ARCG2 in cervical cancer were positive relative(r =0.535,P < 0.05).Conclusions:The increased expressions of ALDH1 and ABCG2 in cervical squamous cancer may play a role in the occurrence,development and metastasis of cervical squamous cell carcinoma.ALDH1 and ABCG2 may have synergistic effect in occurrence of cervical cancer.

Objective To study the expression of E-cadherin and ?-catenin in endometrial and endometriotic tissues of patients with endometriosis (EM) and to compare their expression in endometriosis at different stages. Methods Twenty-four EM patients were enrolled and the expression of E-cadherin and ?-catenin in endometrial and endometriotic tissues were determined by immunohistochemistry method. Normal endometrial tissue from 21 individuals was included as the control group. Results Compared with the control group, in E-cadherin and ?-catenin expression was shown in no difference between the secretory and the proliferative phase of endometrial tissue from EM patients. 8.Establishment of High Performance Method for Isolation and Cultivation of Hair Follicle Stem Cells of Neonatal Rats

Objective To establish a simple , practical , highly-effective and stable method for the isolation and cultivation of rat hair follicle cells. Methods Under sterile condition, single hair follicle was taken out after the skin around the barbel of SD neonatal rats was sheared off. And then the hair follicles were digested with two-step method with Type Ⅰcollagenase and trypsin. The obtained cell suspension was planted into the culture plate which was covered with extracellular matrix, and then was cultivated with K-SFM culture medium containing fetal bovine serum with volume fraction of 1%. On the next day, K-SFM culture medium was replaced with serum-free culture medium. The remaining tissues were cut into pieces and spread out in the culture flask, and then were cultivated with HG-DMEM culture medium containing serum. Two kinds of cells were harvested and then were identified by immunofluorescence. The hair follicle epithelial cells were tested by flow cytometer. Results The hair follicle epithelial cells obtained through the above methods showed rapid adherence, and were round or polygon-like , with typical cobblestone-like morphology. The long spindle-shaped cells were seen around the tissues cultivated, having many protrusions on the surface of the cells, and they were interconnected into reticular structure. The expression of cytokeratin 15, cytokeratin 19 and β1 integrin in epithelial cells were positive. Most of the epithelial cells were in the G1 phase, accounting for 75.6%. The expression of laminin ( LN), fibronectin ( FN) and vimentin in the connective tissue sheath cells were also positive. Conclusion The cells harvested by modified two-step enzyme digestion method have confirmed as hair follicle cells and fibroblasts, and the obtained cells are of rapid adherence, good homogeneity, and active proliferation.
9.Study on Repairing Skin Defect with Pelnac Tissue Engineering Skin

Objective To explore the feasibility of repairing skin defect with Pelnac tissue engineering skin and cultured epidermal stem cells ( ESCs) and fibroblasts. Methods ESCs and fibroblasts in SD rats were isolated and cultured in vitro, and then were detected by immunohistochemistry. Fibroblasts were seeded in Pelnac material to construct dermis, and ESCs were seeded on the surface of dermis to construct tissue engineering skin. Thirty BALC/c nude mice were randomly divided into groups A, B and C. After mouse back skin defect model was established, group A was transplanted with Pelnac material, group B was transplanted with compound of Pelnac and fibroblasts, and group C was transplanted with Pelnac tissue engineering skin. The wound healing was observed after operation. The wound tissues were sampled for pathological histology test on the 5th and 7th day after transplantation. Results The expression of cytokeratin 15 and β1 integrin in ESCs was positive, and the expression of laminin ( LN) and fibronectin ( FN) in fibroblasts was positive. In group C, the wound healing of nude mice was the best, characterized by the proliferation of fibroblasts, obvious new capillaries and regularly arranged epidermal cells at the transplantation sites. Conclusion Pelnac tissue engineering skin has good effects on promoting repair of skin defect of nude mice.
10.Association of fibrinogen ?G-455A polymorphism with plasma fibrinogen level in patients with coronary heart disease

Objective To evaluate whether ?G-455A gene polymorphism increases the risk of coronary heart disease(CHD) due to elevated plasma fibrinogen.Methods A total of 1 485 patients who had received coronary angiography due to chest pain or suspected cardiac ischemia by non-invasive examination were included in the study.According to the angiographic results,all the patients were divided into the control group(n=466) and coronary heart disease group(n=1 019).Patients in the coronary heart disease group were further divided into stable angina pectoris group(SAP,n=674) and acute coronary syndrome group(ACS,n=345) according to their clinical presentation.We investigated G-455A polymorphism of ? fibrinogen gene and plasma fibrinogen level in all the patients.Results Increased plasma fibrinogen levels were observed in CHD groups compared with controls(ACS: 380.92?92.35 mg/dL,SAP: 352.49?94.89 mg/dL,control: 311.72?87.09 mg/dL,P
