3.The study of antiplatelet aggregation activity of small dose clopidogrel combined with paeonol

Objective To investigate the antiplatelet aggregation activity of clopidogrel ( small dose) combined with paeonol. Methods Antiplatelet aggregation activity was evaluated by platelet aggregation induced by adenosine diphosphate in vitro, the weight of arteriovenous bypass thrombosis in rats and bleeding time in mice. Results The combination significantly inhibited platelet aggregation, and reduced formation of arteriovenous bypass thrombosis as well as prolonged the bleeding time. Moreover, the activity of combined group was more superior to clopidogrel and paeonol, respectively. Conclusion The antiplatelet aggregation activity of the combination is better than clopi-dogrel, which has bright prospects in clinical applications.
4.Transfection of maspin/PCR2.1 vector inhibits uPA and uPAR expressions in gastric cancer cells in vitro

Objective To investigate the inhibitory effect of maspin gene on metastasis and invasion of gastric cancer and its mechanism.Methods Maspin gene was ligated to the expression vector PCR2.1 with T4 DNA ligase after the PCR2.1 was digested by HindⅢ/XbaⅠ.The recombinant vector maspin/PCR2.1 was transfected into human gastric cancer cell lines MKN28 and SGC7901,then the mRNA and protein expression changes of maspin gene,uPA and uPAR were detected respectively by PT-PCR and Western blotting.Results After identified by digestion and sequencing,the reconstructed plasmid was confirmed to contain the correct and full nucleotide sequence of maspin gene.The expressions of maspin gene were up-regulated in MKN28 and SGC7901 cell lines,while the expressions of uPA and uPAR were down-regulated.Conclusion The expression vector maspin/PCR2.1 is constructed successfully and can be expressed in eukaryotic cells.Expressions of uPA and uPAR can be inhibited by maspin gene.
5.Feature of immunoglobulin heavy chain complementarity determining region 3 gene in neonates with different gestational age

Objective To study the feature of immunoglobulin complementarity determining region 3 (CDR3) gene in neonates of different gestational age (GA), and the effect of neonatal maturity on the diversity of CDR3 nucleotide sequence. Methods DNA were extracted from cord blood of 10 neonates of very immature (25-30 weeks), 12 immature (31-36 weeks), and 11 mature (37-41 weeks). CDR3 sequence was amplified suing nested PCR technique then cloned and sequenced. Results 1. There was a CDR3 length of 29.4±7.8, 32.4±9.2 and 40.8±10.7bp, including NDN length of 13.5±5.6, 16.1±7.8 and 22.0±8.5bp respectively in very immature, immature and mature neonates. 2. DP73 and DP75 were preferentially used in very immature and immature neonates, VH5, DP73 and DP75 were used in mature neonates. The usage rate of DP73 and DP75 reduced, whereas VH5 was raised with GA increasing. 3. For D gene segment, there was a frequent use of DN, DQ52 and DXP in very immature neonates, whereas DXP, DLR and DN in immature and mature neonates. 4. JH4 usage was preferential, followed by JH6 in all neonates, and their usage rate was decreased with GA increasing. 5. 63.3%, 68.8% and 92.0% of CDR3 sequence respectively in very immature, immature and mature neonates have an open reading frame with 303bp long encoding 101 residues. Conclusion During the early life of neonates, VH-D-JH rearrangement of IgH gene is in an active condition, but the diversity is limited. Humoral immunity is a gradual development with increasing GA. Both heterogeneity and similarity of CDR3 sequence exists in neonates with different GA.

Objective To investigate the relationship among IGF-II expression and cell proliferation and apoptosis in human colorectal adenocarcinoma. Methods 60 paraffin embedded samples of human colorectal adenocarcinomas were used in this study. IGF-ⅡmRNA expression was detected by in situ hybridization, proliferation cell nuclear antigen(PCNA) protein was measured by immunohistochemical staining and apoptosis was determined by TUNEL technology. Fifteen normal colorectal tissues were used as controls. Computerized image analysis system qualitatively analyzed the positive ratio of above three indices in the cancer and normal tissues. Objective To construct a recombinant eukaryotic vector expressing maspin cDNA,and to explore the effect of maspin overexprssion on the proliferation of human gastric carcinoma cell line SGC7901.Methods The fragment of maspin gene was amplified by polymerase chain reaction(PCR).An eukaryotic vector expressing maspin(maspin/PCR2.1)was constructed with PCR2.1,and transfected into SGC7901 cells.The expression of maspin at mRNA and protein level was detected by RT-PCR and Western blotting.The proliferation of SGC7901 cells was observed by MTT.Cell cycle was analyzed by flow cytometry.Results Recombinant plasmid maspin/PCR2.1 was constructed and transfected into SGC7901 successfully.The mRNA and protein levels of maspin were significantly higher in the maspin/PCR2.1 group(33.6?1.2,23.4?1.6)than that in the blank PCR2.1(15.0?1.5,12.3?1.5)and the untreated group(13.7?2.0,12.0?1.3)(P
8.Clinical Analysis on 38 Cases of Laparoscopic Operation in the Treatment of Large Ovarian Cysts

Objective To investigate the feasibility of laparoscopic operations in treating large ovarian cysts.Methods A total of 38 patients with large ovarian cysts underwent laparoscopic cyst cystectomy or adnexectomy on the affected side from December 2002 to October 2005.The 2nd or 3rd trocar(usually on the tumor side)was inserted into the cyst,the fluid in which was drained with a suction unit.The cyst was then removed after being elevated toward the incision,which was extended to 20 mm.Results Large ovarian cysts in the 38 cases were all removed successfully,including cystectomy in 28 cases and adnexectomy in 10 cases respectively.No complication occurred.The follow-up period was 1-24 months(mean:10 months)for 35 cases.No abnormal findings in pelvic examination and B ultrasonography were observed.Conclusions Laparoscopic operation is a safe and feasible therapeutic method for large ovarian cysts.
9.Immunohistochemical and morphological characteristics of the papillary thyroid microcarcinoma

Objctive:To investigate the immunohistochemical and morphological characteristics of the papillary thyroid microcarcinoma(PTM) ,and differential diagnosis of the related disease.Methods:The morphological characteristics of 223 patients with PTM were observed under light microscope.Seventy-four cases of PTM and 32 cases of proliferative lesion of thyroid were observed under light microscope with stains of hematoxylin and eosin and immunohistochemical staining.The antibody included CK19,MC,Galectin-3 and CD56.Results:Eighty-six cases were follicular-patterned and 31 cases nuclear features were untypical in 223 cases of PTM.The positive expression rates of CK19、MC、 Galectin-3、CD56 were 100.0%,98.6%,98.6% and 4.1% in 74 cases of PTM,and were 37.5%,12.5%,18.8%,68.8% in proliferative lesion of thyroid,respectively.Conclusion:Some cases of PTM show a follicular-patterned and the nuclear features.It is untypical compared with classical papillary thyroid carcinoma.It can be differentiated from proliferative lesion with absence of envelope,obviously various and unusually proliferated follicular cells,sclerotic stroma,neoplastic follicles among the collagen and normal follicles.The wide and intensive expression of CK19,Galectin-3,and the negative expression of CD56 are extremely useful in the diagnosis of PTM,four-marker panel with CK19,Galectin-3,MC and CD56 can improve the specificity and accuracy of the differential diagnosis of PTM.
10.Evaluation method about the learning effect of web-based PBL with key factors simulation

Objective This paper aims to use components as the key factors to explore the feasibility of evaluation method about the learning effect of Web-based PBL.Methods We distributed a questionnaire to 101 students of clinical class of 2007 and 2008 grade in Guangzhou Medical University,and electronically distributed after 2 weeks.Cranach's α coefficient WaS used to evaluate intrinsic consistency reliability of the questionnaire.We applied principal axis factoring to extract components and to explain components which were orthogonally rotated by varimax.Multi-factor regression model Was simulated with key factors and the model's performance of fitting WaS tested.Results Multi-factor regression mode(WPBLSUM)=0.545 Collective collaboration capabilities+0.202 Self-learning and quest skills+0.137 linical reasoning and problem-solving capabilities+0.116 memorizing and understanding of medical knowledge.The coefficient of determination R2Was 0.998 and the corrected coefficient of determination R2Was 0.998.indicating that the model's fit results are obvious.Conclusion This paper demonstrates that using components as the key factors to simulate and assessing the Web-based PBL learning effect is feasible,which assesses the Web-based PBL learning effect well from four different aspects.
