1.Clinical efficacy of daniston danshentong capsule in treatment of patie nts with nodular-cystic acne

AIM: To observe the clinical effects of daniston danshentong c apsule in treatment of patients with nodular-cystic acne. METHODS: A group of patients (31 cases finished their observation in all 35 cases) wa s selected to take daniston danshentong capsule ( 0.25 g/capsule), 4 capsules three times daily. After continuous taking the agent for 8 weeks, the dosage ch anged to be 3 capsules three times daily. The treatment lasted for 12 weeks. RESULTS: The total effective rate was 83.9 %. Compared the mean number of the nodular-cystic acne before treatment (23?11) with that after 2 weeks of treatment (19?9), after 4 weeks (15?6), after 8 weeks (9?6), and aft er 12 weeks (7?7), all of the P values were less than 0.01 . The effective rate increased gradually along with administration time lengtheni ng. CONCLUSION: Daniston danshentong capsule is effective in tre atment of nodular-cystic acne, and eight-week course is a suitable for the tre atment.
2.Change of Kv Channel Interacting Protein 1 and Gamma-Aminobutyric Acid-ergic Neurons in Hippocampus of Pentylenetetrazole Induced Epilepsy Rats

Objective To explore the role of Kv channel interacting protein 1(KChIP1)in the process of epileptic seizure and the relationship between KChIP1 and gamma-aminobutyric acid-(GABA)ergic neurons.Methods Normal female Sprague-Dawley rats were treated with pentylenetetrazole to make acute pentylenetetrazole models of epilepsy.Laser Scanning Confocal Microscope(LSCM)combined with double-labeled immunohistochemical technique was applied to observe the expression of the KChIP1 and the GABAergic neurons in the hippocampus of rats.Results The number of KChIP1-postive neurons in the hippocampus was significantly increased in the acute pentylenetetrazole model rats(P<0.05).There was no significant difference in the number of double-labled neurons(P>0.05),nor of the GABA-postive neurons between the model rats and the controls.The ratio of double-labeled neurons/total positive neurons was 63.9% in the hippocampus.Conclusion The KChIP1 might be involved in epileptogenesis of pentylenetetrazole induced seizure.The KChIP1 was associated with GABAergic neurons,whereas it may be functionally different from GABA.
3.Cytomegalovirus pneumonia after allogeneic hematopoietic stem cell transplantation

To compare the clinical features and prognosis in patients with cytomegalovirus pneumonia from other pneumonia after allogeneic hematopoietic stem cell transplantation (allo-HSCT). A total of 118 patients with pulmonary complications after allo-HSCT from March 2016 to June 2019 were analyzed retrospectively, who were divided into cytomegalovirus (CMV) pneumonia group ( n=34) and the non-CMV pneumonia group ( n=84). Compared with non-CMV pneumonia group, CMV pneumonia group presented earlier median onset time (1.8 vs.6.0 months, P=0.015) after allo-HSCT, more dyspnea (41.2% vs. 19.0%, P=0.012), hypoxemia (38.2% vs. 13.1%, P=0.006), and interstitial pneumonia (82.4% vs. 23.8%, P<0.01).The incidence of CMV-viremia and serum viral load in CMV pneumonia group were significantly higher than those in non-CMV pneumonia group. Consistently, and the development of mixed infection in CMV pneumonia group was higher than that of non-CMV pneumonia group (41.2% vs. 16.7%, P=0.013). The median follow-up time was 12.8 (0.4-46.5) months. The 1-year attributable mortality in CMV pneumonia group was significantly higher than that in non-CMV pneumonia group (26.5% vs. 10.7%, P=0.004), while the 1-year overall survival rate was significantly lower than that in non-CMV pneumonia group (61.8% vs. 85.7%, P=0.001). Reduced-intensity conditioning (RIC)( P=0.036), high flow ventilation ( P=0.033) and negative CMV-viremia ( P=0.009) were unfavorable prognostic factors of patients with CMV pneumonia. Compared with those with non-CMV pneumonia, patients with CMV pneumonia had more characteristic clinical manifestations and imaging features. However, due to the higher incidence of mixed infections, the causes of pneumonia need to be identified by bronchoscopic alveolar lavage. In conclusion, patients with CMV pneumonia have worse clinical outcome. RIC, high flow ventilation and negative CMV-viremia are adverse prognostic factors for CMV pneumonia.
4.Effects of EEF1 A2 on the growth and proliferation of pancreatic cancer cell

Objective To investigate the effects of EEF1 A2 on growth and proliferation of the human pancreatic cancer cell line SW1990. MethodsThe EEF1 A2 gene was introduced into pancreatic cancer cell line SW1990 by adenovirus vector. The effects of EEF1 A2 on the growth of human pancreatic cancer cell were measured by MTT. Cell cycle was detected by flow cytometry and cell growth rate was examined by soft agar cloning formation test. ResultsAfter EEF1A2 induction, the expression of EEFA1 A 2 mRNA in pancreatic cancer cell line SW1990 increased, value of A750 at 72 h was 1. 2996 ±0. 2091, number of cells was 81250, cloning efficiency at 14 d was 82%, all of these parameters were significantly higher than those in the groups of blank adenoviras vector and PBS groups (P <0.05 ) ; the percentage of G1 phase cell was 28.5%, which was significantly lower than those in the groups of blank adenovirus vector and PBS groups; the percentage of Sphase ceil was 60.9%, which was significantly higher than those in the groups of blank adenovirus vector and PBS groups (P < 0.05 ). ConclusionsEEF1 A2 gene significantly enhanced cell growth and proliferation in human pancreatic cancer in vitro.
5.Effect of ginkgo biloba exocarp polysaccharides (GBEP) on p53 gene expression and telomerase activity of human gastriccancer SGC-7901 cells

AIM To study the influence of ginkgo biloba exocarp polysaccharides(GBEP) on p53 gene expression and telomerase activity of human gastriccancer SGC 7901 cells in vitro . METHODS The p53 gene expression level of human gastriccancer SGC 7901 cells were measured by immunohistochemistry ABC. The telomerase activity of human gastriccancer SGC 7901 cells were measured by TRAP ELISA. RESULTS GBEP(80~160 mg?L -1 ,48 h) inhibited the expression of mutated type p53 genes and telomerase activity of human gastriccancer SGC 7901 cells. CONCLUSION The action mechanism of GBEP that inhibited the proliferation of SGC 7901 cells may relate to its down regulation effect on the expression of p53 genes and telomerase activity of human gastriccancer SGC 7901 cells.
6.Unusual mtDNA sequencing results from ancient DNA.

Sequence analysis of human mitochondrial DNA(mtDNA) is being used widely to characterize individual identification, particularly when there is insufficient nuclear DNA in samples for typing. Hair shafts, bones, teeth and other samples that are severely decomposed may be subjected to mtDNA analysis. As sample decomposes, however, the possibility of mtDNA to be degraded becomes high and the possibility of spurious results becomes high. In this case mtDNA sequencing results must be carefully analyzed. We got unusual results while typing two human bone samples, which were not compatible with human mtDNA sequence. Bones were about 50 and 35 years old. We report the results with discussions about ancient DNA sequencing.

7.Compatibility of a novel ethylenediamine modified polylactic acid with osteoblasts.

Biocompatibility of a newly developed ethylenediamine modified poly (DL-latic acid) (EMPLA) with osteoblasts was investigated by means of cell morphology and cell proliferation. Films of PLA and EMPLA were made by solvent casting. Osteoblasts obtained from crania of neonatal Wistar rats were cultured on surfaces of PLA and EMPLA, with glass as control. The cell morphology was observed by phase contrast microscope and the cell proliferation was determined by MTT assay. The morphology observations revealed that the osteoblasts cultured on EMPLA spread wider than those on PLA, and much more cells were confluent on EMPLA, compared to those on PLA and glass. The growth curves showed the osteoblasts on EMPLA grew faster than did those on PLA and glass. The results exhibited that the biocompatibility of EMPLA with osteoblasts is better than that of PLA and glass, which suggested wide applications of EMPLA in biomedical area, especially in tissue engineering.

8.Functional genomic approaches to explore secondary metabolites in medicinal plants.

Extensive genomic data concerning medicinal plants are rather scarce and insights of the secondary metabolic pathways and their regulatory mechanism are insufficient, hampering the broad application of cell or tissue cultivation and metabolic engineering to producing high-value secondary metabolites. The integration of cDNA-AFLP based transcript profiling and metabolomics, a new development of functional genomic approaches could establish correlations between the changes of secondary metabolites and expressions of related genes. It has manifested widely applicative prospects in seeking genes involved in biosynthesis of secondary metabolites and exploring secondary metabolic pathways. Functional genomic approaches are promising trends in the field of medicinal plants secondary metabolites research and will lead to better utilization of natural medicinal resources.

10.Visualization Analysis on Research Literature of TCM Regulation of miRNA

Objective To analyze the current status and trends of TCM regulation of microRNA(miRNA)using visualization methods.Methods The literature related to the TCM regulation of miRNA was retrieved from CNKI,VIP and Wanfang Data from the establishment of the databases to May 31,2022.Excel 2019,VOSviewer 1.6.18,and CiteSpace 5.8.R3 software were applied to visualize and analyze the year of publication,journal source,author and keywords of the included literature.Results A total of 787 articles were included,and the number of publications continued to rise.Source journals with more publications includes Chinese Journal of Experimental Traditional Medical Formulas,China Journal of Traditional Chinese Medicine and Pharmacy,Chinese Archives of Traditional Chinese Medicine,etc.Research teams were formed with Wang Jie,Liu Xide,Diao Limei,etc.as the representatives,and the cooperation among the teams was not very close.Analysis on the keywords in the literature showed that the studies mainly focused on apoptosis,miRNA-21,electroacupuncture,atherosclerosis,proliferation and other related fields;TCM intervention that accounted for the most research were extracts of Chinese materia medica,with 42%of the studies;there were 24 miRNAs studied≥3 times,and the most studied miRNA was miRNA-21.Conclusion The research hotspots of TCM regulation of miRNA are mainly the molecular mechanism of various TCM therapeutic tools to improve cell autophagy,apoptosis,inflammatory response and other pathways through regulation of miRNAs.The trend of research is to study the mechanism of empirical prescriptions of famous TCM practitioners based on precise therapeutic strategies.
