1.Anti-tumor Activity of Adriamycin-loaded Chitosan Nanoparticles Surface-modified with Glycyrrhizin on Human Hepatoma Cell Line SMMC-7721

OBJECTIVE: To investigate the anti-tumor activity of adriamycin-loaded chitosan nanoparticles surface-modified with glycyrrhizin on human hepatoma cell line SMMC-7721. METHODS: Inhibition effect of drug-loaded nanoparticle on the proliferation of SMMC-7721 cells was determined with MTT method. Intra-cellular distribution of nanoparticles and morphological change of SMMC-7721 cells before and after treatment were observed by confocal laser scanning microscope (CLSM) and the apoptosis induced by adriamycin was measured by TUNEL assay. RESULTS: The inhibition effect of adriamycin-loaded chitosan nanoparticles surface-modified with glycyrrhizin on the proliferation of SMMC-7721 cells increased 6.36 times lower in comparison with free drugs. The inhibition effect of adriamycin-loaded chitosan nanoparticles had no obvious improvement. Nanoparticles surface-modified with glycyrrhizin promoted the distrubition of adriamycin in the nucleus, so as to promote the anti-tumor activity of adriamycin. TUNEL assay indicated that nanoparticles surface-modified with glycyrrhizin induce DNA fragmentation and nucleus breakdown to induce the apoptosis of SMMC-7721 cells. CONCLUSION: Adriamycin-loaded chitosan nanoparticles surface-modified with glycyrrhizin can used as potential active hepatocyte-targeted delivery carrier. Pharmacokinetic evaluation of it is worthy of further studing.
2.THE SUPPRESSIVE EFFECTS OF CYCLOSPORIN A ON IMMUNE FUNCTION IN MICE

The suppressive effects of cyclosporin A ( CsA ) made in China on immune function were studied in mice. CsA ( 25 and 50 mg/kg ig for 4 d ) caused a significant decrease of spleen plaque forming cell ( PFC ) in mice. At the dose of 25, 50 and 100 mg/kg ig for 4 d, CsA inhibited the production of hemolysin in a dose-dependent manner in mice immunized with SRBC. CsA ( 25, 50 and 100 mg/kg ig for 10d ) also markedly inhibited the delayed cutaneous hypersensitivity ( DCH ) induced by DNCB in mice. CsA 50 mg/kg ig for 14d prolongated survival time of cardiac grafts. The treatment with CsA could not reduce the clearance rate of iv charcoal particles as well as bone marrow cells in mice. There was no significant difference of immu-nosuppressive effect on PFC response between China-made CsA and Sandoz-made CsA

The intention of quitting smoking and its influencing factors were surveyed with questionnaire developed by Chinese Center for Disease Control in 734 smokers in Guangzhou City.The results showed that 57.6%of the interviewed smokers intended to quit smoking.Logistic regression analysis demonstrated that 5 factors were significantly associated with the quitting intention:the attitude against smoking of family,educational background,the cost of cigarettes,addiction to smoking,awareness of the harm of smoking to health.
4.Ultrasound diagnosis of occult inguinal hernia in children

ObjectiveTo evaluate ultrasound diagnosis of occult inguinal hernia in children. MethodsThe contralateral site of 132 established unilateral inguinal hernia cases was checked by ultrasound and 33 outpatient suspected occult inguinal hernia cases underwent B-mode ultrasonography. [WT5”HZ]Results Occult inguinal hernia was diagnosed in 15 cases (11.4%) in the 132 children,and diagnosis of occult inguinal hernia was founded by ultrasound in 17 out of the 33 outpatient cases. No opposite inguinal hernia developed in the ultrasound negative 117 patients during follow up of 2 to 12 months,and hernia developed in one out of the 16 outpatient cases initially judged as negative by ultrasound. ConclusionUltrasonography is useful for the diagnosis of occult inguinal hernia in children.
5.Research of exons 19 and 21 mutations of epidermal growth factor receptor gene in 109 cases of lung adenocarcinoma in Yancheng area

Objective To investigate the 19 and 21 exon mutation status of the epidermal growth factor receptor(EGFR) gene in the patients with lung adenocarcinoma and its distribution differences among different groups .Methods The related samples in 109 cases of lung adenocarcinoma were collected in our hospital during 2013-2014 .The 19 and 21 exons of EGFR gene were detected by adopting the ARMS‐PCR method .Results Among 119 cases ,19 and 21 exons mutations were found in 56 cases(51 .38% ) ,in which 24 cases (22 .02% ) were in exon 19 and 33 cases (30 .28% ) were in exon 21 .59 cases were male with 19 cases(32 .20% ) of mutant type and 50 cases were female with 37 cases (74 .00% ) of mutant type;64 cases were ≥ 60 years old with 34 cases (53 .13% ) of mutant type ,45 cases were <60 years old with 22 cases(48 .89% ) of mutant type;33 male cases were smoking with 7 cases(21 .21% ) of mutant type and 25 male cases were non‐smoking with 12 cases (48 .00% ) of mutant type .Conclusion The 19 and 21 exon mutation rate of EGFR gene in the patients with lung adenocarcinoma is higher in this area ,the 21 exon mutation rate is slightly higher than that of 19 exon;the mutation type and mutation rate are correlated with sex and smoking history ,but have no correlation with age .

7.STUDIES ON SIX MINERAL ELEMENT CONTENTS OF RICE BRAN AND MILLED RICE FROM THREE COLOR RICE CULTIVARS

Objective: To investigate six mineral element contents of rice bran and milled rice from three color rice cultivars (white rice, red rice, black rice). Methods: Select 36 typical cultivars and determine Se, Zn, Fe, Ca, Mn, Cu contents in their rice bran and milled rice with AFS-2201 atomic fluorescent spectrometer and AA-680 atomic absorption spectrometer. Results: 1. The six mineral element contents of rice bran were 2.58, 3.35, 10.04, 4.58, 12.79 and 6.59 times as those of milled rice respectively. 2. The Ca content of black milled rice was significantly higher than that of white and red milled rice. The Mn content in rice bran of white rice was significantly higher than those of black and red rice. Other mineral element contents were not significantly different. 3. For rice bran and milled rice of different cultivars, the contents of each mineral element in three color rices varied greatly. Conclusions: 1. The content of six minerals in rice bran was 5.66 times on average as that in milled rice in all cultivars of three color rice. 2. Eight cultivars of the highest content of each mineral in rice bran and milled rice were recommended for commercial use and human consumption.

Objective To observe the anti-pyretic effect of F022 from Radi x Isatidis.Methods The fever model was induced by intravenous injection of endo toxin into rabbits.Body temperature of the rabbits was measured at 0.5,1,2,3,an d 4h after administration in semi-vivo and in vivo.Results F022 subsided th e fever in rabbits induced by endotoxin in semi-vivo and vivo.Conclusion It is valuable for further research that F022 component is used as the main to scr een anti-pyretic active constituent.

10.Transferrin-labeled magnetolipsomes: preparation and magnetic resonance imaging in vitro

BACKGROUND:Transferrin (Tf) is one suitable ligand to be conjugated to drug delivery systems to achieve site-specific targeting and desired therapeutic effect, due to its specific binding to transferrin receptors (TfR), and high expression on the surface of tumor cells. Contrast agents are also modified with Tf to achieve specific tumor imaging. OBJECTIVE:To prepare Tf-labeled magnetoliposomes (MLs), and characterize their utility as TfR targeted MR specific contrast agent in vitro. METHODS:MLs and Tf-MLs were prepared by lipid film hydration method and covalent coupling method, respectively. Tf-MLs were characterized by their mean size, zeta potential, polyindex, r2 relaxivity, Tf-binding efficacy and cytotoxicity.In vitro MRI contrasting properties of the suspended nanoparticles incubated with HepG2 cells were determined. RESULTS AND CONCLUSION:The mean diameter, polydisperisity index, zeta potential and r2 relexivity of Tf-ML were 95.1 nm, 0.21,-1.25 mv and 94.62 mmol-1/s, respectively. The coupling efficiency was calculated and the values obtained were 59.4 μg Tf/μmol phospholipid corresponding to about 27 molecules of Tf-MLs. After a 2-hour incubation with rhodamine-labeled Tf-MLs, rhodamine fluorescence was detected intensively in the plasma membrane and the cytoplasm of the TfR-overexpressing HepG2 cells. In contrast, Tf-ML showed little binding in MCF-7 cells that had low TfR level. HepG2 cels incubated with Tf-ML showed much higher intracellualar iron density than incubated with non-targeted MLs.In vitro MR T2WI of cells demonstrated the centrifuge tube containing HepG2 cells incubated with Tf-MLs produced a lower visible signal intensity than that treated with non-targeted MLs. Tf-MLs showed their potentials such as high r2 relaxivity, specific binding ability characteristics. These results suggest the availability of Tf-MLs to serve as a targeted contrast agent.
