OBJECTIVE:To discuss the strategy for screening new leading compounds and discover the novel drug candidates,which have the independent intelligent property right,from botanicals METHODS:Screening the new leading componds from the single plant which plays the main therapeutic role in classical prescriptions or proved recipes were reviewed RESULTS:The results showed that the frequency of use of certain kinds of plant was higher than that of the others in treatment of certain disease,it is hypothesized that the plant may contain the active ingredient for the therapy of certain kinds of disease CONCLUSION:Screening new leading compound from Chinese materia medica is a shortcut with less risk,which may promote the development of new drugs in China
2.Chronic Renal Failure Complicated by Primary Hypothyroidism

Objective: To investigate the clinical features of chronic renal failure (CRF) complicated by primary hypothyroidism.Methods: Thyroid function, thyroid microsome antibody ( TM), and thyroid globulin antibody (TG) were determined withradioimmunoassay in 23 CRF complicated by primary hypothyroidism patients, and the serum calcium, phosphorus, potassi-um, cholesterol, glucose, urea nitrogen, hemoglobulin, and endogenous creatinine clearance rate were also assayed. Thirty-onepatients with only CRF were in control group. Results:The level of thyroid-stimulating hormone (TSH), Ch, and Ca in pa-tients with CRF complicated by primary hypothyroidism were higher than that in control group,and serum free T4 (FT4),K,and P were lower than that in control group. The others had no difference between two groups. TG and TM increased in2 cases. Conclusion:The most important criteria in diagnosing CRF complicated by primary hypothyroidism is TSH twotimes higher than normal upper limit. The others include decreased FT4, normal level of Ca, P, and K, and increased Ch,TG,and TM. The diagnosis should be performed combined with clinical manifestations.
3.Association between apoptosis induced in hepatoblastoma HepG2 cells by Cdk7 silencing and decreased pRb and Cdk2 phosphorylation

Aim To evaluate the effect of Cdk7 silencing on the c ell cycle control, the phosphorylation level changes of Cdk2 and pRb in human he patoblastoma HepG2 cell culture in vitro, and to validate Cdk7 as a novel ta rget for anticancer therapeutics.Method Levels of Cdk7 and the phosphorylation levels of Cdk2 and pRb were measured by Western-blotting. DNA c ontents, cell cycle and apoptosis induced by Cdk7 silencing were analyzed by flo w cytometry and ultrastructural changes of cells were observed with transmission electron microscopy.Result The phosphorylation levels of pRb a nd Cdk2 and the levels of Cdk7 decreased in a concentration-dependent manner wh en the concentration was above 100 nmol?L -1. 4.Clinical Observation of Acupuncture plus Bobath Therapy for Motor Dysfunction of Core Muscles in Patients with Post-stroke Hemiplegia

Objective To investigate the therapeutic efficacy of acupuncture plus Bobath therapy in treating motor dysfunction of the core muscles in patients with hemiplegia after cerebral stroke.Method Totally 144 patients with post-stroke hemiplegia were divided into a control group, treatment group 1 and treatment group 2. The treatment group 1 was intervened by Bobath therapy, while the treatment group 2 was by abdominal acupuncture and scalp acupuncture in association with Bobath therapy. The recruited patients were examined for the manual muscle strength rating of the abdominal core muscles, Berg Balance Scale (BBS) score, and Barthel Index (BI) score on the day of admission, the 30th day and 60th day after admission.Result There were no significant inter-group differences in comparing the core muscle strength, BBS, and the Activities of Daily Living (ADL) on the day of admission (P>0.05). The core muscle strength, BBS, and ADL were significantly increased on the 30th and 60th day after admission in the three groups (P<0.05), and the scores in treatment group 2 were significantly higher than that in the control group and treatment group 1 (P<0.05).Conclusion Acupuncture plus Bobath therapy can significantly improve the motor dysfunction of the core muscles in patients with hemiplegia after cerebral stroke.
5.Effect of fluoxetine on the viability and lipopolysaccharide-induced tumor necrosis factor-α release of primary cultured rat astrocytes

ObjectiveTo investigate the effects of fluoxetine on viability and lipopolysaccharide (LPS)-induced TNF-α release of primary cultured rat astrocytes.MethodsThe cells were plated on 96-well tissue culture plates and treated with (5,10,20,40) μM fluoxetine for 24 hours,MTT method was used to measure the cell viability.The cells were plated on 48-well tissue culture plates,treated sequentially with (5,10,20,40) μM fluoxetine and 1 μg/ml LPS,and enzymatic linked immunosorbent assay(ELISA) was used to detect the TNF-α level of the cell supematant.ResultsCompared to viability of the control group( OD value:0.20 ± 0.017 ),fluoxetine at the dose of 20 μM,40 μM increased the viability of astrocytes ( OD value:0.23 ± 0.013,0.24 ± 0.012 ) (P <0.05,P<0.01 ).Treatment with LPS for 24 hours,the level of TNF-α was significantly increased ( 53.84 ±24.84) pg/ml compared to the control group( 8.00 ± 10.87)pg/ml (P < 0.01 ),fluoxetine at a dose of 10 μM can suppressed LPS-induced TNF-α release from astrocytes ( 28.85 ± 3.36 ) pg/ml (P < 0.05 ).ConclusionFluoxetine can increase astrocytes viability and suppress LPS-induced TNF-α release from astrocytes.
6.Trend prediction of chaotic time series

To predict the trend of chaotic time series in time series analysis and time series data mining fields, a novel predicting algorithm of chaotic time series trend is presented, and an on-line segmenting algorithm is proposed to convert a time series into a binary string according to ascending or descending trend of each subsequence. The on-line segmenting algorithm is independent of the prior knowledge about time series. The naive Bayesian algorithm is then employed to predict the trend of chaotic time series according to the binary string. The experimental results of three chaotic time series demonstrate that the proposed method predicts the ascending or descending trend of chaotic time series with few error.

Objective To Summarize experence of Dynamic Hip Screw for the treatment of femoral intertrochanteric fracture. Methods 54 cases with femoral intertrochanteric fracture were treated by DHS internal fixation. Results All patients were followed up for 4~36 months with an average of 7 months. Satisfactory effect was obtained. Conclusions The recovery of intact posterointernal cortex of femoral intertrochante, the standard postiton of fixation of DHS and an incarease in postoperative functional exercise are the key to obtain a satisfactory therapeutic effect.
8.Effect of 5-Aza-2′-deoxycytidine on experimental lung metastasis of breast cance

Objective To investigate the effect of 5-Aza-CdR on experimental lung metastasis of breast cancer and the possible mechanisms.Methods MDA-MB-231 cells were divided into control group and 5-Aza-CdR-treated group.The mRNA expressions and promotor methylation status of BRMS1 and CXCR4 of the MDA-MB-231 cells were evaluated by SqRT-PCR and MSP respectively.Then,the MDA-MB-231 cells of control group and 5-Aza-CdR-treated group were injected into BALB/c nude mice through lateral tail veins,respectively.five weeks later,the mRNA abundance of the target gene HPRT and internal control gene GAPDH of the lung tissues from the mice were evaluated by FqRT-PCR.Results 5-Aza-CdR upgraded the BRMS1mRNA expression significantly than that in control group(0 versus 0.39?0.001,P0.05) and the status of unmethylated CXCR4 CpG island 1 in promotor were not changed significantly.The Ct values of HPRT and GAPDH in control and 5-Aza-CdR-treated groups were 24.75?1.55,16.19?0.69 versus 27.61?1.67,17.48?0.96 respectively,2-??Ct=0.34.The mRNA abundance of the HPRT was lower and there were fewer metastases in the lungs of 5-Aza-CdR-treated group compared with control group.Conclusions 5-Aza-CdR can activate tumor metastasis suppressor gene BRMS1 by demethylation mechanism,and thus,decreased the ability of breast cancer MDA-MB-231 cells for experimental metastasis to lungs.

Objective To investigate principle and clinic pathologic characteristics of autoimmune thyroid disease(AITD) associated nephrosis. Methods By introducing 5 cases and related reference to review clinic pathologic characteristics of AITD associated nephrosis. Results AITD associated nephrosis regard proteinuria as principal, and a handful of performance is nephritic syndrome and nephritis syndrome. Pathologic characteristics are diversity. It is usually membranous nephrology, mesangial proliferative glomerulonephritis and focal and sclerosing glomerulonephritis. Conclusion Treatment of AITD associated nephrosis itself and immunosuppressive agent therapy might be useful to get remission of it.
10.Clinical analysis of infectious complications in 189 patients with lupus nephritis

Objective To investigate the clinical and epidemiologic features of infection in patients with lupus nephritis (LN). Methods The incidence, sites and pathogen species of infection 1 year after diagnosis in 189 patients with LN were retrospectively analyzed, and the relationships between the clinical, histological types, treatment programs and infection were analyzed. Results Among 189 LN patients (16 males and 173 females), 97 patients had infection (infection group), and 92 patients had not infection (non-infection group). The incidence of infection was 51.3% (97/189), while the incidence of nosocomial infection was 39.7%(75/189). In infection group, the patients had 133 infection episodes, and the principal infection sites were lung and urinary tract system, accounting for 67.7% (90/133); the principal pathogen was bacteria, accounting for 72.9%(97/133), and the second was virus, accounting for 17.3% (23/133). Different clinical or pathological types and treatment options of LN had different infection rates, but there were no statistical differences (P>0.05). The incidence of nosocomial infection in patients with glucocorticoid + cyclophosphamide double impact was significantly higher than that in patients treated with glucocorticoid + mycophenolate mofetil: 52.9% (45/85) vs. 7.7% (2/26), and there was statistical difference (P<0.01). One year after diagnosis, the mortality in infection group was significantly higher than that in non-infection group:8.2%(8/97) vs. 1.1%(1/92), and there was statistical difference (P<0.05). Conclusions The more serious the clinical and pathological types, the higher the incidence of infection. The incidence of infection is associated with the treatment options. Infection is still the most important reason for LN patients′death.
