2.Effects of Jian-Xing on three animal models to mimic muscle tremors of Parkinson's disease

ObjectiveTo develop three animal models to mimic muscle tremors of Parkinson's disease and observe effects of Chinese herb compounds Jian-Xing on these models.MethodsPure muscle tremor mice models were developed by single intraperitoneal injection of arecoline or oxotremorine. The mitochondria damaged model was developed by intraperitoneal injection of MPTP(1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine) 20mg/kg for 8 days. Duration time of tremoring was recorded. The climbing time and spontaneous movements of MPTP mice were recorded. HPLC was used to detect the content of dopamine and it's metabolites in MPTP mice.ResultsAfter injection of oxotremorine or arecoline,model mice exhibited significant tremoring, duration time of tremor of mice in Jian-Xing group shortened significantly. As to MPTP model mice, climbing time of model mice prolonged, times of spontaneous movements of model mice decreased and the content of dopamine in striaturn decreased compared with control group. Climbing time of mice in Jian-Xing group shortened, spontaneous movements increased and content of dopamine increased distinctively. ConclusionOxotremorine, arecoline and MPTP all can produce tremor animal models. Chinese herb compounds Jian-Xing can shorten the duration time of tremor.As to MPTP model, Jian-Xing can shorten the climbing time of model mice, increase the spontaneous movements and increase the content of dopamine in striaturn.
3.A DOUBLE LABELLING IMMUNOCYTOCHEMICAL STUDY OF NEUROTENSIN AND SUBSTANCE P IN DORSAL HORN OF SPINAL CORD IN RAT——A LIGHT AND ELECTRON MICROSCOPIC CORRELATIVE STUDY

Immunoreactivity of neurotensin(NT) and substance P (SP) were studied by means of PAP method with simultaneous immunocytochemical double staining on the same sections using diaminobenzidine (DAB) for SP and benzidinc dihydrochloride(BDHC) for NT as the chromogens for light and electron microscopy in dorsal horn of spinal cord of rat treated with colchicinc. In our hands the reaction products of DAB and BDHC is quite discernible, under LM the former is brown and later is blue; under EM DAB reaction product is homogeneous and diffuse electron opaque while BDHC reaction product is patch electron opaque, it is a new but simpler approach to demonstrate two antigens simultaneously in the same ultrathin section. It would be conformable further to the studies of morphological and functional relationships between different kinds of neurons. The results showed that NT-like immunoreactive (NT-LI) perikarya were mainly located in lamina Ⅱi and out layer of lamina Ⅲ and NT-LI terminals were mainly located in laminae Ⅰ-Ⅲ. Under EM the NT-LI axon terminals may synapse with the unlabeled axon or unlabeled dendrites. SP-like immunoreactive (SP-LI) perikarya were mainly located in lamina Ⅱ. The density of SP-LI terminals were higher in laminae Ⅰ-Ⅱ, while axo (SP)-axonic (SP), axo (SP)-somatic (SP) synapses were identified with EM. In double labelling sections, furthermore under EM, it was found that NT-LI axon terminal (BDHC patch like electron opaque reaction products)can synapse or contact with SP-LI axon (DAB diffuse electron opaque reaction products). Our results suggested that NT-and SP-containing neurons and terminals in dorsal horn might participate in regulating process of primary sensory transmission.
4.Serum B-type natriuretic peptide changes in patients with acute coronary syndrome during percutaneous coronary intervention

Objective To observe the serum B-type natriuretic peptide(BNP)changes in patients with acute coronary syndrome during percutaneous coronary intervention(PCI).Method The serum BNP level was determined and the heart function was assessed in 236 consecutive patients with acute coronary syndrome(ACS) admitted to CCU and in 54 normal normal suvjects as control from January 2005 to December 2006 in Tongji Hospital.The ACS patients were further divided into various subgroups according to the involved arteries and performance of PCI.Serum BNP,hypersensitive c respose protein(HsCRP)level,amd heart comstitution and function were analyzed.Results The serum BNP and HsCRP level were significantly increased in patients with ACS [(332.06?483.17)ng/L and(31.06?52.15)mg/L]more than those in normal subjects [(81.44?195.55)ng/L and(11.15?20.78)mg/L,respectively,P
5.Effect of Jintong Capsule on the Mice Models of Tourette Syndrome

Objective To observe effect of Jintong capsule on model mice of tourette syndrome (TS).Methods TS models of mouse were established by intraperitoneal injection of amphetamine (AMP) or subcutaneous injection of apomorphine (APO). The animals were randomly divided into the normal control group, model group, Jintong capsule large dose group, Jintong capsule small dose group and haloperidol control group. The spontaneous movements and grasping action of model animals were observed and the contents of dopamine and its products of metabolism in striatum of animals were detected.Results The abnormal hyperactivity and the content of dopamine of Jintong capsule groups in striatum of AMP model mice were lower than that of the model group ( P< 0.05 ), and the grasping time of Jintong capsule groups in APO model mice shorten compare with the control group ( P<0.05).Conclusion Jintong capsule can ameliorate hyperactivity behaviors of these two animal models, and decrease the content of dopamine in striatum.
6.Changes of proliferating kinetics in Plasmodium berghei of sodium artesunate-resistance

In the present paper, we measured DNA content of uninuclear Plasrnodium berghei traced with DAPI by means of fluoromi-crospectrophotmeter. The results indicate that the DNA replication of parasite was continuous and it's content was high polyploidy and phase G1 of proliferating cycle was not evident. The dispersion degree of distribution of DNA content in P. R. was markedly lower and the peak sitewas more concentrated and obviously shifted to the right compared with P. N. It was suggested that the speed of DNA replication and proliferating vitality of parasites after producing resistance to SA markedly decreased, indicating that the changes of proliferating kinetics of P. R. happened.
7.Comprehensive yield and quality comparison of Bupleurum chinense' s seeds of different harvesting time from Qingchuan County with Topsis approach.

In order to investigate the comprehensive quality differences of the seeds harvested in different growing time, and offer theoretical guide for the optimization of seeds' production technology, we analyzed the apparent size, 1 000-grain weight, water absorbency, germination indexes, postharvest embryo rate change, relatively contents of coumarins and the yield of single plant of its seeds of different harvesting time, and compared their comprehensive quality by Topsis analysis. The results showed that there existed obvious differences in yield and quality between seeds of 3 harvesting times. While the harvesting time postponed, the yield of single plant raised, and the shrunken seeds rate decreased, the quality of seedlings increased, while the contents of coumarins showed a steady increase, and the germination rate decreased. The comprehensive quality of the seeds harvested in the black ripe time rank the first place, followed by the brown ripe time and the yellow ripe time. As the harvesting time delays, the seeds' comprehensive quality increases, therefore, we could put off the seeds' harvesting time properly for the high efficient seed production.

8.Potential risk factors of excessive epistaxis after endoscopic endonasal surgery.

OBJECTIVE:
To investigate the potential risk factors and management of excessive epistaxis after endoscopic endonasal surgery (EES).
METHOD:
Six hundred and forty-one patients who underwent EES in our hospital from December 2011 to December 2012 were reviewed retrospectively. Factors which potentially affect the incidence of excessive epistaxis after EES were analyzed with univariate and multivariate logistic regression model.
RESULT:
The incidence rate of excessive epistaxis after EES was 8.4% in our study. Multivariate logistic regression analysis revealed that history of previous EES, along with other four factors, correlated significantly with the occurrence of excessive epistaxis after EES.
CONCLUSIONS
Previous EES, along with other three factors, may increase the chance of excessive epistaxis after EES, while pre-operative corticosteroid therapy may reduce the risk to some extent.

9.Study on antiinflammatory effect of a compound TCM agent containing ant extractive in animal models.

OBJECTIVETo study the antiinflammatory effect of a compound TCM (Traditional Chinese Medicine) agent on animal models. The agent contains ant extractive and a blent of three herbal products, herba epimedii, fructus cnidii, and fructus lycii.
METHODThree animal models to induce experimental inflammation in rats, including carrageenin--induced paw edema, cotton-ball granuloma and adjuvant induced arthritis, were chosen to study the antiinflammatory effect of the TCM agent.
RESULTThe TCM agent showed a marked inhibitory effect on edema induced by all three types of inflammation in rats, the inhibitory rate of the TCM agent at the dose of 0.20, 0.40 and 0.80 g.kg-1 in granuloma model bing over 25% at 1 hour post oral administration, and being 23.8%, 22.7%, 39.7% at 6 hour. In addition, the TCM agent also showed a significant preventive as well as therapeutic effect on adjuvant induced arthritis in rats, and improved the pathological changes of the animal joints with the induced arthritis.
CONCLUSIONTCM agent has significant antiinflammatory effects on the three above mentioned animal models.

10.Prognostic value of acute heart block after alcohol septal ablation in patients with hypertrophic obstructive cardiomyopathy

Objective To investigate the prognostic value of acute beart block (AHB) after percutaneous transluminal septal myocardial ablation (PTSMA) in patients with hypertrophic obstructive cardiomyopathy (HOCM). Methods Ninety-four HOCM patients underwent PTSMA were included in this study. Twelve-lead electrocardiograms were obtained during and post PTSMA. Association between clinic events and incidence of post-PTSMA AHB was analyzed. Results AHB was induced in 26 patients by PTSMA and disappeared in 11 patients shortly post PTSMA, subacute intraventricular conduction disturbances was seen in 11 (42.3%), subacute I°AVB in 2(7.7% ) and subacute Ⅲ°AVB in another 2 (7.7%) patients. Among 68 patients without AHB during PTSMA, intraventricular conduction disturbances was evidenced in 14 patients (20.6% ), I°AVB in 2 (2.9% ) and Ⅲ°AVB in 1 patient (1.5%) after PTSMA. AHB patients with subacute heart block were associated with poor prognosis (conduction block duration was 42.00 h) while patients without AHB was associated with benign prognosis even with new onset of subacute heart block (conduction block duration was 7.33 h, P <0.01). Conclusion Patients with AHB during PTSMA are at higher risk for subacute heart block, especially intraventricolar conduction disturbances. AHB patients with subacute heart block were associated with poor prognosis and longer recovery time conducting system.
