A highly phytase-producing strain B.licheniformis LL8 was obtained by several mutagenesis of UV with B.licheniformis as starting strain.The new strain produced about two folds of phytase activity as compared with the starting strain. The production performance of the strain was stable. The cultivation conditions were optimized by single factor and orthogonal experiment. When the mutant B.licheniformis LL8 was cultivated at 55℃, initial pH 7.5 with the inoculation size of 10% for 30h in WBE medUm, the phytase activity was up to 2268.4U/mL.

Objective:To construct pEGFP-C3-B7.2-MAGE-1 eukaryotic expression plasmids and to study the immunological effects of esophageal cancer cell vaccine modified by human B7.2/MAGE-1 gene.Methods:The B7.2 and MAGE-1 cDNA,which was amplified by RT-PCR, and the eukaryotic expression plasmid pEGFP-C3-B7.2-MAGE-1 was constructed. Objective To investigate the distribution of pathogenic bacteria,drug sensitivity,and the relationship between drug sensitivity and Escherichia coli(E.coli) induced enzymes in childhood diarrhea in the last 2 years in Chongqing area,so as to provide important evidence for pediatric clinical therapy.Methods Thirty-one E.coli induced enzymes,extended spectrum ?-laetamases(ESBLs),cephalosporinase(AmpC)detected in different phenotype methods,and drug sensitivity was measured in paper strip method,and the specimens were collected from children′s hospital affiliated to chongqing university of medical sciences from Jan.2005 to Dec.2006 were determined.Among the total,there were 18 enteropathogeic E.coli(EPEC) strains,8 enterotoxigenic E.coli(ETEC) strains and 5 enteroinvasive E.coli(EIEC) strains.In addition,drug resistance tests by paper strip included chloramphenicol(CHL),amikacin(AMK),gentamicin(GEN),norfloxacin(NOF),ciproflocacin(CIP),cefazolin(CEZ),cefoperazone(CPZ),ceftriaxone(CRO),ceftazidime(CAZ),cefotacime(CTX),cefepime(FEP),imipenem(IPM).SPSS 12.0 software was used to analyze the data.Results Three point two percent of the 31 E.coli were drug resistant to IPM,and 35.5%,38.7% to NOF,CIP individually,but more than 60% to AMK,GEN,even more than 67.7% towards cephalosporin(except ceftazidime and cefepime);the gross enzyme-produced rate was 87.1%,rate of single ESBLs,AmpC,and induction of both enzymes simultaneously presented 64.5%,6.5%,16.1% respectively;and there was marked difference in drug resistance when bacteria that produced single AmpC versus bacteria that produced single ESBLs or that produced both ESBLs and AmpC(Pa﹤0.05).Conclusions The relationships among enzyme′s quantity,sort and bacterial resistance are different.These data show E.coli infected by bacterial diarrhea children in Chongqing due to a high rate of induced enzymes,and their drug resistance vary according to the state of induced enzymes.

Objective To study the mechanism of pregnancy immune tolerance induced by adoptive transferring of FasL gene-modified dendritic cells(DC). Methods The mouse models of spontaneous abortion(CBA/J ? DBA/2) and normal pregnancy(CBA/J ? BALB/c) were established.Five different experimental groups were included: mice of normal pregnancy(CBA/J?BALB/c)(n=17),served as control group;mice of spontaneous abortion without treatment(CBA/J?DBA/2)(n=37),mice injected with DC culture medium(DCCM)(n=25);mice immunized with empty plasmid pcDNA3.1-DC(n=6);and mice immunized with pcDNA3.1-FasL-DC(n=5).Embryo resorption rates of pregnant mice in each groups were observed.Annexin V-FITC was used to detect the apoptosis of T cells.Immunohistochemical staining(SABC) was used to detect the expression of FasL in decidual membranes of pregnant mice. ResultsThe embryo resorption rate of mice immunized with FasL-DC was decreased significantly as compared with that of mice of spontaneous abortion without treatment,DCCM group and immunized with pcDNA3.1-DC(P0.05). Conclusion The decreased apoptosis rate of peripheral T cells and the reduced expression of FasL in decidual membranes may be an important mechanism for the pathogenesis of abortion.Adoptive transfer of FasL gene-modified DC may induce pregnancy immune tolerance by increasing FasL expression of maternal-fetal interface and decreasing embryo resorption rate.

OBJECTIVETo explore the effect of Renshen Jianxin Capsule (RJC) on insulin resistance in patients with coronary heart disease (CHD) and glucose tolerance impairment (GTI).

METHODSEighty patients with CHD-GTI of qi-deficiency blood-stasis syndrome were randomly assigned to 2 groups equally, the treated group was treated by RJC and the control group by metformin, based on the conventional Western medical treatment with nitric esters for 20 weeks. Changes before and after treatment in clinical symptoms and levels of blood glucose insulin, and insulin sensitivity index (ISI) were observed.

RESULTSThe scores of clinical symptoms of Chinese medicine decreased in both groups, which showed statistical significances compared with those before treatment (P<0.01, P<0.05). On alleviating the angina pectoris, the markedly effective rate of the treated group is 47.5%, the total effective rate was 80.0%, and the difference between the two groups showed statistical significance (P<0.05). FBG, INS and ISI were improved significantly in both groups after treatment (P<0.05, P<0.01); while the three indices showed in significant difference between the two groups after treatment (P> 0.05).

CONCLUSIONRJC was effective in improving insulin resistant, which may be one of the mechanisms of its therapeutic effect on CHD.
