1.Rhabdomyolysis Associated with Viral Encephalitis

Viral encephalitis is infectious diseases caused by many kinds virus which infects the brain and produces a series of clinical manifestations,it is an common disease in children.Rhabdomyolysis is a sort of syndrome characterized by striated muscle necrosis and the release of intracellular muscle constituents into the circulation,most notably high level of creatine phosphokinase and myoglobin,and myoglobi-nuria,which is often complicated with acute renal failure.It was reported that rhabdomyolysis could be associated with many viral encephalidities,the clinician should pay attention to this condition because it was not rare especially,when hematuria appearred in the course of viral encephalidities,serum creatine phosphate kinase,serum and urine myohemoglobin should be detected to identify whether rhabdomyolysis was existed.Once the diagnosis was confirmed,it should be treated with fluid administration and basification in order to avoid acute renal failure,and to improve prognosis on these patients.This study reviewed the literature on viral encephalidities and rhabdomyolysis.
2.Central Nervous System Damage in Children with Mycoplasma Pneumoniae Pneumonia and Its Mechanism

Objective To observe the central nervous system(CNS)damage in children with mycoplasma pneumoniae pneumonia(MPP)and explore its mechanism.Methods A total of 325 cases of MPP were selected to investigate the incidence of CNS damage,and the levels of IL-8,IL-18 and soluble interleukin-2 receptor(sIL-2R)in both serum and cerebrospinal fluid(CSF)were detected in those patients with CNS complications.Children with viral encephalitis at hospital during the same period were taken as control group.Use SPSS 12.0 software to analyze the data.Results There were 22 cases(6.8%)of MPP who had CNS complications,presented mainly with fever,vomiting,headache and convulsion.The levels of IL-8,IL-18 and sIL-2R were higher in MPP patients who had CNS complications than those in control group,especially the level of IL-18 and sIL-2R in CSF(Pa

ObjectiveTo investigate the effect of all-trans retinoic acid (atRA) on the renin-angiotensin system (RAS) in 5/6 renal ablation model. MethodsAtRA was administered to 5/6 renal ablation rats by three dosages: 5 mg·kg-1·d-1 (n=8), 10 mg·kg-1d-1 (n=8) and 20 mg·kg-1 d-1 (n=8) and vehicle (vehicle group, n=8) for 10 weeks. Healthy rats consisted of shamoperation group (n =8). The level of renin and angiotensin Ⅱ in renal tissues were measured by radioimmunoassary. The level of angiotensin type 1 receptor (AT1R) in remnant renal cortex was measured by Western blot. The mRNA expression levels of two subunits of activative protein 1(AP-1),c-jun and c-fos was quantitated by real-time PCR. ResultsAfter 10 weeks of atRA treatment by gavarge, artery blood pressure decreased (P<0.05). AtRA reduced the levels of renin (P<0.05) and angiotensin Ⅱ (P<0.05) in kidney and down-regulated the expression of AT1R protein in renal cortex. Larger dose of atRA (20 mg·kg-1·d-1) performed higher activity in inhibiting renin and AT1R. Compared with vehicle group, atRA could significantly inhibit the expression of renal c-jun and c-fos mRNA (P<0.05). Conclusion atRA can decrease the over-expression of main components of RAS.
4.Research on Quality Differences in Benzalkonium Chloride from Domestic and Abroad

Objective:To compare the quality differences in benzalkonium chloride samples from domestic and abroad to provide references for the quality standard revision for benzalkonium chloride in Chinese pharmacopoeia. Methods: The ratio of alkyl compo-nents was determined according to the method described in USP 38, and the total content of benzalkonium chloride was determined ac-cording to the method recorded in Chinese Pharmacopoeia (2015 edition) and USP 38, respectively. Results:According to the results of composition ratio of alkyl, the fraction defective of domestic samples and imported samples was 100% and 50%, respectively. The content difference between the values calculated by the methods in the two pharmacopoeias showed that the total content of domestic samples changed from 3. 86% to 4. 15%, and that of imported samples changed from 1. 15% to 3. 90%. Conclusion:There are sig-nificant differences in the quality of benzalkonium chloride between domestic samples and imported samples. It is recommended that the ratio of alkyl components should be supplemented in our pharmacopoeia referring to the method in USP 38 and the total content calcula-tion formula for benzalkonium chloride should be revised to improve the quality standard for benzalkonium chloride.
5.Effects of 2,3,5,4'-tetrahydroxy stilbene-2-O-β-D-glucoside on water content and the metabolism of free radicals in brain of ischemia mice

ObjectiveTo explore the mechanism of protection of 2,3,5,4'-tetrahydroxy stilbene-2-O-β-D-glucoside(TSG) on brain.MethodsThe bilateral carotid arteries of mice were occluded and then reperfused. The mice were sacrificed to get the brain tissue. Brain water content(BWC) was assayed by dry-wet method; malondialdehyde (MDA) content was determined by thiobarbiturate method and the activity of superoxide dismutase(SOD) was estimated by nitrite salt assay.ResultsBWC of model group was significantly higher than that of sham-operation group, while the TSG treatment reduced the BWC remarkably in mice with ischemia-reperfusion. SOD activity of model group was significantly lower than that of sham-operation group, while MDA content increased remarkably. Comparing with the model group, the SOD activity of mice treated with TSG was higher and the MDA content was lower.ConclusionTSG may reduce brain edema, improve the anti-oxidative ability of the brain tissue, and therefore, have protective effects on the brain.
6.Progress on the application of artificial intelligence technology in ligand-based and receptor structure-based drug screening

Artificial intelligence technology is being widely applied in drug screening. This paper introduces the characteristics of artificial intelligence, and summarizes the application and progress of artificial intelligence technology especially deep learning in drug screening, from ligand-based and receptor structure-based aspects. This paper also introduces how to apply artificial intelligence to drug design from these two aspects. Finally, we discuss the main limitations, challenges, and prospects of artificial intelligence technology in the field of drug screening.
7.Development of HTS model on SERT inhibitors combined biological screening model with HTVS.

In order to improve the efficiency of drug screening on serotonin transporter (SERT) inhibitors, a high-throughput screening (HTS) model is established in RBL-2H3 cells. The RBL-2H3 cells are very similar to the serotonin genetic neuro, in modulation of post-receptor mechanisms and transduction pathway of SERT reactivated. Depending on a fluorescence substrate ASP+ used in detection method of inhibitor rates, it's convenient, quick, accurate and effective, not making the environmental biohazard compared with radioactive experiments. Furthermore, biological screening model combined with computer aided virtual screening technique describing high-throughput virtual screening (HTVS). Bayesian classification method and molecular fingerprint similarity were applied to virtual screening technique, for screening compounds in compound library. Some compounds have been found, and then validated further by biological screening model. Combination of HTS and HTVS improves the efficiency of screening SERT inhibitors.

10.Absorption of extractive Polygonum orientale in rat everted gut sacs.

Using in vitro everted gut to investigate the intestinal absorption of the extracts from Polygonum orientale at different concentration. UPLC-MS/MS was used to detect the content of protocatechuic acid, isoorientin, orientin, vitexin, cynaroside, quercitrin, kaempferol-rhamnoside in different intestinal segments, then compared the results with the absorption of chemical components of extractive P. orientale in each intestinal segments, and calculated the absorption parameter. We took the statistic analysis with SPSS statistic software. The influence significance of each factors were analyzed to describe the character of absorption. The absorption of each component is linearity in different intestinal segments and different dose, and the square of coeficient correlation exceed 0.95, which consistent with zero order rate process. The K(a) increase along with the raised dosage of the extractive P. orientale (R2 > 0.95), indicated it is the passive absorption; different intestinal segments have different absorption. And the absorption trend in intestinal is duodenum, jejunum, ileum are greater than the colon. As ingredients are selectively absorbed in intestinal sac, the everted intestinal sac method is selected to assess the intestinal absorption charcteristics of ingredients of extractive P. orientale.
