1.Relationship between serum parathyriod hormone level and renal anemia in patients with chronic kidney dis-ease

Objective To explore the relationship of serum parathyriod hormone ( PTH) level with renal ane-mia in patients with chronic kidney disease (CKD).Methods Serum levels of PTH,Hb,Hct,BUN,Cr were observed in 126 patients with CKD at stages 3-5,the changes of 42 patients in maintenance hemodialysis(MHD) with recombi-nant human erythropoietin (rhEPO) and calcitriol-1.25(OH)2D3 treatment for three months were determined. Results The serum PTH level was significantly increased in CKD patients at stage 3 and more in CKD patients at stage 4 and 5.There were positive relationship between blood PTH and BUN ,Cr(P<0.01),but a negative relation-ship between PTH and Ccr,Hb(P <0.01).42 patients of MHD treated with rhEPO,1.25(OH)2D3 after three months,according to the changes of HCT ,the effective rate was 73.8% and the ineffective rate was 26.2%.The PTH,Hb,HCT of the effective group had statistically significant differences compared with the ineffective group ( all P<0.01).Hb and Hct significantly increased and PTH significantly decreased in the effective group .Conclusion CKD patients had decreased renal function ,elevated blood PTH .High serum PTH might aggravate renal anemia and fail to respond to rhEPO.The high serum PTH corrected by using 1.25(OH)2D3 might improve renal anemia,with high plasma PTH correcting .
2.Evaluation of effect of behavioral intervention of secondary prevention on stroke

Objective The purpose of this study was to describe the effect of behavioral intervention of secondary prevention on stroke. Methods 204 patients with stroke were divided into the observation group and the control group with 102 patients in each group at random. The health records were set up and self-designed scale for assessment of risk factors of recurrence of stroke was used to assess the patients' general conditions. Individual intensified education and behavior intervention were adopted in the observation group. Routine treatment and health education were used in the control group. The collected data before intervention, the third and 6th month after intervention were evaluated. The medication compliance, limbs rehabilitation exercises, bad living style, coming to hospital at regular intervals and recurrence rate of stroke were compared between the two groups. Results The observation group was superior to the control group in the following aspects, the mastering rate of disease knowledge, changes in bad living style and therapy compliance (P<0.01). Conclusions Behavioral intervention of secondary prevention could effectively reduce the recurrence of stroke and improve the living quality of patients.
3.Relationship of human microvascular endothelial cell activation and adenosine level under the hypoxia

Background Retinal hypoxia is one of primary causes of retinal neovascularization,and its mechanism is research hot topic.Studies showed that hypoxia stimulates the activation of many trancripation factors and vascular endothelial cells,which leads to angiogenesis.Besides to the vascular endothelial growth factor,the effect of adenosine on angiogenesis is increasingly concerned.Objective This study was to invastigate the relationship of biological behaviour of human microvascular endothelial cells (HMEC-1) under the hypoxia and adenosine,and to explore the effect of adenosine on angiogenesis under hypoxia.Methods HMEC-1 cell line was cultured in vitro,and the cells were divided into normoxia group and hypoxia group.The cells in the normoxia group were cultured under the 5% CO2 environment,and those in the hypoxia group were cultured under the 1% O2,94% N2 and 5% CO2 environment.The proliferation ability and percentage of the cells were assayed by cell counting kit-8 (CCK8) and EDU.The migration number and invasive number of the cells were detected by transwell chamber.The expressions of CD39 and CD73 proteins in the cells were tested by Western blot and immunofluorescence technique,and the level of adenosine was measured by high performance liquid chromatography (HPLC).Results The proliferation values (absobancy) were 0.715-±0.067 and 0.821 ±0.056 in the normoxia group and the hypoxia group in 12 hours after culture,and those in 24 hours were 0.946±0.028 and 0.998±0.028,showing significant increase in the hypoxia group compared with the normoxia group (t12h =3.805,t24h =3.222,all at P < 0.01).The precentage of the proliferation in the hypoxia group was evidently higher than that in the normoxia group (t =-6.868,P<0.01).The number of the cell migration and invasion in 24 hours after culture was 185.3 ± 10.594 and 74.2± 10.741 respectively in the normoxia group,and that in the hypoxia group was 300.7±22.853 and 107.5±7.007,with significant differences between the two groups (t=-12.124,-6.367,both at P<0.01).The expression levels of CD39 and CD73 proteins in the hypoxia group were significantly higher than those in the normoxia group in bothl2 hours and 24 hours after culture(all at P<0.05),and the adenosine content in the cells is gradually increased in 2,6,12,24 and 36 hours after culture,with the highest content in 36 hours.The adenosine content was significantly higher in the hypoxia group than in the normoxia group at various time points (t2h =2.469,P =0.017;t6h =5.442,P<0.001;t12h =3.841,P<0.001;t24h =4.458,P<0.001;t36h =2.757,P =0.008;t48h =3.319,P =0.002).Conclusions Compared with the normoxia group,the proliferation,migration and invasion abilities of HMEC-1 are stronger,meanwhile,the expression of CD39 and CD73 as well as the adenosine level in the cells are all increased under the hypoxic condition.It is suggested that the activation of human microvascular endothelial cells might be significantly related to the level of adenosine and its key enzymes.
4.Effects of Jian-Xing on three animal models to mimic muscle tremors of Parkinson's disease

ObjectiveTo develop three animal models to mimic muscle tremors of Parkinson's disease and observe effects of Chinese herb compounds Jian-Xing on these models.MethodsPure muscle tremor mice models were developed by single intraperitoneal injection of arecoline or oxotremorine. The mitochondria damaged model was developed by intraperitoneal injection of MPTP(1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine) 20mg/kg for 8 days. Duration time of tremoring was recorded. The climbing time and spontaneous movements of MPTP mice were recorded. HPLC was used to detect the content of dopamine and it's metabolites in MPTP mice.ResultsAfter injection of oxotremorine or arecoline,model mice exhibited significant tremoring, duration time of tremor of mice in Jian-Xing group shortened significantly. As to MPTP model mice, climbing time of model mice prolonged, times of spontaneous movements of model mice decreased and the content of dopamine in striaturn decreased compared with control group. Climbing time of mice in Jian-Xing group shortened, spontaneous movements increased and content of dopamine increased distinctively. ConclusionOxotremorine, arecoline and MPTP all can produce tremor animal models. Chinese herb compounds Jian-Xing can shorten the duration time of tremor.As to MPTP model, Jian-Xing can shorten the climbing time of model mice, increase the spontaneous movements and increase the content of dopamine in striaturn.
5.Effects of 2,3,5,4'-tetrahydroxy stilbene-2-O-β-D-glucoside on water content and the metabolism of free radicals in brain of ischemia mice

ObjectiveTo explore the mechanism of protection of 2,3,5,4'-tetrahydroxy stilbene-2-O-β-D-glucoside(TSG) on brain.MethodsThe bilateral carotid arteries of mice were occluded and then reperfused. The mice were sacrificed to get the brain tissue. Brain water content(BWC) was assayed by dry-wet method; malondialdehyde (MDA) content was determined by thiobarbiturate method and the activity of superoxide dismutase(SOD) was estimated by nitrite salt assay.ResultsBWC of model group was significantly higher than that of sham-operation group, while the TSG treatment reduced the BWC remarkably in mice with ischemia-reperfusion. SOD activity of model group was significantly lower than that of sham-operation group, while MDA content increased remarkably. Comparing with the model group, the SOD activity of mice treated with TSG was higher and the MDA content was lower.ConclusionTSG may reduce brain edema, improve the anti-oxidative ability of the brain tissue, and therefore, have protective effects on the brain.
6.Application of molecular absorbent recycling system in auto liver transplantation

Objective To investigate the application of molecular absorbent recycling system (MARS) in auto liver transplantation (ALT).Methods The clinical data of 46 patients who underwent ALT in Zhongnan Hospital of Wuhan University from September 2014 to August 2015 were retrospectively studied.The patients were randomly divided into the MARS group (n =30) and the hemodialysis group (n =16).In the MARS group,20 patients were male and 10 were female with an average age of (15 ±67) years,and the median age was 46.In the hemodialysis group,8 were male and 8 were female with an average age of (22 ± 54) years.A heparinized left iliac vein was linked to a centrifugal pump of a MARS and an output tube was linked to a left jugular vein.Before the anhepatic phase,physical pretreatment was carried out by ligating the porta hepatis and inferior vena cava (IVC) to adapt the patient to the anhepatic phase.Subsequently,a bypass was established followed by ligation of the IVC.The duration of operation,length of stay in ICU after operation,one-year survival rate and hepatorenal function were studied.Results The duration of operation in the MRAS group and in the hemodialysis group was not significantly different (P >0.05).The length of stay in ICU after operation in the MARS group was (12.0 ±3.0) d,which was significantly shorter than that of the hemodialysis group (20.0 ± 2.0) d (P < 0.05).The one-year survival rate in the MARS group was (100%),which was significantly higher than that of the hemodialysis group (93.8%,P <0.05).Conclusion The application of MARS in ALT shortened the length of stay in hospital and improved one-year survival rate,resulting in better outcomes than the conventional method.
7.Weekly dosage of docetaxel combined with cisplatin and 5-fluorouracil in the treatment of advanced gastric cancer.

OBJECTIVETo observe the effects and toxicity of docetaxel given once weekly combined with cisplatin and 5-fluororacil in the treatment of advanced gastric cancer (AGC).
METHODSThirty-one patients with AGC were given docetaxel at 35 mg/m square by intravenous infusion completed in 1 h on the 1st, 8th and 15th days. Cisplatin was given at 20 mg/m square for 4 h between the 2nd and 6th days, during which time 5-fluororacil was also given at 750 mg/m square for 24 h. One treatment cycle lasted for 28 days and 2 consecutive cycles were administered.
RESULTSIn the 31 patients available for therapeutic effect evaluation, the overall effective rate was 48.4%. The effective rate in patients without previous treatments was 64.7% (CR in 2 cases), and 28.6% in the retreated patients. The median TTP, median survival time and one-year survival rate were 6 months, 11 months and 41.9%, respectively. The main toxic reactions of the combined treatment were myelosuppression, gastrointestinal tract reaction and alopecia of mostly grade 1 or 2.
CONCLUSIONWeekly dosage of docetaxel combined with cisplatin and 5-fluororacil is an effective regimen for AGC with only mild toxic reactions and good patient tolerance.

Objective To explore the feasibility and superiority of the simple controllable aerocyst pressurization wrapped-up bondage for battlefield first aid in the future high tech local war. Methods The disposable infusion bag is wrapped up in the bottom of the sling in stead of partial surgical dressing. Sphygmomanometer, pressure gauge and heparin hat are connected. Inflate and pressurize the sphygmomanometer and keep the pressure between 3kPa and 5kPa. Results The structure of this bandage is simple and can be operated easily. The pressure is controlled nimblely and conveniently. Except for the accurate effect, it is versatile and inexpensive. In addition, it can be employed in the bone fracture.
10.Effects of Taurine on Oxidative Stress and Calcium Overload Induced by Cerebral Cortex Contusion in Rats

OBJECTIVE:To study the effects of taurine on oxidative stress and calcium overload induced by cerebral cortex contusion.METHODS:SD rats were randomly divided into normal control group,brain contusion model group,taurine groups(high dose,middle dose and low dose respectively),and nimodipine group.After being fed with corresponding drugs for7days,all rats were subjected to modeling by brain contusion.For intracellular calcium detection,rats were sacrificed2h after modeling,and the brain slices were prepared to fluorescence labeling and confocal microscopy detection.For the detection of oxidative stress,rats were sacrificed24h after modeling,the cortex of contusion side was homogenated and then the activity of superoxide dis?mutase(SOD)and content of malondialdehyde(MDA)were detected through biochemical method.RESULTS:Compared with model group,all taurine groups were shown to have markedly less MDA and intracellular calcium content,and the high dose group had markedly stronger SOD activity.CONCLUSION:Taurine is effective in counteracting the oxidative stress and calcium overload caused by brain contusion.
