Chinese Journal of Postgraduates of Medicine 2012;35(19):25-27
Objective To investigate the clinical significance of the detection of the levels of serum uric acid (UA) and serum eytokine tunor necrosis factor-oα (TNF-α),soluble intercellular adhesion molecule-1 (sICAM-1),interleukin (IL)-6 and IL-8 in patients with chronic obstructive pulmonary disease (COPD).Methods UA of 40 healthy people (control group) and 158 COPD patients (COPD group) in acute exacerbation and remission stage was detected respectively by automatic biochemical analyzer, and the levels of serum TNF-α,sICAM-1,IL-6 and IL-8 were detected by ELISA.The percentage of first second forced expiratory volume in expected value (FEV1%) of COPD patients in acute exacerbation and remission stage were detected. Results TNF-α,sICAM-1,IL-6,IL-8 and UA of COPD group in acute exacerbation were significantly higher than those of control group (P< 0.01 ),and FEV1% was significantly lower than that of control group (P< 0.01 ).TNF-α,sICAM-1 and UA of COPD group in remission stage were significantly higher than those of control group (P <0.01 or <0.05),IL-6 and IL-8 had no statistical significance compared with those of control group (P > 0.05),and FEV1% was significantly lower than that of control group (P < 0.01 ).Correlation analysis showed that TNF- α,sICAM-1,IL-6,IL-8,UA of COPD patients in acute exacerbation were negatively conrelated with FEV1% (r =-0.681,-0.579,-0.545,-0.652,-0.357,respectively,P < 0.01 or < 0.05 ).In remission stage,TNF- α,sICAM-1,IL-6 and IL-8 of COPD patients were negatively correlated with FEV1% (r =-0.623,-0.335,-0.349,-0.623,P<0.01 or <0.05),and UA had no significant correlation with FEV1% (r =-0.092,P >0.05).Conclusions The serum UA and cytokines TNF-α,sICAM-1,IL-6,IL-8 are involved in the progression of COPD and correlated with lung function. They can be used as the judge indexes of COPD.

7.Rapid Content Determination and Pharmacokinetic Study of Gliquidone in Human Plasma by HPLC
China Pharmacist 2014;(11):1843-1845
Objective:To develop a simple and rapid HPLC method to determine the concentration and pharmacokinetics of gliqui-done in human plasma. Methods: Acetonitrile was used to precipitate plasma proteins directly and gliquidone was detected by an HPLC-fluorescence assay. The mobile phase consisted of acetonitrile-20 mmol·L-1 biphosphate potassium (80∶20, v/v, pH 2. 5). The flow rate was 1. 0 ml·min-1 . The excitation wavelength was 315 nm and the emission wavelength was 410 nm. Results:The line-arity of gliquidone in plasma was good from 0. 034 to 2. 2 μg·ml-1 and the quantitative lower limit was 0. 034 μg·ml-1 . The range of method recovery was 99. 23%-108. 11%, and that of extraction recovery was 85. 78%-88. 77%. The intra-and inter-day precision was both less than 10%. Both specificity and stability met the requirements. Gliquidone in plasma was determined using the above method after a single oral dose of 60 mg gliquidone tablets in 20 healthy volunteers, and then the pharmacokinetic parameters were ana-lyzed. The variation coefficients were close to or more than 50%. Conclusion:The method is simple, rapid and sensitive, and suit-able for the detection of gliquidone in a large quantity. Individualized administration of gliquidone should be considered due to the obvi-ous individual difference in pharmacokinetics.

Objective To study the effect of calcitriol combined with metformin in the treatment of polycystic ovary syndrome(PCOS)and its influence on serum monocyte chemotactic factor -1(MCP -1).Methods 96 patients with PCOS were randomly divided into the control group(48 cases)and the the observation group(48 cases). The two groups were treated with metformin tablets,0.25g/time,2 times/d.On the basis of the above treatment, the observation group was given calcitriol,0.25μg/time,1 time /d.The two groups were treated for 3 months. Before and after treatment, the number of common symptoms of PCOS(menstrual scarce, acne, hairy, ovarian volume increase, polycystic change)and the determination of serum MCP -1 changes were summarized. Results Before treatment, the menstruation, acne, hairy, ovarian volume increase, polycystic change in control group were 46 cases,21 cases,32 cases,19 cases, 47 cases, respectively, which were 18 cases,11 cases,18 cases,9 cases and 10 cases after treatment. Before treatment, the menstruation, acne, hairy, ovarian volume increase, polycystic change in observation group were 47 cases,25 cases, 29 cases,22 cases,45 cases, respectively, which were 10 cases,6 cases,10 cases,4 cases and 5 cases after treatment. After treatment, the two groups were treated with less menstruation, acne, hair, ovarian volume and polycystic change (χ2 =9.86,7.25,9.06,10.17,13.42;14.28,12.62,11.28,16.52,19.66,all P <0.05).And after treatment, which of the observation group were significantly better than the control group (χ2 =4.06,4.86,5.62,4.71,9.27,all P <0.05).The serum MCP -1 levels of the control group before and after treatment were (159.64 ±27.12)μg/L, (118.54 ±21.38)μg/L,which of the observation group were (162.57 ±29.06)μg/L,(82.59 ±10.72)μg/L.After treatment, the serum levels of MCP -1 in the two groups were significantly decreased(t =2.968 2,4.163 8,all P <0.05),and the serum MCP -1 level in the observation group was significantly lower than that in the control group (t =2.661 2,P <0.05).Conclusion Calcitriol combined with metformin can significantly improve the efficacy of patients with PCOS,and decrease the expression of MCP -1 in serum.

China Pharmacy 2001;0(07):-
OBJECTIVE: To study the pharmacokinetics and bioavailability of pulse controlled-release tablets of ketoprofen(KRT) in dogs.METHODS: A total of 6 dogs were randomly assigned to receive KRT(trial group,n=3) or KP enteric-coated capsules(control group,n=3) 150 mg,then a crossover trial was conducted 1 week later.Plasma samples were taken at different time points and the plasma concentration of ketoprofen in dogs was determined by HPLC.The pharmacokinetic parameters of the two KP preparations were computed and the correlation between absorption percentage in vivo and release rate in vitro was compared.RESULTS: The pharmacokinetic parameters of KRT versus those of its control preparation were as follows: tmax:(5.00?0.41) vs.(3.75?0.00) h;Cmax:(15.40?1.94) vs.(20.73?1.72) ?g?mL-1;AUC0~t(101.33?17.49) vs.(93.21?25.03) ?g?h?mL-1.The relative bioavailability of the trial preparation was 108.71%.The test preparation showed a good correlation(r=0.987) between absorption percentage in vivo and release rate in vitro.CONCLUSIONS: The KRT and KP enteric-coated capsules were bioequivalent.
