Chinese Journal of Postgraduates of Medicine 2012;35(19):25-27
Objective To investigate the clinical significance of the detection of the levels of serum uric acid (UA) and serum eytokine tunor necrosis factor-oα (TNF-α),soluble intercellular adhesion molecule-1 (sICAM-1),interleukin (IL)-6 and IL-8 in patients with chronic obstructive pulmonary disease (COPD).Methods UA of 40 healthy people (control group) and 158 COPD patients (COPD group) in acute exacerbation and remission stage was detected respectively by automatic biochemical analyzer, and the levels of serum TNF-α,sICAM-1,IL-6 and IL-8 were detected by ELISA.The percentage of first second forced expiratory volume in expected value (FEV1%) of COPD patients in acute exacerbation and remission stage were detected. Results TNF-α,sICAM-1,IL-6,IL-8 and UA of COPD group in acute exacerbation were significantly higher than those of control group (P< 0.01 ),and FEV1% was significantly lower than that of control group (P< 0.01 ).TNF-α,sICAM-1 and UA of COPD group in remission stage were significantly higher than those of control group (P <0.01 or <0.05),IL-6 and IL-8 had no statistical significance compared with those of control group (P > 0.05),and FEV1% was significantly lower than that of control group (P < 0.01 ).Correlation analysis showed that TNF- α,sICAM-1,IL-6,IL-8,UA of COPD patients in acute exacerbation were negatively conrelated with FEV1% (r =-0.681,-0.579,-0.545,-0.652,-0.357,respectively,P < 0.01 or < 0.05 ).In remission stage,TNF- α,sICAM-1,IL-6 and IL-8 of COPD patients were negatively correlated with FEV1% (r =-0.623,-0.335,-0.349,-0.623,P<0.01 or <0.05),and UA had no significant correlation with FEV1% (r =-0.092,P >0.05).Conclusions The serum UA and cytokines TNF-α,sICAM-1,IL-6,IL-8 are involved in the progression of COPD and correlated with lung function. They can be used as the judge indexes of COPD.

Objective To detect the expression of insulin-like growth factor Ⅱ mRNA-binding protein 3 (IMP3) and p16 in cervical carcinoma and investigate their clinical significance. Methods Immunohistochemical Envision method was used to detect the expression of IMP3 and p16 proteins in 67 cases of carcinoma of the uterine cervix,58 cases of cervical intraepithelial neoplasia (CIN) and 15 cases of normal cervical epithelium. Results IMP3 was mainly dedected in the cytoplasm, whose positive expression rate was 85.07% (57/67) in cervical carcinoma,12.07% (7/58) in CIN (x2 =66.32,P < 0.05) and 0 (0/15) in cervical epithelium (x2 =44.38,P < 0.05).No significance variations of the expression of IMP3 were observed in cervical adenocarcinoma and squamous cell carcinoma (x2 =0.35,P > 0.05).p16 was found both in nucleus and cytoplasm of cervical carcinoma, whose expression rate was 83.58% (56/67),higher than 41.37% (24/58) in CIN (x2 =24.85,P < 0.05) and 0 (0/15) in cervical epithelium (x2 =42.61,P < 0.05).Conclusion Expression of IMP3 and p16 is high in cervical carcinoma, maybe can be used as a potential biomarker of metastasis and progression.
5.Rapid Content Determination and Pharmacokinetic Study of Gliquidone in Human Plasma by HPLC
China Pharmacist 2014;(11):1843-1845
Objective:To develop a simple and rapid HPLC method to determine the concentration and pharmacokinetics of gliqui-done in human plasma. Methods: Acetonitrile was used to precipitate plasma proteins directly and gliquidone was detected by an HPLC-fluorescence assay. The mobile phase consisted of acetonitrile-20 mmol·L-1 biphosphate potassium (80∶20, v/v, pH 2. 5). The flow rate was 1. 0 ml·min-1 . The excitation wavelength was 315 nm and the emission wavelength was 410 nm. Results:The line-arity of gliquidone in plasma was good from 0. 034 to 2. 2 μg·ml-1 and the quantitative lower limit was 0. 034 μg·ml-1 . The range of method recovery was 99. 23%-108. 11%, and that of extraction recovery was 85. 78%-88. 77%. The intra-and inter-day precision was both less than 10%. Both specificity and stability met the requirements. Gliquidone in plasma was determined using the above method after a single oral dose of 60 mg gliquidone tablets in 20 healthy volunteers, and then the pharmacokinetic parameters were ana-lyzed. The variation coefficients were close to or more than 50%. Conclusion:The method is simple, rapid and sensitive, and suit-able for the detection of gliquidone in a large quantity. Individualized administration of gliquidone should be considered due to the obvi-ous individual difference in pharmacokinetics.

Journal of Chongqing Medical University 1986;0(04):-
Objective:To observe the effect of GSK-3? on the cell cycle and metastasis of human gastric carcinoma cell. Methods:Influencing the GSK-3? activity of human gastric carcinoma cell line SCG-7901 by LY294002 with or with not LiCl treatment. The expression of total GSK-3? and p-GSK-3?ser9 were detected in protein level by western blotting, cell proliferation and cell cycle distribution were tested by MTT and FCM respectively, the expression of E-cadherin and ?-catenin were measured with S-P immunohistochemistry and the cell-ground substance adhesion was tested by cell adherence assay. Results:The expression of total GSK-3? was constant and the protein level of p-GSK-3? ser9 was down regulated after exposured to LY294002 by 72 h,the cell proliferation was inhibited, cell cycle was arrest at G0/G1 phase, the E-cadherin level was positively regulated, the expression of ?-catenin and the adhesion rate were decreased. The effect on the proliferation、cell cycle、the expression of E-cadherin and ?-catenin and the adhesion by the LY294002 were partially abrogated by lithium chloride.Conclusion:GSK-3?could negatively regulate the proliferation of human gastric carcinoma cell by leading cells to G1-S arrest and inhibit its metastasis by positively regulating the expression of E-cadherin.
9.Research of UVA in affecting Cx43 protein among rabbit lens epithelia cells
Journal of Chongqing Medical University 2007;0(07):-
Objective:To probe the mechanism of inhibition in gap junction-mediated intercellular communication (GJIC) of rabbit lens epithelia cells(RLECs) irradiated by UVA. Methods:RLECs of 2~3 generation were divided into normal control group and ultraviolet irradiation group (The UV intensity was 0.2 mW/cm2 under a UV lamp with fixed position 50 cm, RLECs were exposed to ultraviolet lamp at different times: 5,10,15 minutes). Western blot techniques were employed to detect the protein levels of Cx43 irradiated by UVA. By indirect immunofluorescence histochemistry we analyzed the localization of Cx43 in the RLECs. Results:In UVA irradiation group and normal control group, there was expression of Cx43. With the increasing intensity, the expression of Cx43 was decreasing. The immunnostaining of Cx43 in RLECs revealed that the internalization of Cx43 from membrance to plasma and nuclear was found in UVA irradiated RLECs. Conclusion: Cell molecule level study proved UVA irradiation can reduce GJIC of RLECs,which wis associated with perturbations in localization and number of Cx43 among RLECs.
