Journal of Chongqing Medical University 1986;0(04):-
Objective:To establish the method for the expression, purification and characterization of metallo-?-lactamases(IMP-1) and to explore the influence of experimental conditions on its separation and purification.Methods:pET9a/d-IMP-1 plasmid was transformed into E.coli competent BL21(DE3) cell and inoculated in LB medium. The supernatant were applied to SP Sepharose Fast Flow column, following by Sephadex G-75 column, and the purified enzymes were identified by SDS-PAGE and MALDI-TOF-MS and their activity was determined using Nitrocefin as substrate. Results:The purified enzyme showed high catalytic activity in the hydrolysis of most antibiotics. Its Michaelis constant K_m9.28?mol/L,and molecular weight Mr equalled 25111.9(determined by MALDI-TOF-MS).Conclusion:The method was established for the expression, purification and characterization of metallo-?-lactamases(IMP-1),which can be applicable to other metallo-?-lactamases.

China Journal of Traditional Chinese Medicine and Pharmacy 2005;0(08):-
Objective:To investigate the mechanism of therapeutic effects of Rhein on insulin resistance from aspects of the expression of resistin mRNA and plasma free fatty acid(FFA) in diabetic obese rats. Methods:The streptozotocin(STZ) and high fat diet-induced diabetic obese rats were divided into three groups at random. They were differently treated with Rhein, pioglitazone and saline by gavage. The expressions of resistin mRNA,plasma free fatty acid(FFA),as well as blood lipid in all groups were detected after administration by gavage for 8 weeks. Results:Similar to pioglitazone,Rhein can reduce the expression of resistin mRNA and the level of FFA,TC,TG and LDL(P
4.Effect of exectinib on the expression of EGFR genes 19 and 21 in patients with advanced lung cancer
Chinese Journal of Biochemical Pharmaceutics 2017;37(6):292-294
Objective To investigate the effect of ectectin on the expression of EGFR gene 19 and 21 in patients with advanced lung cancer.Aim to provide scientific reference.Methods68 patients with advanced lung cancer were treated with 3 months of ectectin.Serum free DNA was extracted from patients before and after treatment.RT-PCR was performed to amplify EGFR exon 19 and exon 21, EGFR gene mutations were detected by gene sequencing, and the mutation status of exon 19 and exon 21 of EGFR gene before and after treatment were evaluated.ResultsIn 68 patients with lung cancer before and after treatment with ectatinib, Histopathological types were correlated with the efficacy of ectectin hydrochloride.DCR of adenocarcinoma was significantly higher than that of squamous cell carcinoma.18 cases of EGFR mutations, there is no CR and PD cases, PR 11 cases, SD 7 cases, ORR 61.1%, DCR 100%.In the remaining 50 patients with unknown EGFR status, PR 24, SD 4, PD 15, ORR 48.0%, DCR 56.0%.There was no significant difference in the mutation rate of exon 19 and 21 between before and after chemotherapy. The mutation status of serum EGFR was correlated with the pathological type of patients (P<0.02).The EGFR gene mutation rate was 35.4% (17/48) before chemotherapy and 50% after chemotherapy. The rate of EGFR gene mutation was 55% before and after chemotherapy.ConclusionEctectin is effective in the treatment of advanced lung cancer, and it is effective in lung adenocarcinoma and may lead to the change of serum EGFR gene mutation in lung cancer patients, for the first-line application of patients treated with ectectin hydrochloride should always observe the changes in serum EGFR gene, to avoid blindness in treatment of patients.
5.Establishment of a rat model of hyperuricemia associated with uric acid excretion disorder
Chinese Journal of Comparative Medicine 2017;27(8):55-59
Objective To explore a reliable method to establish a rat model of hyperuricemia associated with abnormal uric acid excretion, and to lay the foundation for the study of pathogenesis of uric acid excretion disorder and the optimization of the treatment plan. Methods The models were established respectively by potasium oxonate(300 mg/kg) with pyrazinamide (300 mg/kg) or ethambutol(250 mg/kg).Continuous dosing for 1, 3 and 5 weeks, to determine the content of uric acid in rat blood, urine, and stool, the function of liver and kidney was detected and pathological examination was performed. Results The blood uric acid in the potasium oxonate and ethambutol group was increased first and then decreased, while in the potasium oxonate and pyrazinamide group were increased steadily and the excretion of uric acid in urine was stable during the continuous administration. The two methods showed no harmful effect on the liver and kidney function. Conclusions A stable rat model of hyperuricemia associated with uric acid excretion disorder can be effectively established by potassium oxonate and pyrazinamide, exhibiting similar manifestations of clinical hyperuricemia and uric acid excretion disorder.

Journal of International Oncology 2017;44(7):496-499
Objective To determine the levels of serum anterior gradient 2 (AGR2) before and after treatment in nasopharyngeal carcinoma (NPC) patients, and investigate the relationship of AGR2 and clinical pathological characteristics of NPC patients. Methods The serum levels of AGR2 were detected by enzyme linked immunosorbent assay (ELISA) in 55 NPC patients (NPC group) before and after treatment, 30 patients with nasopharyngeal inflammation (inflammation group) and 20 healthy controls (health control group). The correlations between serum AGR2 before and after treatment and clinical pathological characteristics of NPC were analyzed. The NPC patients received radiotherapy and were followed up for 6 months, and the therapeutic effect was evaluated. Results The serum AGR2 levels in NPC group before treatment, inflammation group and health control group were (22.92±5.24)μg/L, (9.50±4.15)μg/L and (8.75±2.18)μg/L respectively, and the difference was statistically significant (F=268.400, P=0.000).The level of serum AGR2 in NPC group was obviously higher than that in inflammation group (t=14.241, P=0.000) and health control group (t=15.254, P=0.000).The level of serum AGR2 in NPC group after treatment was significantly lower than that before treatment [(15.15±10.33)μg/L vs.(22.92±5.24)μg/L, t=12.774, P=0.000].In NPC patients, serum AGR2 levels of clinical Ⅲ-Ⅳ stage group before and after treatment were higher than those of clinical Ⅰ-Ⅱ stage group (t=5.938, P=0.000;t=0.759, P=0.032).Serum AGR2 levels of lymph node metastasis group before and after treatment were higher than those of no lymph node metastasis group (t=6.879, P=0.000;t=2.700, P=0.009).Serum AGR2 levels of carotid sheath and skull base involvement groups before treatment were higher than those of non-involvement groups (t=8.342, P=0.000;t=8.255, P=0.009).Serum AGR2 levels of cranial nerve involvement group before and after treatment were higher than those of non-involvement group (t=7.743, P=0.000;t=3.021, P=0.004).The serum AGR2 level after treatment in complete response patients [(13.86±2.93)μg/L] was significantly lower than that in partial response patients [(15.85±3.24)μg/L, t=2.267, P=0.028] and invalid patients [(20.65±6.59)μg/L, t=4.935, P=0.000].The serum AGR2 level in partial response patients was significantly lower than that in invalid patients (t=3.196, P=0.004).Conclusion The level of serum AGR2 in NPC patients increases obviously.AGR2 plays an important role in the genesis and development of NPC, and can be used as a new marker of NPC for judging the clinical therapeutic efficacy and prognosis.
