1.Nitric oxide, nitric oxide synthase and cerebral vasospasm after subarachnoid hemorrhage

Over the years,cerebral vasospasm has been regarded as one of the important factors influencing the prognosis in patients with subarachnoid hemorrhage.Studies have shown that cerebral vasospasm is associated with nitric oxide and nitric oxide synthase after subarachnoid hemorrhage,and regulating the synthesis of nitric oxide may play an active role in the treatment of cerebral vasospasm.
2.A clinical investigation on factors influencing postoperative serum bilirubin level in patients with primary liver cancer

Objective To discuss clinical factors associated with high serum bilirubin level postoperatively in patients with liver cancer. Methods In this study,130 postoperative cases of primary liver cancer were divided into two groups of high serum bilirubin and of normal bilirubin according to serum bilirubin level during two weeks of postoperation. Results Preoperative serum bilirubin、 Child grade、 operation time、 blood loss and types of hepatic vascular exclusion were correlated with the level of postoperative serum bilirubin. Types of hepatic vascular exclusion was a independent factor predicting postoperative elevation of serum bilirubin level. Conclusions Preoperative liver function, operation time, types of hepatic vascular exclusion and blood loss are all among factors influencing postoperative serum bilirubin level.

Recently,the techniques for proteomics have made remarkable progress.They are widely used in various fields of life sciences,providing strong technical supports for relevant researches.Especially,proteomics technology has made prominent contributions to drug development and mechanism studies,magnificently improving the efficiency of discovering new drugs.This paper summarizes the classical method and new technology of proteomics.Also,its applications in drug study,including targets screening,mechanism studies,drug toxicology,researches of resistance mechanisms and clinical medical studies have been reviewed.
4.Expression of myostatin gene mRNA in the muscle tissue from 75 patients with muscular weakness

Objective To investigate myostatin gene mRNA expression in the muscle tissue from patients with muscle weakness suffering from different illness.Methods The clinical data of our patients were all from the Muscular Disease Center,Department of Neurology,People' s Liberation Army General Hospital.A total of 75 patients suffered from muscular weakness were included consecutively.Skeletal muscle biopsies were obtained with informed consent from all 75 patients.The diagnosis was confirmed by two senior doctors for muscular disease according to the clinical feature,the results of electromyography,serum creatine kinase activity and histopathological examination.Among of them,21 cases were diagnosed as polymyositis,15 cases progressive muscular dystrophy,5 cases neurogenic amyotrophy,4 cases chronic muscle fiber damage,4 cases mitochondrial myopathy,4 cases lipid storage myopathy,4 cases myotonic dystrophy,3 cases muscular dystrophy in adults,2 cases dermatomyositis,and 2 cases inclusion body myositis.There were 2 cases characterized by pure high activity of creatine kinase.And the other 9 cases were diagnosed as non-neuromuscular disease.The expression of myostatin gene mRNA in muscle tissue was evaluated by reverse transcription polymerase chain reaction method,with glyceraldehyde-3-phosphate dehydrogenase as internal reference.Results The expression of myostatin gene mRNA was detected in 63 patients,but not in other 12 cases,and the percentage of positive expression was 84%.The expression index was with great variation,from 0 to 3.52.In positive cohort,the index was correlated positively with the duration of disease (r =0.236,P =0.041).The activities of creatine kinase in positive expression cohort were higher than that of negative one,but nonsignificantly.Conclusion The expression of myostatin gene mRNA in muscle tissue may not correlate to the entity of atrophic muscular disease because of its great variation.
5.Development of fluorescence imaging based assay for screening compounds with anti-migration activity.

In the present study, A fluorescent imaging-based high-throughput screening method was developed for identifying anti-migratory compounds with 96-well Transwell plates. The correlation, precision and stability of this method were examined and the incubation time of dye Hoechst 33342 in addition to migration time was optimized. In addition, The inhibitory activity of anti-cancer drug paclitaxel on tumor cell migration was assayed and an IC50 value of 0.717 micromol x L(-1) was obtained. Using this method, 24 components from Rhizoma Alismatis were screened and one component with anti-migration activity was found. These results show that the new proposed method with good precision, stability and linear range has the potential to assay the inhibitory activity of anticancer compounds.
6.Strategy of antibody-drug conjugates in preclinical safety evaluation

Recently,increasing cancer researches focus on antibody-drug conjugates(ADCs) which can improve the anti-tumor potency with less adverse effect while benefiting patients in the future. However,safety evaluation of ADCs is a big challenge because of complex components as well as in experience in preclinical studies. In this review,the authors reviewed the mode of action,hazard risks,and toxicity observed in preclinical/clinical studies of ADCs,summarized the preclinical studies of Adcetris(brentuximab vedotin)and Kadcyla(ado-trastuzumab emtansine),and suggested a better strategy of ADCs in preclinical safety evaluation.
7.Hemodynamic changese during the procedure or double cardiac valve replacement.

The hemodynamics of 49 scheduled patients during anesthesia and opration for mitral and aortic valve replacement were studied with the insertion of SwanGanz catheter and by the method of thermodilution technics.Pre-anesthesia CI and LVWI were in the nor mal range with abnormal MPAP, PCWP, PTRI and RVWI.Increase of RAP and decrease in MPA,PCWP, MAP, CI, LVWI and RVWI were observed during induction of anesthesia. The hemodynamic parameters returned to or exceded the pre-anesthesia values after trachcal intubation and sternotomy. The CI, MAP, MPAP, PCWP, LVWI and RVWI decreased signifi- cantly after insertion of aorta and vena cava catheter. The reduction of MPAP, P(IWP,PTRI and RVWI post-cardiopulmonary bypass (CPB) were significant and persistent with no change of PVRI.The lower MAP,SVRI and deterioration of cardiac performance post-CPB indicate that the administeration of positive inotropir agents and restoration of hematocrit are nec essary as early as possible.

Aim To establish an ideal two-dimensional polyacrylamide gel electrophoresis( 2-DE) method for serum proteomic research. Methods The 2-DE experiment of serum was opti-mized by adjusting the conditions of sample pretreatment and silver nitrate staining. Results An ideal 2-DE experiment method with higher repetition was established. The number of protein spots and the resolution were both increased,and this method was compatible with MS analysis. Conclusion After the adjustments and optimizations,an ideal method of 2-DE technique for serum is established,which has a high reference value for the relevant researches,and lays an experimental foundation for the search,analysis as well as identification of the disease related proteins in serum.
