1.Application experience of DR in fatigue fracture

Objective To discuss the advantages of DR in fatigue fracture diagnosis compared with the plain X-ray photography.Method 30 cases performed DR and 30 cases performed plain X-ray check were collected respectively as two groups,all of which have been diagnosed as fatigue fracture clinically.Research and comparison were made to the two groups.Results In DR group,the percentage of showing fracture line,across medullary cavity of bones compact zone and periosteal proliferation were 86.7%,80% and100% respectively,while the plain X-ray group were 43%,33% and 66.7%.Conclusion Compared with plain X-ray,DR shows better imaging with more diagnostic information,and meanwhile it can greatly reduce the X-ray radiation and computerize the imaging.
2.Effect of recombined RNAi plasmids on apoptosis of cisplatin-resistant cell line Acc-3/CDDP

Objective:To investigate the effect of the silence of Bcl-2 oncogene on the apoptosis of cisplatin-resistant cell line Acc-3/CDDP.Methods:The recombined RNAi plasmids for Bcl-2 oncogene were constituted by the four successive steps-designing of Oligo DNAs,and were transfected into Acc-3/CDDP cells by culturing together.The apoptosis of each cells were evaluated by MTT test,electron microscope,and AO/EB fluorescent dyes respectively.Results:The OD value of RNAi group decreased with time last by MTT test,and dropped to the lowest at 48 h.The apoptosis of RNAi group were observable through electron microscope and fluorescent dyes.The apoptosis rate increased with time from 24 to 72 h after transfection.The apoptosis on each cell cycle were clear and classical.Conclusion:Recombined interfering plasmids can effectively silence the expression of Bcl-2 oncogenes,and the apoptosis of cisplatin-resistant cell line Acc-3/CDDP cells can be enhanced.
3.Establishment and drug-resistance gene expression of cisplatin-resistant cell line Acc-3/CDDP

Objective:To establish a CDDP-resistant cell line from human oral adenoid cystic carcinoma cell line Acc-3 and compare the drug-resistance gene expression between them.Method:The CDDP-resistant cell line Acc-3/CDDP was established by increasing the concentration of CDDP added to Acc-3 cells step wisely.The index of drug-resistance was estimated by MTT assay.The expression of MDR-1,MRP-1,GST-?and Bcl-2 were measured byRT-PCR optical densityOD.Result:The resistance factor(sRF=IC5(0Acc-3/CDDP) /IC5(0Acc-3))for CDDP,5-Fu,VCR,MTX and PYMwere 8.9,1.97,2.83,2.42,1.1 respectively.The expression of drug-resistance gene MRP-1,GST-?and Bcl-2 increased in Acc-3/CDDP cells.MDR-1 was expressed in Acc-3/CDDP,but none in Acc-3.Conclusion:MDR-1,MRP-1,GST-?and Bcl-2 were significantly over expressed in CDDP-resistant cell line Acc-3/CDDP,which can provide effective models for the study on multi-drug resistance of oral carcinoma.
4.Simultaneous Determination of Lithospermic Acid and Rosmarinic Acid in Guanxin Danshen Capsule by UPLC

OBJECTIVE:To establish a method for the simultaneous determination of lithospermic acid and rosmarinic acid in Guanxin danshen capsule. METHODS:UPLC was performed on the column of ACQUITY UPLC-BEH C18 with mobile phase of 0.1% formic acid-methanol solution (gradient elution) at a flow rate of 0.3 ml/min,detection wavelength was 280 nm,column temperature was 30 ℃,and the injection volume was 5 μl. RESULTS:The linear range was 0.019-0.76 mg/ml for lithospermic ac-id(r=0.999 9)and 0.005-0.2 mg/ml for rosmarinic acid(r=0.999 7);RSDs of precision,stability and reproducibility tests were lower than 2%;recoveries were 97.34%-101.24%(RSD=1.57%,n=6) and 98.30%-100.39%(RSD=0.86%,n=6). CONCLU-SIONS:The method is simple,accurate and reproducible,and can be used for the contents determination of lithospermic acid and rosmarinic acid in Guanxin danshen capsule.
5.Application Value of Aer-barium Sulphate Double Contrast Radiograph in Diagnosis of Digestive Tract Diseases

Objective To discuss the application value of aer-barium sulphate double contrast radiograph in digestive tract diseases diagnosis. Methods The author analyzed 197 cases' images retrospectively which had positive performance in radiograph and had been clinical defined in our hospital from January to June 2008. Results In all these cases, there are 83 cases origin from mucous membrane which include inflammation, tumor and anabrosis; 24 cases origin below mucous membrane which include interstitialoma, lymphoma and scleroderma; 38 cases mainly changed in the shape, structure and position of organs which include hiatus hernia, Oblique inguinal hernia, diverticulum, malrotation of intestine and megacolon; 29 cases mainly changed in the power and function of organs which include functional disorder of esophagus, mesentery-compressed syndrome funtional disorder of small intestine and irritable colon; 17 cases' performance caused by the factor outside intestinal canal which include mesentery shift, stomach shift and small intestine shift, and other cases which include fistula between esophagus and mediastinum, rectovaginal fistula, foreign body in esophagus and small intestine ascariasis. Conclusion Aer-barim sulphate double contrast radiograph is the best method to diagnose the digestive tract diseases because it has forte as follow: convenient, easy prepared, less painful, large-scale examine limit and reliable diagnosis.
6.Cerebral oxygen saturation monitoring for improving postoperative cognitive dysfunction

Postoperative cognitive dysfunction (POCD) is a common complication of the central nervous system after major surgery with anesthesia,which severely impairs patients' quality of life.Near infrared spectroscopy (NIRS) is advantageous for its convenience,inexpensiveness,noninvasiveness,continuous operation and bedside monitoring.Routine monitoring of regional cerebral oxygen saturation has the potential of optimizing the anesthesia plan and reducing the incidence of POCD.The aim of this review is to describe the NIRS technology and its clinical application.
7.Interstitial Implantation of ~(125)I seeds for Recurrent or Metastatic Malignant Tumor at the Head and Neck

Objective To evaluate the feasibility of interstitial implantation of ~(125)I seeds for recurrent or metastatic malignant tumor at the Head and Neck.Methods From January 2003 to July 2007,29 patients received interstitial implantation of ~(125)I seeds in our hospital because of recurrent or metastatic malignant tumors at the head or neck after surgery or chemotherapy.Under local anesthesia and guidance of CT,B-ultrasonography,or endoscopy,~(125)I seeds were implanted into the tumor tissues with intervals of 0.5-1.0 cm.The matched peripheral dose(MPD)was 90-160 Gy,and the radioactive activity was set at 29.6 MBq per seed.Results The operation was completed in all the patients.A median of 23 seeds(12-67)were implanted in each case.No complications,such as hemorrhage,infection,or seed migration,occurred in the patients.The patients were followed up for 3-24 months(mean,16 months).During the period,the rate of local control was 55%(16/29)at 3 months,71%(20/28)at 6 months,45%(10/22)at 12 months,and 36%(5/14)at 2 years.Conclusion Interstitial implantation of ~(125)I seeds is feasible for recurrent or metastatic tumor at the head or neck with a good short-term outcome.
8.Influence of milrinone on systemic inflammation in perioperative patients undergoing cardiac valve replacement

Objective To investigate the influence of milrinone on systemic inflammation in the perioperative cardiac valve replacement Methods Twenty patients, scheduled for cardiac valve replacement were randomly divided into 2 groups: milrinone and control groups Before anesthesia induction, a bolus of milrinone 30?g/kg was given in 10min, followed by a continuous infusion of 0 5?g?kg 1 ?min 1 during operation in milrinone group and the same volume of saline was administrated in control group The mixed venous blood samples were obtained from the pulmonary artery at the following points : before induction of anesthesia (T 1), 30min after anesthesia (T 2), 30min after the cross clamping of aorta (T 3 ), 10min and 30min after the declamping (T 4,T 5), 30min and 8 h after CPB(T 6,T 7), 24h and 72 h after operation (T 8,T 9) IL 6, IL 8 and IL 10 were determined by enzyme linked immunosorbent assay Results Compared with those at T 1, the levels of WBC,PMN,IL 6 and IL 8 increased significantly from T 5 to T 9, and IL 10 level rose markedly from T 3 to T 9 (P0 05) Conclusions During CPB and cardiac valve replacement the systemic inflammation is obvious , reaches the peak 30 min after CPB and persists to 72h after operation Perioperative administration of low dose milrinone is effective against the inflammation
9.Inflammatory mechanism of estrogen in the pain modulation of ovariectomized mice

Objective·Giving the ovariectomized (OVX) mice exogenous estrogen replace therapy (ERT) so as to explore the inflammatory mechanism of estrogen in the pain modulation of OVX mice.Methods·Twenty-four 12-week-old female C57BL/6J mice were randomized into three groups:sham group (S),ovariectomy+placebo group (P),ovariectomy+estrogen group (E).The ERT began from one week after OVX and last for four weeks.Paw withdraw threshold (PWT) and paw withdraw latency (PWL) were measured at one day before OVX,and three days,one week,two weeks,three weeks,four weeks and five weeks after OVX.Concentrations of inflammatory cytokines in serum were measured at three days and five weeks postoperatively.Five weeks after operation,the lumbar intumescentia of spinal cord was taken and the expression of p38 was analyzed by Western blotting.Results·All the OVX mice's PWTs or PWLs were decreased after operation compared with non-operation mice;PWTs and PWLs were significantly different from group S separately from one week and three days after OVX (P<0.01).The inflammatory cytokines of OVX mice were significantly higher than group S (both P<0.01) at three days after OVX.Western blotting results showed that the p38 of group P was significantly higher than that of group S after five weeks postoperatively (P<0.01).By receiving ERT,both PWTs and PWLs of group E increased slightly and were significantly different from group S (P<0.01) at four and five weeks after OVX.At five weeks after OVX,inflammatory cytokines of group E were significantly lower than group P (P<0.01) but still much higher than group S (P<0.01).Meanwhile the content of p38 in group E was less than that in group P (P<0.01).The p38 in group E was still more than group S,however it was no longer significantly different from group S (P>0.05).Conclusion·Estrogen may reduce concentrations of inflammatory cytokines through the p38 MAPK pathway and plays an important role in the pain modulation of OVX mice.
