OBJECTIVE To study the effect of total flavonoids of Dracocephalum heterophyllum (TFDH),a Uygur medicine,on cardiomyocyte hypertrophy induced by norepinephrine(NE),and to provide insights into the mechanism.METHODS Cardiomyocytes of primarily cultured neonatal rats were used as models. Myocardial hypertrophy was induced by NE 2 μmol·L-1. The cells were divided into cell control group,NE 2 μmol·L-1model group, and TFDH 10,25 and 50 μmol·L-1+NE 2 umol·L-1groups.CCK-8 method was used to observe the activity of myocardial cells, while RT-PCR technique was used to detect the expression of mRNA of cardiac hypertrophy gene atrial natriuretic peptide (ANP) and β-myosin heavy chain (β-MHC). The internal factors were glyceraldehyde-3-phosphate dehydrogenase (GAPDH). Confocal laser scanning was used to detect the surface area of myocardial cells and[Ca2+]i.The activity of Ca2+-ATP was measured with enzymatic reaction of fragmentation cells. The concentration of NO and the activity of NOS were determined with colorimetry. RESULTS Compared with the cell control group, cardiomyocytes were stimulated at 48 h by NE 2 μmol·L-1, which could decrease the survival rate of cardiomyocytes from(95±1)% to(78+5)%,surface area increased from (178±29)μm2to(274± 38)μm2(P<0.05),the expression of mRNA of ANP and β-MHC increased from(1.00±0.01)and(1.00± 0.02)to(2.76±0.55)and(2.69±0.31),respectively(P<0.05).The concentration of[Ca2+]iincreased from 1.00±0.12 to(1.52 ± 0.41)μmol·L-1,while the activity of Ca2+-ATP decreased from 1.01±0.14 to(0.41± 0.06)μmol·L-1(P<0.05).The concentration of NO decreased from 1.50±0.14 to(1.12±0.05)μmol·L-1,and the activity of NOS decreased from 0.86±0.06 to(0.52±0.10)μmol·L-1(P<0.05).TFDH 10,25 and 50 μmol·L-1could inhibit the decline of the survival rate, increase of the surface area and the increased expressions of mRNA of ANP and β-MHC that were induced by NE(P<0.05).At the same time,it also could inhibit the increase of the concentration of[Ca2+]i,the decreased of activity of Ca2+-ATP,and the decline of the concentration of NO and the activity of NOS (P<0.05). CONCLUSION TFDH can improve the activity of cardiomyocyte hypertrophy induced by NE, down-regutate mRNA expressions of proto oncogene ANP and β-MHC,and reduce the surface area of cardiomyocytes induced by NE.The mecha-nism may be related to promoting the release of NO and regulating the concentration of [Ca2+]iand the activity of Ca2+-ATP.

Objective To quantitatively analyze the epidemic situation of tuberculosis(TB)by modeling the data of tuberculosis in prefectures of Xinjiang, and predict the new cases of tuberculosis in prefectures of Xinjiang. Methods A dynamic model was used to fit the data of TB in 14 prefectures in Xinjiang from 2005 to 2014,the results of the fitting were verified by tuberculosis data in 2015-2017,verified results were evaluated, estimated values and basic reproductive numbers (R0)of parameters in each region were obtained, data of new TB in 2018-2022 were predic-ted.Results The verification of TB data in 2015-2017 showed that the actual values fell within the 95% confidence interval of the predictive value curve, model was fit well.R0in Southern Kashgar was 1 1.38 (95%CI:1 1.33-11.50),R0in Urumqi City in Eastern Xinjiang and Ili Kazak Autonomous Prefecture in Northern Xinjiang were 5.46 (95% CI :5.28-5.50)and 2.22 (95% CI :2.18-2.28)respectively.The epidemic situation of TB in Southern Xinjiang was more serious than that in Northern and Eastern Xinjiang, epidemic situation of TB in Kash- gar Prefecture was most serious. The predicted results showed that the number of new TB from 2018 to 2022 will slowly grow in most prefectures. Conclusion The dynamical model of TB fits well and is feasible in this study,it can be used for prediction of new TB cases, intervention and management in Southern Xinjiang should be strength-ened to control the prevalence of TB.

Objective To propose a novel algorithm model based on YOLOv7 for detecting small lesions in ultrasound images of hepatic cystic echinococcosis. Methods The original feature extraction backbone was replaced with a lightweight feature extraction backbone network GhostNet for reducing the quantity of model parameters. To address the problem of low detection accuracy when the evaluation index CIoU of YOLOv7 was used as a loss function,ECIoU was substituting for CIoU,which further improved the model detection accuracy. Results The model was trained on a self-built dataset of small lesion ultrasound images of hepatic cystic echinococcosis. The results showed that the improved model had a size of 59.4 G and a detection accuracy of 88.1%for mAP@0.5,outperforming the original model and surpassing other mainstream detection methods. Conclusion The proposed model can detect and classify the location and category of lesions in ultrasound images of hepatic cystic echinococcosis more efficiently.

Objective To explore the in vivo exposure levels of cigarette smoking ( CS) by measuring the biomar-kers nicotine and cotinine. Methods One hundred and sixty male SD rats were divided into 15 cigarette exposure groups (10, 20 and 30 nonfilter cigarettes/day, for 2, 4, 6, 8 and 12 weeks) and a control group (without CS exposure). The rats were sacrificed at different time?points. The concentration of plasma nicotine and cotinine were measured by GC?MS/MS. Results The CS?exposed rats displayed decreased locomotor activity, ataxic gait, irregular respiration, nasal noise, and salivation after smoking exposure for 3 weeks. Rats in the CS exposure groups had lower body weight, and the reduction of body weight was time and dose related (P<0. 01). The retention time of nicotine was 7. 5 to 8. 5 min. The concentra?tion of plasma nicotine in the CS exposure groups was higher than control group (155 ± 56. 65) ng/mL. The retention time of cotinine was 11. 5 to 12. 5 min, the concentrations of plasma cotinine in CS exposure groups were higher than control group (340 ± 41. 97) ng/mL, the increase of plasma cotinine in CS groups was time?related (P<0. 05), and the exposure concentration and duration had synergistic effect on the level of plasma cotinine ( P<0. 05 ) . Conclusions CS exposurecauses structural damages in male rats. The plasma concentration of cotinine can effectively reflect the in vivo exposure lev?els of cigarette smoking, and well presents a dose?response relationship.
