1.Analysis on correlation between PDK 1 and Ki-67 expression levels with survival and prognosis of patients with ovarian serous carcinoma

Objective To analyze the relationship between the expression levels of pyruvate dehydro-genase kinase 1(PDK 1)and Ki-67 with the survival and prognosis of the patients with ovarian serous carci-noma(OSC).Methods The cancer tissues of 93 patients with OSC treated by surgery in this hospital from January 2000 to June 2020 were collected,and the expression levels of PDK1 and Ki-67 in the tissues were de-tected by immunohistochemistry.The relationship between the expression levels of PDK1 and Ki-67 with the clinicopathological characteristics of the patients with OSC was analyzed by the single factor and binary logis-tic regression model.The relationship between the PDK1 and Ki-67 expression levels with the recurrence and platinum resistance within 3 years was analyzed.The Kaplan-Meier survival curve was adopted to analyze the effect of PDKI and Ki-67 expression levels on the survival and prognosis in the patients with OSC.Results The expression levels of PDK1 and Ki-67 were related with the International Federation of Gynecology and Obstetrics(FIGO)stage,tissue differentiation level,lymphatic metastasis,ascites formation and positive asci-tes(P<0.05).The high expression of PDK1 was a risk factor for the late FIGO stage(Ⅲ-Ⅳ),lymphatic metastasis and ascites positive in the patients with OSC(P<0.05);the high expression of Ki-67 was a risk factor for the late FIGO stage(Ⅲ-Ⅳ)and ascites formationin in the patients with OSC(P<0.05).The re-currence rate and platinum resistance rate within 3 years in the patients with PDKi and Ki-67 high expressions were significantly higher than those in the patients with low expressions(P<0.05).The progression-free survival(PFS),overall survival(OS)and 5-year survival rate in the patients with high PDK1 and Ki-67 ex-pressions were significantly lower than those in the patients with low expressions(P<0.05).Conclusion The expression levels of PDK 1 and Ki-67 play an important role in judging the survival and prognosis of the patients with OSC.
2.Expression and clinical significance of DNMT1 and CCNA1 in different grades of cervical lesions

Objective To investigate the expression and clinical significance of DNMT1 and CCNA1 in different grades of cervical lesions. Methods Cervical tissues were selected from normal cervical(NC),low-grade cervical intraepithelial lesion(LSIL),high-grade cervical intraepithelial lesion(HSIL)and cervical squa-mous cell carcinoma(SCC)from each thirty patients.The expression of DNMT1 and CCNA1 mRNA and protein was examined by Western Blot analysis and qRT-PCR in cervical tissues. Results The expression of DNMT1 mRNA and protein in LSIL,HSIL and SCC was higher than in NC(F=117.93,P<0.05;F=61.24,P<0.05). Expression of DNMT1 mRNA and protein was increased steadily according to severity of cervical lesions(χ2trend=26.25,P<0.05;χ2trend=26.60,P<0.05).The expression of CCNA1 mRNA and protein in HSIL and SCC was lower than that in NC and LSIL(F = 77.04,P < 0.05;F = 57.15,P < 0.05). Expression of CCNA1 mRNA and protein was decreased steadily according to severity of cervical lesions(χ2trend=64.19,P<0.05;χ2trend=60.24,P<0.05). There was a negative correlation between expression of DNMT1 and CCNA1 protein in LSIL,HSIL,SCC (r=-0.75,-0.56,P<0.05). Conclusion In DNMT1 mRNA there is high expression of protein and in CCNA1 mRNA there is low expression of protein,but both may be related to the occurrence and development of cervical cancer.
3.Clinical value of long chain noncoding RNA-LINP1 in evaluating the prognosis of patients with endometrial cancer

Objective:To detect the level of long non-coding RNA LINP1 (lncRNA-LINP1) in endometrial carcinoma and to explore the prognostic value of the expression leve.Methods:From January 2015 to December 2016, the tissue samples of 82 patients with endometrial carcinoma in North China University of Science and Technology Affiliated Hospital were used as endometrial carcinoma group, and the normal adjacent tissues were selected as the paracancerous control group.The expression of LINP1 mRNA was measured by real-time fluorescence quantitative PCR (qRT-PCR). The clinicopathological data of the patients were collected, and the relationship between the expression of LINP1 mRNA and clinicopathological parameters was analyzed.Kaplan-Meier method was used to analyze the relationship between the expression of LINP1 mRNA and the survival rate of endometrial patients; and COX multivariate analysis was used to identify the risk factors of death in patients with endometrial cancer.Results:The expression of LINP1 mRNA in endometrial carcinoma group and paracancerous control group were (2.38±0.43) and (1.00±0.24). There was significant difference between the two groups ( t=25.376, P<0.001). The results of clinicopathological parameters showed that the expression of LINP1 mRNA was not related to the age and histological type of endometrial carcinoma (all P>0.05). It was related to lymph node metastasis, FIGO stage, tumor differentiation and vascular invasion (all P<0.05). the three-year total survival rate of the group with high expression of LINP1 mRNA (48.78%) was lower than that of the group with low expression of LINP1 mRNA (80.49%), and the differences were statistically significant (χ 2=6.306, P<0.05). COX Regression analysis showed that, high expression of LINP1 mRNA and FIGO Stage III, IV and lymph node metastasis were the risk factors for death in endometrial cancer patients (HR(95% CI) were 2.898(2.031-4.136), 1.831(1.448-2.316), 1.708(1.364-2.138), P values were 0.002, 0.004 and 0.008, respectively. Conclusion:The high expression of LINP1 mRNA in endometrial carcinoma is closely related to lymph node metastasis, FIGO stage, tumor differentiation, vascular invasion and prognosis.It is a risk factor for the death of patients with endometrial cancer and may be used as an index to evaluate the prognosis of patients with endometrial cancer.
