1.Study the high effect process of collecting hematopoietic stem cell from umbilical cord blood

Background: Umbilical cord blood - Stem cell CD34 are attracted to study and apply to cure many serious diseases. But the quantity of the cell is small, so it only used to children. Therefore, studying to find out the high effect collected process is necessary. Objective: studying to find out the high effect process of collecting hematopoietic stem cell from umbilical cord blood. Subjects and methods: the study included 112 umbilical cord blood specimens collected from healthy mother delivering at National Hospital of Obstetric and Gynecology.Results: The condition of the mother was healthy, with no risk factors of infection or genetic disease. The average age of mothers was 28 \xb1 5 years old, average weight: 63 \xb1 5 kg. Age of fetus was from 39-41 weeks, average weight: \ufffd?3000g. Collection technique: cutting the umbilical cord after delivery 6-10 seconds, cutting the umbilical cord far from neonatal 6 cm, blood collection continued after the collection of placenta, all processes must be implemented rapidly in the 4 -5 minutes in aseptic condition. With the above process it can be obtained with high effect blood: volume was 90 - 120ml (48%). CD34 cell count: 1.6 \xb1 0.3x106, the number of eukaryotic cells: 13.53 \xb1 1.7x108/unit of the umbilical cord. 6.Investigation on volume of cord blood

At the first step of studying about “the volume of cord blood” in Vietnamse, a total of 50 samples have been collected. We obtained some results: - The mean volume of cord blood was: 50.06 +/- 20,73ml. -The mean number of total mononuclear cells was 352.4 +/- 161.6 (x106). -Some cord blood colections had many total MNC as some larger cord blood harvests. So, it is our belief that total MNC numbers are more useful indicator of clinical utility than more sample volume.

7.Screening of CMV, HBsAg, HCV, HIV and syphilis in cord blood

51 cord blood samples were screened for CMV, HbsAg, HCV, HIV by Elisa technique and syphilis by VDRL. The results showed that 100% were positive for IgG anti-CMV, 100% negative for IgM anti-CMV, and 1.96% positive for HBsAg and HV, while there were no positive cases for HIV and syphilis. These rates were higher in comparison with those found by foreign studies but comprehensible concerning epidemiology and rate of these mentioned viruses in the Vietnamses population. For safe reasons, all cord blood found positive with any of the above tests should be rejected and corresponding mothers should be also included in the programmed tests.

8.A study on cord blood collections

A total of 54 cord blood collections were analysed for lymphocyte subsets. No significant differences in percentages of major lymphocyte subsets were found on CB in comparison with peripheral blood (CD 2+ 65%+/- 60, CD 3+ 69% +/- 5,4, CD 4+ 57%+/-7.6, CD 7+ 78% +/- 7.4 CD 8+ 16% +/- 6.1CD 16/56+ and CD 3+ 0.9% +/- 0.5, CD 19+ 16% +/- 3.9). The CD4: CD8 ratio is 3.56 compared with 1.4 in adult PB.CD 3+ with cytotoxic activity as determined by the expression of CD 56 are practically absent on CB.

9.A study on cord blood collections

A total of 47 cord blood collection were analysed for CD 34+ progenitor cells. Results: volume (ml) without anticoagulant 64ml (40-90). Total NC 9.82x108+/- 0.45. Total CD 34+ 1.79x10 +/-1.4. Mean percentage of CD34+: 0.32%+/ 0.24. Given that a transplanted cell dose of 20x106 NC/kg body weight would be sufficient for hematopoiesis, 95% of all stored CB samples would be sufficient for patients up to 10 kg body weight and 36% for patients up to 40 kg body weight.

10.Infection situation in cord blood collection

From the beginning of cord blood collection, we considered bacterical contamination. Fifty samples have been collected from the obstetrical Hung Vuong hospital for studying of blood culture. Using a bag collection set (Baxter Health Care). The contamination of sample is due to skin flora: staphylococcus aureus.
