A high throughput measurement method of human red blood cells (RBCs) deformability combined with optical tweezers technology and the microfluidic chip was proposed to accurately characterize the deformability of RBCs statistically. 2.Effects of different temperature and time on the period of validity and quality in blood preservation.

AIMTo examine the effects of different temperature protection on measures on preservation damages in liquid blood and explore the corresponding.
METHODSTake equal half blood samples from 10 healthy blood donors and divided each sample into two groups, put the fresh blood into CP2D-A solution at 0 degrees C and 4 degrees C, respectively and take the samples 21 days and 42 days, later and then measured the contents of membrane phospholipids with shafig-UR-rehman method, CaM with purification PED test, LPO with spectrophotometry.
RESULTSAt the same temperature, when the preservation time was prolonged, peroxidation was increased, the preservation damages were also augmented; the damages were declined when the temperature was lower during the same period, the aging of blood was more evident at 4 degrees C.
CONCLUSIONBlood peroxidation temperature is lower. The author pointed out the questions and prospects of blood preservation.

3.Two mathematical models of estimating RBC membrane elastic modulus and a comparison of them by use of bottom attached method.

4.Changes of blood viscosity and erythrocyte rheology in acute hypoxic hypoxia mices.

OBJECTIVETo observe the changes of blood viscosity and erythrocyte rheology in mice after acute hypoxic hypoxia (AHH).
METHODSThirty-two Kui-ming mice were randomly divided into control group, AHH group (duplicating AHH model, and divided into 5 min, 8 min, 11 min subgroups), the blood sample was taken out from heart after neck dislocation at corresponding times, for detecting the blood viscosity and erythrocyte rheology indices.
RESULTSCompared with control group, the whole blood viscosity at different shears, whole blood reduced viscosity, whole blood relative viscosity were lower and the erythrocytes aggregation index was higher in AHH 5 min group; the electrophoresis time was longer and the electrophoresis length, migration of erythrocyte were lower in AHH 8 min and AHH 11 min groups. The whole blood reduced viscosity, whole blood relative viscosity, erythrocytes aggregation index in AHH 8 min group were higher, and the erythrocyte deformability index was lower significantly than that of AHH 5 min group, respectively.
CONCLUSIONThese data suggested that the AHH could induce the blood viscosity and electrophoresis ability.

5.Alterations of erythrocyte deformability and membrane protein after high intensity training and recovery in rats.

AIM AND METHODSThe purpose of this investigation was to determine the changes of erythrocyte deformability, band-3 protein and actin in the definite volume of erythrocyte membrane after high intensity running training and recovery in rats.
RESULTSLong-term training could significantly improve erythrocyte deformability and the quantity of membrane proteins. Erythrocyte deformability, band-3 protein and actin decreased transitorily at varying degrees after inadaptable high intensity exercise. One and two week training could improve erythrocyte deformability, the quantity of band-3 protein and actin after recovery.
CONCLUSIONAlterations of erythrocyte membrane protein after high intensity training could cause the change in erythrocyte membrane structure and hence influenced erythrocyte deformability. That was maybe one of mechanisms of training effecting erythrocyte deformability.

6.The effects of mesenteric lymph drainage on erythrocyte rheology in rats with hemorrhagic shock.

OBJECTIVETo observe the effects of mesenteric lymph drainage on erythrocyte rheology and blood viscosity in hemorrhagic shock rats.
METHODSWistar rats were randomly divided into sham-shock group, shock group (establishing hemorrhagic shock model), drainage group (establishing hemorrhagic shock model plus drainaging shock mesenteric lymph from hypotension 1 h). At 3 h of hypotension or corresponding time, blood samples were harvested from the abdominal aorta for determining the erythrocytic parameters, erythrocyte electrophoresis, erythrocyte sedimentation rate (ESR) and blood viscosity, and the erythrocytes aggregation index and erythrocyte deformability index were calculated.
RESULTSCompared with the sham-shock group, the red cell contents, hematocrit (HCT), hemoglobin (Hb), mean corpuscular hemoglobin concentration (MCHC), erythrocyte electrophoretic rate and mobility, erythrocyte deformability index, whole blood viscosity, whole blood relative or reduced viscosity at low and high shear rates in shock group were observably lower, and mean corpuscular volume, electrophoretic time of erythrocyte, ESR, K value of equation and K value of emendation, erythrocytes aggregation index, plasma viscosity in shock group were increased markedly; the MCHC, erythrocyte electrophoretic rate and mobility, whole blood viscosity, whole blood relative viscosity at low and high shear rates in drainage group were reduced, and the red blood cell volume distribution width -SD (RDW-SD) was increased remarkably. At the same time, in drainage group, the HCT, RDW-SD, erythrocyte deformability index, whole blood viscosity and relative viscosity at low and high shear rates were higher, the ESR, K value of equation and K value of emendation, erythrocytes aggregation index, plasma viscosity were lower than that of shock group.
CONCLUSIONThe results indicate that the mesenteric lymph drainage could improve the erythrocyte rheological behavior, as a result, improve the hemorrheological properties in hemorrhagic shock rats.

7.Experiment studies on viscoelastic properties of erythrocyte membrane in patients with pulmonale during acute exacerbation.

The membrane viscoelasticity of erythrocyte taken from both normal subjects and patients with cor pulmonale during acute exacerbation was investigated using a micropipette aspiration technique. Experimental results were analysed with vogit viscoelaticity model based on pioneering theory of Chein et al. The results showed that the erythrocyte membrane elastic moduli ((6.970 +/- 1.050) x 10(-3) dyn/cm) and viscous coefficients ((0.936 +/- 0.242) x 10(-4) dyn x s/cm) of the cor pulmonale patients was significantly higher than those of the normal subjects ((5.203 +/- 1.051) X 10(-3) dyn/cm, (0.620 +/- 0.053) x 10(-4) dyn x s/cm). The membrane elastic moduli, viscous coefficients, rigidity of erythrocyte, and viscosity were all increased. It may be the important subcellular mechanism to cause the decrease of erythrocyte deformability and hyperviscosity of blood in these patients.

8.On the biophysics characteristics of reticulocytes.

This paper reports an in vivo study on the biophysics characteristics of reticulocytes. Anemia was induced by injection of phenylhydrazine in rabbits. The measurements, including electrophoresis rate, hematolytic rate, fluorescent polarization and the changing anisotropic value, were performed in vivo for 72 hours in the process of reticulocytes growing into erythrocytes. It was shown that there were obvious changes in the biophysics characteristics of reticulocytes in this course. Therefore, the findings are of significance to basic, theoretical and clinical studies.

9.A Study on Erythrocyte Deformability in Essential Hypertension.

BACKGROUND: To pass through the microcirculation, erythrocytes need to undergo considerable deformation. We performed an experiment to investigate the erythrocyte deformability in healthy adults and in patients with uncontrolled hypertension and controlled hypertension. METHODS: 5ml of whole blood is anticoagulated with 2mg ethylenediaminotetra-acetic acid(EDTA). 1ml graduated plastic syringe, without its plunger, is filled with anticoagulated whole blood to the 1ml mark, And the syringe is put into the assembled membrane holder. Under standard condition, whole blood is passed through a 5micro polycarbonate membrane filter under negative pressure of 20cm H2O. The studies are performed at roon temperature and within two hours of venipuncture. RESULTS: Erythrocyte deformability was observed in 91 normal healthy adults and 25 patients with uncontrolled hypertension and 18 patients with controlled hypertension. Following results were obtained. 1) In male control age group I, II, III, the erythrocyte deformability indices were 0.776+/-0.125, 0.792+/-0.189, and 0.714+/-0.172 respectively and in female the indices were 0.880+/-0.138, 0.865+/-0.182 and 0.796+/-0.170 accordingly. 2) THe RBC deformability indices in female control troup were significantly higher than the male group in statistics(p<0.05). 3) No significant differences were observed among each age group in control. 4) In patients with uncontrolled hypertension, each age group I, II and III, the erythrocyte deformability indices were 0.785+/-0.050, 0.775+/-0.122, and 0.692+/-0.133 respectively and in control group, the indices were 0.8365+/-0.140, 0.8252+/-0.186 and 0.7534+/-0.173 accordingly. And in patients with controlled hypertension, the indices were 0.820+/-0.00, 0.788+/-0.135 and 0.716+/-0.141 accordingly. 5) The erythrocyte deformability inddex in patients with uncontrolled hypertension were significantly lower than the control group(p<0.05), and no significant differences were observed among each age group between male and female in patients with uncontrolled hypertension. 6) There was no significant statistical difference of deformability indices between uncontrolled group and controlled group(p>0.05). CONCLUSION: This study reveals that the erythrocyte deformability in patients with essential hypertension is significantly lower than the healthy adults.

10.Dynamic changes of ultrastructure of erythrocytes in prolonged preservation at 4 degrees C.

To study the dynamic changes of ultrastructure of erythrocytes in prolonged preservation of blood with preservative fluid containing superoxide dismutase (SOD), the whole blood samples were preserved at 4 degrees C in SOD-containing solution, the morphologic changes of erythrocyte were dynamically ob served by transmission microscopy after preservation for 42, 75 and 85 days, an d the blood samples preserved in GMA solution served as control. Three variance was applied to analyze the data with SAS software. The results showed that the metamorphotic rates of erythrocyte preserved in SOD-containing solution for 42, 75 and 85 days were lower than those of erythrocytes preserved in GMA solution. Most of metamorphotic rates of erythrocyte preserved in SOD-containing solution for 42, 75 and 85 days were correspond to those of erythrocytes preserved in GMA solution for 42 days, or even lower. It is concluded that SOD-containing preservative fluid might help to maintain the normal morphology of erythrocytes in prolonged preservation at 4 degrees C.
