2.Study on characteristic of human rotavirus strains G9P8 on cell culture

Background: In recent years, the rotavirus diarrhea increased significantly in Vietnam, an estimated 50%-70% of children\u2019s hospitalization for diarrhea in 2005. The virus subtype causing disease mainly in Vietnam today was not only G1, G2, G4, like other countries in the world but also the rare strain was G9. Objectives: to isolate G9 rotavirus strains on MA104 cell culture; to selecting G9 rotavirus which developing well on MA104 cells. Subjectives and Method: an experimental research in the laboratory. 20 stool samples derived from 20 children with acute diarrhea caused by rotavirus G9P8 (using RT-PCR method). The samples were processed according to standards of the Centers for disease control and prevention (CDC), Atlanta, USA. Results: eight out of 20 human rotavirus strains G9P8 were positive on Ma104 cell suspension after 3 consecutive cultures (OD indexes of these sample were over 0.100,sample No 2 had the highest OD (1.347)). Sample No 2 was chosen for the first time cloning (25 clones, 17/25 with OD>0.100). And clone No 16 was selected for the second purifying (25 clones, 24/25 with OD>0.100). Ten out of 25 clones in the second time were adapted on monolayer Ma104 cell culture and only clone No 14 and No 18 with highest OD (2.648 and 2.644, respectively) will be used for next studies. Conclusions: cloning method was a basic method for adapting rotavirus clone on cell cultured. Rate of G9P8 rotavirus strain which isolated on Ma104 cells\ufffd?suspension was relatively high. Among the second time clones, 2/10 samples adapted on Ma104 cells with high OD.

3.Adaptation of human rotavirus strain VNHR203-027 on cell culture

Using human rotavirus strain VNHR203-027 to inoculate and passage 11 consecutive times on Africa green monkey kidney (Vero) cells with inoculum medium at different trypsine concentrations, their results are VNHR203-027 have been adapting and growing on Vero cells with trypsin concentration of 30g/ml. Using human rotavirus strain VNHR203-037 to multiply on 4 lots of primary monkey kidney cells and 6 lots of Vero cells gave very high titre on both kinds of cell, the average titre on primary monkey kidney cells was 8,47 Iog10 FFU/ml and of Vero cells was 8.65 Iog10 FFU/ml. No significant difference in potency is found among Vero cells and primary monkey kidney cells (T=10-2 < t=1.86 with =0.05).

6.Relationships among Workplace Bullying, Organizational Culture and Nursing Performance in Nurses

BACKGROUND: This study was aimed to examine relationships among workplace bullying, organization culture and nursing performance in clinical nurses.METHODS: A cross-sectional questionnaire survey was conducted from October 16 to 26, 2018 with 168 nurses participating in RN-BSN and graduate programs in a university. Data was analyzed with SPSS 22.0 by t - test, ANOVA, Pearson's correlation coefficient and hierarchical multiple regression.RESULTS: Of nurses, 23.2% were found as victims of workplace bullying. Higher scores for workplace bullying positively correlated with rank-oriented culture (r=.19, p=.015), negatively correlated with relational-oriented culture (r=−.43, p<.001) and innovation-oriented culture (r=−.35, p<.001). After adjusting for other variables, better nursing performance was associated with lower scores for workplace bullying (β=−.18, p=.026) and innovation-oriented culture (β=.24, p=.023). However, there was no moderating effect of organizational culture in the relationship between workplace bullying and nursing performance.CONCLUSION: Efforts to creating innovative organizational culture and reducing workplace bullying can contribute to improving nursing performance.

INTRODUCTION Early onset sepsis is difficult to diagnose due to nonspecific symptoms and a lack of
reliable tests. It can progress quickly, and lead to neurodevelopmental consequences or be fatal if not
treated. However, approximately 10-fold more newborns are treated with antibiotics empirically and often
unnecessarily. This study aimed to compare the management recommendations of the Neonatal Early
Onset Sepsis Calculator with those of the Centers for Disease Control/American Academy of Pediatrics
guidelines.
METHODS Neonatal Early Onset Sepsis Calculator was applied to the data set to examine how an alternative
model would perform compared to current guidelines published by the CDC and compared to current
practice within the institution. Chi square and kappa value agreement was used to determine the difference
between treatment recommendations of NEOS calculator and AAP guideline.
RESULTS Of the 330 patients who received therapy, only 14.2% were recommended empiric antibiotics by
the EOS calculator, compared to the 39% recommended by the CDC guidelines (p < 0.001, ? = 0.372).
Eleven patients were identified to have culture-positive sepsis.
CONCLUSION The number of infants suspected with EOS and subsequently require antibiotic use at birth
may be dramatically reduced with the use of the neonatal EOS calculator.

8.The change of IL-2, IL-10 in cell culture supernatants of nasopharyngeal carcinoma (NPC) patients

In order to investigate the changes of some cytokines in NPC patients, we have made an measure of IL-2 and IL-10 in cell culture supernatants on 22 NPC patients, at II, III, IVa, IVb stage, positive anapathological test, compare with 12 healthy age-matched controls. The obtained results showed that: IL-2 production was significantly depressed in cell culture supernatants of NPC patients compare with control group. The production of IL-10 was increased in cell culture supernatants of NPC patients at late stages. It suggested that, IL-10 may be an prognostic factor in severe NPC patients and may prove valuable in selecting with NPC who are candidates for aggressive therapy

9.In vitro embryo culture of Epimedium wushanense.

OBJECTIVETo study the in vitro embryo culture of Epimedium wushanense and provide scientific basis for large scale production of tissue culture.
METHODCullus and buds were induced from embryo of E. wushanense on a MS medium supplemented with different 2,4-D,6-BA, NAA, IBA.
RESULTThe optimal compositions of medium that induced callus and buds from embryo were the MS medium supplemented with 2,4-D 2 mg x L(-1), IBA 2 mg x L(-1) and NAA 0.5 mg x L(-1) and the MS medium supplemented with IBA 2 mg x L(-1) and 6-BA 0.5 mg x L(-1), respectively. The optimum medium for callus differentiation was MS + 6-BA 1 mg x L(-1) + NAA 0.5 mg x L(-1) + IBA 1 mg x L(-1), and MS +6-BA 1.0 mg x L(-1) + NAA 0.5 mg x L(-1) for shoots proliferation.
CONCLUSIONUsing embryo as explants, the method of induction and culture of E. wushanense was established by the callus and buds, and the embryo of E. wushanense can be quickly propagated.

10.Optimization of noni callus induction and establishment of callus suspension system.

The aim of the study was to obtain the secondary metabolites in the stem segment of noni and to establish genetic transformation system. The stem segments (no axillary buds) of noni were used as explants to induce the callus, and then to establish the cell suspension system. The factors affecting callus induction and cell suspension were studied. The results showed that the optimal culture medium for induction was MS with 1.0 mg/L 6-Benzylaminopurine (6-BA) and 0.1 mg/L 2,4-dichlorophenoxyacetic acid (2,4-D), and the optimum culture medium for suspension was MS with 1.0 mg/L 6-BA and 0.1 mg/L 2,4-D, 3% sucrose and the pH of 5.85, with the initial inoculation amount of 37.5 g/L, and the speed of 110 r/min and 25±2 °C applying darkness culture. The suspension cells grew well and showed the maximum growth rate. The growth curve of the suspension cells from the stem segment of noni was in "S-typed" trend, and it should be transformed to the fresh medium between 12 and 20 d. During the culture, the pH of the culture medium decreased and then slowly increased, and the optimum pH for the suspension cells culture of callus from noni's stem segments was 4.5-5.0. In this study, the stable cell suspension system of the stem segment of noni was successfully established.
