2.The Effect of Histone Deacetylases-1 siRNA on the Growth and Apoptosis of HeLa Cells

Objective To investigate the effect of histone deacetylases-1(HDAC-1)siRNA on the HDAC-1 protein expression,cell growth and apoptosis in HeLa cells.Methods The HDAC-1 protein was knocked down by HDAC-1 siRNA.HDAC-1 protein was detected by Western blot.The cell growth inhibition was assesses by MTT and clone forming assay.Apoptosis was measured by flow cytometry.Results Forty-eight h after transfection of HDAC-1 siRNA,HDAC-1 protein expression in HeLa cells was down-regulated obviously.The down-regulation of HDAC-1 significantly reduced the colony formation rate,inhibited cell proliferation and induced apoptosis in HeLa cells.Conclusion HDAC-1 siRNA may play an anti-proliferation role in HeLa cells by inducing apoptosis.
3.The clinical features and treatment of autoimmune hemolytic anemia in patients after orthotopic liver transplantation

Objective To analyze the clinical feature,pathogenesis and treatment of autoimmune hemolytic anemia (AIHA) in patients after orthotopic liver transplantation.Method A retrospective analysis was conducted on 7 patients who developed AHA after orthotopic liver transplantation.The clinical feature,pathogenesis and treatment of AIHA were reviewed.Result The incidence of AIHA was 0.98% (7/713).AIHA occurred (7.9-± 4.0) (3-14) days after transplantation.All patients were characterized by progressive anemia and jaundice.Laboratory examination showed lower hemoglobin and platelet count,higher lactate dehydrogenase,higher indirect bilirubin and higher reticuloeyte count.The causes of AIHA included ABO-incompatible liver transplantation,infection and repeated transfusion.Conclusion AIHA is a rare and serious complication after liver transplantation.The most cause is ABO-incompatible liver transplantation.The patients can be successfully cured if the diagnosis is clarified and the treatment is definitive.

5.Breeding of Arachidonic Acid Producting Strain with Mortierella Sabellina by Ultraviolet Mutation

In order to gain the strain which has high growth vigor and whose biomass concentration satisfy further fermentation to produce arachidonic acid, we took the yield of microbes olein and arachidonic acid as evaluative index, adopted twice ultraviolet mutation, determined the time of ultraviolet irradiation by single factor expriment, and then the content of the arachidonic acid was measured by GC. The results of this experiment showed that the power of viltalight lamp was 20 W, exposure distance was 30 cm and exposure time was 80 s, lethality of spores of Mortierella isabellina was 76.4%. After twice ultraviolet mutation and repeatedly screen, a mutation of Mortierella isabellina Z80s2-109 whose arachidonic acid concentration in biomass were 3.77 times of the control strains was obtained and genetic stability.
6.Acute exacerbation in idiopathic pulmonary fibrosis(IPF):a case report and review

Objective To improve the diagnosis and treatment of acute exacerbation in idiopathic pulmonary fibrosis(IPF)Methods A case with clinical proven acute exacerbation in IPF and treated with corticosteroid was described and the foreign literature was reviewed.Results Most patients with idiopathic pulmonary fibrosis(IPF)show slowly progressive deterioration.However,accelerated deterioration also occurs without any definite factors on infection,heart failure,et al in a few of patients with IPF who have previously shown slowly progressive deterioration,and for part of patients corticosteroid treatment is effective.Conclusion Some acute exacerbation in idiopatic pulmonary fibrosis can be idiopathic.
7.Determination of Angelica Sinensis Polysaccharide ( ASP1 ) in Rats by HPLC-FD with Pre-column Derivat-ization

Objective:To establish an HPLC-FD method for determining the content of Angelica sinensis polysaccharide (ASP1) to lay foundation for its pharmacokinetic study in rats. Methods: Purified ASP1 was labeled with FITC by the method of Belder and Granath to obtain ASP1-FITC. The tissue samples were treated with 30% trichloroacetic acid and 11% NaOH before injection. The samples were determined by HPLC-FD. A PL aquagel-OH MIXED column was used,and the mobile phase was phosphate buffer( dis-solve NaH2PO32. 34g , Na2HPO34. 33 g and NaCl 11. 70 g into 1000 ml water) with pH of 7. 0. The flow rate was 0. 5 ml·min-1. The excitation and emission wavelengths was set at 495 nm and 520 nm, respectively. Results:The linear calibration curve was within the concentration range of 0. 25-40. 00 μg·ml-1(r=0. 9996)with the lower limit of quantification of 0. 20μg·ml-1 in tissue sam-ples. The extraction recovery of ASP1 was determined at low, medium and high concentration with the recovery of 91. 98%-114. 20%. The intra and inter-day RSDs were lower than 8. 31% and 2. 94%, respectively. Conclusion:The method to determine the content of ASP1 in rats by HPLC-FD with pre-column derivatization has been esablished. It is simple, accurate and reliable, which can be suc-cessfully applied in the study of pharmacokinetics and tissue distribution of ASP1 in rats.
8.Influence of intra-abdominal hypertension after orthotopic liver transplantation on the recipients' respiratory function and management

Objective To observe and analyze the reasons of intra-abdominal hypertension,and to discuss the influence of intra-abdominal hypertension (IAH) on recipients' respiratory function and its changes after disposition.Method During January to June in 2012,50 patients who had performed orthotopic liver transplantation were involved in this study.Intra-abdominal pressure was measured by AbViser Intra-Abdominal Pressure Monitoring Kit.According to the value of intra-abdominal pressure,the patients were divided into two groups (IAH group and control group).The reasons of IAH were observed and analyzed.Respiratory rate,oxygenation index,and the arterial partial pressure of carbon dioxide between IAH and control groups were compared.IAH was dealt with restricting oral input,inserting nasogastric tube,administering enema,inserting ilues tube or placing percutaneous catheter drainage according to reasons.Intra-abdominal pressure,respiratory rate,oxygenation index,and arterial partial pressure of carbon dioxide were also observed after disposition.Result The incidence of IAH was 44% (22/50).The respiratory rate of patients in IAH group was quicker than that in control group.The oxygenation index in IAH group was lower than that in control group.The arterial partial pressure of carbon dioxide in IAH grade I group was not higher than in control group.However,the arterial partial pressure of carbon dioxide in IAH grade Ⅱ and Ⅲ groups was higher than in control group.After disposition,the intra-abdominal pressure,respiratory rate and arterial partial pressure of carbon dioxide in IAH groups were decreased from 16.4 ± 3.1 mmHg,33.0 ± 5.2 L/min and 47.6 ± 9.4 mmHg to 10.5 ± 1.8 mrnHg,19.7 ± 4.1 L/min and 39.9 ± 2.9 mmHg respectively,while the oxygenation index was increased from 202.7 ± 72.9 mmHg to 350.5 ± 98.5 mmHg.Conclusion The incidence of IAH after orthotopic liver transplantation was high.The influence of IAH on patients' respiratory function was serious.After timely and proper disposition,the respiratory function of patients with IAH could be improved.
10.Effects of ischemic time factors on reduction of myocardial ischemia-reperfusion injury by sulfentanil postconditioning in rats

Objective To evaluate the effects of ischemic time factors on reduction of myocardial ischemia-reperfusion injury by sulfentanil postconditioning in rats .Methods Healthy adult male Sprague-Dawley rats ,weighing 230-250 g ,were heparinized and anesthetized with intraperitoneal 5% chloral hydrate 8 ml/kg .The hearts were excised and perfused in a Langendorff apparatus with K-H solution .After 30 min of stabilization ,48 isolated rat hearts were randomly assigned into 6 groups (n=8 each) using a random number table .In I/R1-3 groups ,the hearts were subjected to 30 ,45 ,and 60 min of myocardial ischemia ,respectively ,followed by 90 min of reperfusion . In SP1-3 groups , the hearts were subjected to 30 , 45 , and 60 min of myocardial ischemia , respectively ,followed by 90 min of reperfusion ,and sulfentanil 100 nmol/L was added to K-H solution at 15 min of reperfusion for postconditioning . Left ventricular end-diastolic pressure (LVEDP ) , left ventricular developed pressure (LVDP) ,+dp/dtmax ,-dp/dtmax ,HR and coronary flow (CF) were measured at the end of equilibration and 15 ,30 ,60 and 90 min of reperfusion .Creatine kinase (CK) and lactate dehydrogenase (LDH) activities in coronary effluent were measured at 5 min of reperfusion .Myocardial infarct size was determined in 5 hearts chosen randomly at 90 min of reperfusion .Results LVEDP was significantly higher ,and LVDP , ± dp/dtmax and CF were lower at each time point of reperfusion than at the end of equilibration in I/R1-3 groups ( P<0.05) .Compared with I/R1 and I/R2 groups ,LVEDP was significantly increased ,and LVDP and ± dp/dtmax were decreased at 30 ,60 and 90 min of reperfusion ,HR and CF were decreased ,and CK and LDH activities in coronary effluent and infarct size were increased at 15 and 30 min of reperfusion in group I/R3 ,and LVEDP was significantly decreased ,LVDP , ± dp/dtmax and CF were increased ,CK and LDH activities in coronary effluent and infarct size were decreased at 30 , 60 and 90 min of reperfusion in groups SP1 and SP2 ( P< 0.05 ) . Compared with group I/R3 , LVDP was significantly decreased at 30 ,60 and 90 min of reperfusion ( P<0.05) ,and no significant change was found in CK and LDH activities in coronary effluent and infarct size in group SP3 ( P>0.05 ) .Conclusion Sulfentanil postconditioning can attenuate myocardial ischemia-reperfusion injury when the rats are subjected to 30 or 45 min of ischemia ,however ,it provides no myocardial protection when the rats are subjected to 60 min of ischemia .
