- Abstract: OBJECTIVE: To determine the pectin content in Panax ginseng. METHODS: Soluble sugar was removed firstly, then pectinase was added for enzyniolysis, and 3, 5-dinitrosalicylic acid colorimetry was employed for determination of pectin. RESULTS: The optimal conditions of determination were as follows: 500 mg of Panax ginseng powder was extracted with 80% alcohol in water bath at 80°C, then filtrated, and the residue was enzymolyzed with pectinase of 1200 u at 50°C for 6 h in 50 mL of 0.1 mol · L-1 citric acid-sodium citrate buffer (pH 3.5). The galacturonic acid from degraded pectin in Panax ginseng was determined with 3, 5-dinitrosalieyfic acid colorimetry. CONCLUSION: This method has been proved to be convenient and accurate, which can be applied to the determination of pectin content in Panax ginseng.
