Effect of trichostatin A and sodium butyrate on variation of Thl/Th2 cell balance after differentiation of peripheral blood CD4+T cells

Abstract:
Objeciive To investigate the effects of teichostaein A(TSA) and sodium butyrate(NaB), histone deacetylase inhibitors, on the balance of Th1/Th2 after differentiation of peripheral blood CD4+T cells. Methods Peripheral CD4+T cells were grown in vitro and were dirided into TSA group, NaB group, and CON group The concentrations of IL-4 and IFN-γ (Th1/Th2 factors) in the supernatantsWere examined by ELISA in each group 48 h later; the expresdcms of IL-4 and IFN-γ were armlyzed by qRT-PCR; and the Tllymphocytes in peripheral blood (PBL) were measured by flow cytometry Resutts ELISAshowed that the supernatant IFN-γ and IL-4 levels n the TSA and NaB groups were eignieicanfly higher than those in the CON group 48 h after treatmerut with TSA(0. 33 jumol/L) and NaB (L 4 mmol/L); real-iime quantitative PCR also showed that the ejpresricms of IFN-γ and IL-4 mRNA n TSA and NaB groups were significantly higher than those in the CON group (3. 12: 1, 2. 3: 1; P<0. 05). However, flow cytometry showed that the raios of IFN-γ+ cells/IL-4+ cells were significantly higher n TSA and NaB groups than that n the CON group((1. 93: 1,1 55: 1;P<0. 05). Conclution Treatment with TSA and NaB, berth histone deacetylase inhibitors,can induce a shfft of Th1/ Th2 cell balance toward Th1 cells in peripheral CD4+T cells.
