Effect of recombinant adenovirus livin siRNA on proliferation and apoptosis of U251 cells

Abstract:
Objective To investigate the silencing effect of siRNA targeting livin gene on the proliferation and apoptosis of U251 cells. Methods Adenoviral Vector of siRNA targeting livin was constructed and transfected into U251 cells. The gene silencing efficiency was monitored by RT-PCR and Western blotting analylis; the proliferation and apoptosis of U251 cells were detected by MTT assays and flow cytometry, respeclwey Mean while, the therapeijlic effect of the recombrnarrt adenoVirus was also observed en tumor-beaeing mice. Resutts The constructed recombinant adenoVirus effeclwely lnhlbited the expreseion of livn at both mRNA and proton leVlls; it also lnhlbited the proeferalion and promoted the apoptds of U251 cells. In addilton, the recombinant adenoVirus effetively inhibited the humor growth in mice and improved the mean survival period of tumor-beaeing mice. Conclusion Sileneing livin gene expression with recombination adenovirus carrying siRNA targeting liven can inhibit the proliferation and promote the apoptosis of U251 cells, which casts new lights on the treatment of glioma.
