Abstract: Fatigue is one of the most common symptoms of primary biliary cirrhosis (PBC) and greatly affects patients′ quality of life. The pathogenesis of fatigue in PBC remains unknown and there are no effective therapeutic methods. This article reviews the latest advances in the pathogenesis and treatment of fatigue in PBC. Studies have shown that the causes of fatigue in PBC include various central and peripheral factors, such as the changes in the brain, sleep disorders, depression, autonomic nervous dysfunction, and abnormal muscle metabolism. Modafinil does not have a definite therapeutic effect in fatigue in PBC. The causes and treatment of fatigue in PBC still need in-depth systematic studies.
