Therapeutic effect of clopidogrel combined atorvastatin on aged UAP patients and its regulation on mTOR protein

Abstract:
Objective:To explore therapeutic effect of clopidogrel combined atorvastatin on aged patients with unsta-ble angina pectoris(UAP)and its regulation on mammalian target of rapamycin(mTOR).Methods:A total of 120 aged UAP patients treated in our hospital from Jan 2013 to Jan 2016 were selected.The patients were randomly and equally divided into clopidogrel group,atorvastatin group and combined treatment group(received clopidogrel com-bined atorvastatin based on routine treatment),and all groups were continuously treated for eight weeks.Duration and onset frequency of angina pectoris,serum expressions of PI3K,Akt,mTOR,connexin(Cx)40 and 43,endo-thelin and its receptor,leptin and its receptor were measured and compared among three groups.Results:Compared with before treatment,there were significant reductions in duration and onset frequency of angina pectoris,serum expressions of PI3K,Akt,mTOR,endothelin and its receptor,leptin and its receptor except PI3K of clopidogrel group,and significant rise in serum expressions of Cx40 and Cx43 after eight-week treatment in all groups after eight-week treatment,P< 0.05 or < 0.01. Compared with clopidogrel group and atorvastatin group after eight-week treatment,there were significant reductions in duration[(5.23 ± 1.02)min,(5.36 ± 0.89)min vs.(3.12 ± 0.24)min]and onset frequency[(3.45 ± 0.34)times/d,(3.64 ± 0.28)times/d vs.(2.64 ± 0.37)times/d]of angi-na pectoris,serum expressions of PI3K[(0.49 ± 0.08),(0.46 ± 0.19)vs.(0.35 ± 0.04)],Akt[(0.40 ± 0.07), (0.41 ± 0.11)vs.(0.32 ± 0.02)],mTOR[(0.31 ± 0.06),(0.30 ± 0.05)vs.(0.26 ± 0.06)],endothelin[(0.51 ± 0.09),(0.50 ± 0.11)vs.(0.45 ± 0.08)]and its receptor[(0.30 ± 0.07),(0.31 ± 0.05)vs.(0.26 ± 0.06)],leptin [(0.34 ± 0.05),(0.33 ± 0.06)vs.(0.27 ± 0.05)]and its receptor[(0.32 ± 0.06),(0.34 ± 0.05)vs.(0.27 ± 0.07)],and significant rise in serum expressions of Cx40[(0.29 ± 0.07),(0.30 ± 0.05)vs.(0.38 ± 0.07)]and Cx43[(0.32 ± 0.06),(0.31 ± 0.04)vs.(0.37 ± 0.08)]in combined treatment group,P<0.05 or <0.01. Conclu-sion:Clopidogrel combined atorvastatin possesses significant therapeutic effect on aged patients with unstable angina pectoris,and it may be related to regulating expression of PI 3K/Akt/mTOR pathway proteins.
