Abstract:
Objective To investigate the diagnostic value of plasma exosomal long non-coding RNA (lncRNA) H19 for breast cancer.Methods The levels of lncRNA H19 were detected by quantitative reverse transcription PCR (qRT-PCR) in plasma exosome and whole plasma from 50 patients with breast cancer,25 patients with benign breast diseases and 25 healthy controls.The correlations be tween plasma exosomal H19 levels and clinical pathological characteristics of breast cancer were analyzed.The receiver operating characteristic (ROC) curve was used to evaluate the diagnostic value of plasma exosomal lncRNA H19 in breast cancer.Results The expression of exosomal lncRNA H19 in plasma of patients with breast cancer was significantly higher than those in whole plasma from the patients with both breast cancer and benign breast diseases and healthy controls (all P < 0.01).The expression level of exosomal lncRNA H19 in breast cancer patients correlated with the levels of estrogen receptor (ER),progesterone receptor (PR),c-erbB-2,ki67 and P120,and lymph node metastasis (all P < 0.05).The area under the ROC curve (AUCROC) was 0.779,and the sensitivity and specificity were 70%.The diagnostic capability of exosomal lncRNA H19 was significantly higher than that of whole plasma.Conclusion Plasma exosomal lncRNA H19 may serve as a potential biomarker for diagnosis of breast cancer.
