The analysis of glucocorticoid receptor of rat liver with HPLC

Abstract:
Objective:To isolate and purify glucocorticoid receptor(GR) of rat liver with HPHIC and verify the existence of the low affinity GR as the further evidence for theoretical basis of large dose glucocorticoid treatment.Methods:The supernatant of the rat liver was performed with Pseudoscatchard assay and HPHIC analysis after ultracentrifuge.SDS PAGE and Western blot were used to identify GR.Results:At the standard atmosphere,when the temperature was at 0 4℃,only one peak appeared at 9 11 min.When the temperature was at 25℃ and the concentration of 3H Dex exceeded 50 nmol/L,two peaks appeared at 3 7 min and 9 11 min.The protein was verified with SDS PAGE and Western blot, its molecular weight was 60 000,which could be bound with monoclonal antibody of GR.Conclusion:Two peaks of GR appears with HPHIC analysis, suggesting they may be high and low affinity receptor.Further researches are necessary to ascertain it.
