Migration of neural stem cells on the fibronectin-coated matrix induced by transforming growth factor beta 1

Abstract:
BACKGROUND:How to migrate transplanted or autologous neural stem cells(NSCs) into the exact focus region in a high survival rate to repair or replace injured tissues is a key problem using NSCs in treatment of damage to the nervous system.OBJECTIVE:To investigate the effect of transforming growth factor-beta 1(TGF 0 1) on the directional migration of NSCs on growth matrix with fibronectin.DESIGN,TIME AND SETTING:The randomized control animal experiment was performed at the Institute of Neurology of Jiamusi University from March to November 2007.MATERIALS:A total of 72 Wistar rats of the same gene background were randomly assigned into 3 groups,a model control group,a simple NSC transplantation group,a TGF ?1 and fibronectin coated NSC transplantation group(combined transplantation group),24 in each group,Six rats in each time point(1,3,7 and 14 days after transplantation).METHODS:Rat models of focal cerebral ischemia were established by Zea Longa suture method in each group.5 u L of Brdu-labeled NSC suspension was injected into left lateral cerebral ventricle of rat models in the simple NSC transplantation group.2 ? L of 100 ?g/L TGF was infused into the sensor motor cortex of left side,and 5 ? L Brdu-labeled fibronectin coated NSC suspension into the left lateral cerebral ventricle in the combined transplantation group.Brain tissues were serially sliced into sections at days 1,3,7 and 14 after transplantation in each group.MAIN OUTCOME MEASURES:Migration of Brdu-labeled NSCs in the brain of host.RESULTS:After in vitro cultured NSCs transplanted into rat models of cerebral ischemia,cells were living in the the subventricular zone at day 1 in each group.The cell number gradually increased at day 3.A large number of Brdu-labeled cells migrated from the lateral ventricle at day 7.Brdu-labeled cells in the subventricular zone became more and reached a peak.The number of positive cells was more in the combined transplantation group than in the model control group and the simple NSC transplantation group,with a significant trend of migration.Number of NSCs increased in the cortex at day 3 after transplantation,and reached a peak at day 7.Number of NSCs was more at each time point compared with the model control group,showing a significant difference.NSCs could differentiate into neurons in the cortex.CONCLUSION:TGF-?1 can accelerate the oriented migration of NSCs on the fibronectin-coated matrix.TGF-?1 and fibronectin in inducing NSC transplantation have synergistic effects.
