Effect of DAPK carboxylic terminal peptide on TNF-? inhibition of human embryonic lung fibroblast proliferation

Abstract:
Objective To investigate the effect of death associated protein kinase(DAPK) carboxylic terminal peptide(DCTP) on inhibition of TNF-? to human embryonic lung fibroblast proliferation.Methods Eukaryotic Expression vector pcDNA3.1(+)-DCTP was constructed and transferred into human embryonic lung fibroblast in vitro.The expression of DCTP was detected by RT-PCR and Western blotting.The proliferation of human embryonic lung fibroblast was examined with MTT assay.Results Eukaryotic Expression vector pcDNA3.1(+)-DCTP expressed DCTP in human embryonic lung fibroblast.pcDNA3.1(+)-DCTP did not affect the proliferation of human embryonic lung fibroblast.However,DCTP inhibited the toxicity of TNF-?.Conclusion The toxicity of TNF-? is blocked by DCTP.It offers theoretical and technical guidance to the clinical study of DCTP for protecting normal cells from damage by TNF-? therapy.
