Abstract:
Objective To investigate the effects of endothelin-1(ET-1) on the expressions of matrix metalloproteinase-3(MMP-3) and tissue inhibitor of metalloproteinase-1(TIMP-1) in human mesangial cells(MCs).Methods Cultured human MCs were exposed to ET-1,the expressions of MMP-3 and TIMP-1 proteins were measured by ELISA.Results The expressions of MMP-3 and TIMP-1 in MCs were enhanced by ET-1,but the ratio of MMP-3/TIMP-1 decreased in a dose-dependent and time-dependent manner.Conclusion ET-1 may decreased the ratio of MMP-3/TIMP-1 in MCs,which then contribute to the inhibition of ECM degradation in glomerulus.
