Abstract:
The method of preparing the high purity factorⅧ complex was introduced.Itwas prepared from the fresh frozen plasma as raw material by thawing to formcryoprecipitate,adsorbing with Al(OH)_3 gel,and then precipitating with glycineand NaCl.The further purified product was yielded by collecting the componentrich in FⅧ coagulant activity(FⅧ:C)and von Willebrand factor(vWF)afterit was subject to Sephacryl S-1000 colomn chromatography.Thereafter,its totalcontent of protein was 0.188mg/ml,FⅧ:C specific activity-15.43IU/ml,FⅧ-related antiegen(FⅧ R:Ag)specific activity-9.85 U/ml,vWF's Ristocetin co-factor(FⅧ R:RC of)specifical activity-24.2 U/ml.And the determination of itby the rocket electrophoresis didn't show any containing of fibrinogen,fibronec-tin,IgG and other foreign proteins.
